Hi @GavinLi2016, It depends how the spot colours are encoded. TIFF is very flexible, unfortunately, and there are several conventions for spot colours. If it's CMYKX, where X is the spot colour and it's a band interleaved TIFF, then yes it can. If it's a multipage TIFF with C, M, Y, K and X in separate pages, then also yes.
