La primera cadena de ADNc fue sintetizada mediante el Sistema de Sintesis RiboClone[R] ADNc (Promega 2005) con la pareja de cebadores oligo(dT)16 (5'-[AACATATGCGGCCGCATTATGGCCTTT.sub.16]-3' y 5'-GATCTTCCACGCGTCGAC[(T).sub.16] 3') y a partir de las condiciones de amplificacion recomendadas en el prospecto del juego de reactivos Marathon[TM].

Para la construccion del ADNc, 1 [micron]g del ARNm se puso en contacto con 50 ng/ml de cebadores aleatorios, 0,5 mM de dNTP, 0,005 M de DDT y 200 U de la enzima SuperScript III, en un volumen final de 20 [micron]l (Invitrogen).
