The refuge is exactly that, a high conservation value habitat of the Grey-crowned Babbler, Hooded Robin, Speckled warbler, Spectacled hare-wallaby, Inland forest Bat, koalas, Horsfield's Bronze-cuckoo, Black-faced Cuckoo-shrike, Yellow-throated Miner, Tree Martin, and the Rainbow Bee-eater.

As dingoes are known to eat the occasional mala, the Wildlife Commission baited the dogs to give the hare-wallaby a better chance.

Biodiversity Conservation Officer at the Conservation Council of Western Australia (CCWA) Cameron Poustie says Barrow Island's primary significance is its mammals, particularly endemic subspecies such as the Barrow Island boodie, Barrow Island euro, Barrow Island mouse, Barrow Island golden bandicoot, and Barrow Island spectacled hare-wallaby.
