It consists of [[Doctor Octopus]], [[Sandman (Marvel Comics)|Sandman]], [[Shocker (comics)|Shocker]], [[Vulture (comics)|Vulture]], [[Rhino (comics)|Rhino]], and [[Electro (Marvel Comics)|Electro]]. Like in the original version of the team, Doctor Octopus is the leader of the team. The team's members have all previously been members of the Sinister Six in either the comic books or other animated Spider-Man programmes. The villains pooled their resources and organized a well-planned jailbreak orchestrated by Hammerhead on Tombstone's orders, then went after Spider-Man. After realizing that his previous strategy's wouldn't work, Spider-Man beat a hasty retreat. Going to bed that night, he wished that he could wake up tomorrow and see the Six back behind bars. The black suit, like a malignant [[genie]], took over his body as he slept and met Dr. Octopus and his team. Spider-Man—or, specifically, the symbiote acting as Spider-Man—defeated all six handily, using both a few new tricks which were previously impossible in his standard [[blue]]-and-[[red]] costume and the Six's own talents and weaknesses against each other. Peter later awoke, bone-tired, late in the day to find his idle wish had been granted. The Bugle reported the Sinister Six has been captured, and his own byline below photos of the battle stared him in the face. He realized the symbiote suit was "wearing me" and was horrified. Of note is that the team never actually uses the codename "Sinister Six" amongst themselves; it's only used in the Bugle headline reporting their defeat and capture. [[Kraven the Hunter]] busted out Electro from Ravencroft, but Doctor Octopus was unwilling to join them (though this was in the following episode to be Doctor Octopus feigning unwillingness to fool the Ravencroft staff and that like in the comics, he was the Master Planner). Electro and Vulture attacked the skating rink near the town's Christmas tree causing Spider-Man to lure them away. After taking out Vulture and Electro, Sandman and Rhino attacked. Upon driving them to the pier, Spider-Man doused Sandman with a hydrant (with some of it freezing him through the cold night air) and then sent Rhino under the icy water. With Sandman completely frozen, Mysterio and Kraven attack using illusions of Doctor Octopus and Shocker. Upon hitting Kraven's face with a perfume bottle, Spider-Man is attacked by Mysterio's technological Homunculis. Kraven unwittingly gave away Mysterio's robot duplicate which was about to explode. Spider-Man swings away the nearby Santa Claus and elf as Kraven is knocked out by the explosion and webbed up. Upon trapping and making sure it's the real Mysterio, Spider-Man tries to get him to tell him who the Master Planner is. With Mysterio taken into custody, Tinkerer had managed to extract the other members at the last minute. It should also be noted that the latter incarnation has the most members of the original Sinister Six from the comics, with only Rhino replacing Doctor Octopus.

When Spider-Man arrives to rescue Dr. Curt Connors, he is ambushed by Doctor Octopus and his villain allies as Doctor Octopus mutates Dr. Curt Connors into [[Lizard (comics)|Lizard]] with the help of a control collar on his neck to be the sixth member of Doctor Octopus' Sinister Six. The Sinister Six then attack Spider-Man where Doctor Octopus mentions that he has jammed all the signals. Unable to get through to his team, Spider-Man had to use some tactics in an echo tunnel to fool the Sinister Six and send them down different tunnels. When Spider-Man corners Lizard who temporarily changes back, Doctor Octopus reactivates the control collar on Lizard who knocks out Spider-Man as the rest of the Sinister Six arrives. When Doctor Octopus states that they should have Spider-Man unmasked to complete his revenge, Kraven the Hunter suggests that Lizard should do the honors of unmasking Spider-Man. As Lizard prepares to unmask Spider-Man, Doctor Octopus controls Lizard to remove Spider-Man's mask as Kraven the Hunter plans to do the job himself. Lizard attacks Kraven the Hunter as Spider-Man tricks Rhino into flooding Doctor Octopus' lab. Spider-Man swims to the docks as the Sinister Six follows him. Spider-Man rides the helicopter to the [[Statue of Liberty]] as the Sinister Six catches up to Spider-Man. Things weren't looking good for Spider-Man when the Sinister Six make a comeback in their fight against Spider-Man. In the nick of time, Spider-Man's team arrives to help him fight the Sinister Six. When Spider-Man has his team switch opponents, Spider-Man fights Doctor Octopus as Lizard breaks free from the control collar. Spider-Man then removes the control collar from Lizard. While Spider-Man's team watches over the other Sinister Six members until S.H.I.E.L.D., Lizard escapes into the sewers. The rest of the Sinister Six are arrested by S.H.I.E.L.D.
