- It was a building with two sides, on two streets (the Kalverstraat and Rokin) which lie parallel to each other. This map image from the Zwaailichten site in the external links (which is a very knowledgeable site about disasters in the Netherlands, but sadly not run by a major mass media organization though it should qualify under the WP:ELPEREN criteria as giving additional information) clarifies the situation. There is a more detailed diagram in the Amsterdam Fire Department report on page 50 (source: [1]). BTW you can also see the geographical situation on this Google Maps link: [2], the Rokin Plaza which lies on Rokin no. 14 occupies the space where Hotel Polen used to be. - Regarding your question "Hotel Polen was once known as a fashionable hotel - when? Any more info?" it was LITERALLY mentioned as this by the Los Angeles Times in this link which states "Fire raced through the fashionable Polen Hotel near the royal palace in downtown Amsterdam Monday, leaving at least six people dead and 30 others injured, police said." (sadly most of the article is behind a paywall and I don't have the opportunity to access that article). The LA Times information was outdated though in 1977 it was not so fashionable anymore and may have qualified as "run down" although I couldn't find sources mentioning that so I removed it. Also please note that the various news sources give varying casualty rates but this was because the articles were published soon after the fire (and there was no Twitter back then so updates about casualty numbers took a long time to gel out). - Cuypers designed the adjacent building on the Rokin which became the Hotel Polen together with the previously used building on the Kalverstraat. "There was no sign of fire yet" seem contentious on a second view and I can't find clarifying information so I will remove that.
