Acid phosphatase activity in the presence of 50 mM tartrate was assayed with naphtol AS-MX phosphate (Sigma) as a substrate and freshly prepared Fast Violet B (Sigma).

Following this step, sections were incubated in the fast red/TR naphtol mixture until the specific regions were stained red, and then the sections were either briefly put into Mayer's hematoxylene in order to visualize the nuclei, or were not subjected to counterstaining.

Transverse sections (8-10[micro]m) were cut with an AO 810 rotatory microtome and placed on slides used for the following histochemical tests: periodic acid-Schiff reagent to detect non soluble polysaccharides, naphtol blue-black for proteins, oil red O for lipids, phloroglucinol-HCl for lignin, vainillin-HCl for hydrosoluble tannins and lugol for starch (Jensen, 1962; Lopez et al., 2005).

For this procedure to take place target molecules are directly conjugated to redox active and electrocatalytic moieties through coordinative bonds with purine bases in the miR molecule.20 A novel electrochemical miR discovery process based on "enzyme amplified biosensing" of miR-21 from cell lysate of total RNA was established by Kilic et al.21 In this method total RNA encom- passing miR was secluded from breast cancer cell lysates and "oxidation signal of the enzymatic reac- tion product, alpha naphtol" was measured.

One unit of activity was defined as 1 pg of naphtol [min.sup.-1] at a CME of 0.02 mL [micro] [g.sup.-1] [cm.sup.-1] (Alvarez-Gonzalez et al., 2006; Rivera-Perez et al., 2010).
