ABI1 (ABA Insensitive 1) is an important component of the core regulatory network in early ABA (A... more ABI1 (ABA Insensitive 1) is an important component of the core regulatory network in early ABA (Abscisic acid) signaling. Here, we investigated the functions of an ABI1 ortholog in Brassica oleracea (BolABI1). The expression of BolABI1 was dramatically induced by drought, and constitutive expression of BolABI1 confers ABA insensitivity upon the wild-type. Subcellular localization and phosphatase assays reveal that BolABI1 is predominantly localized in the nucleus and harbors phosphatase activity. Furthermore, BolAB-I1 interacts with a homolog of OST1 (OPEN STOMATA 1) in B. oleracea (BolOST1) and can dephosphorylate ABI5 (ABA Insensitive 5) in vitro. Overall, these results suggest that BolABI1 is a functional PP2C-type protein phosphatase that is involved in the negative modulation of the ABA signaling pathway.

ABSTRACT Studies on the accumulation of oak-derived volatile compounds in wine and on the effects... more ABSTRACT Studies on the accumulation of oak-derived volatile compounds in wine and on the effects of different factors on the final concentration of these compounds are usually conducted with wine samples and with model solutions; the results obtained with model solutions are generally assumed to be comparable to results that would have been obtained with real wine. This present study was designed to test this assumption and to assess whether the studies with model solutions could be useful for explaining the behavior of these volatile compounds during aging. The evolution of the concentration of oak-derived volatile compounds (furfuryl and guaiacyl compounds, cis- and trans-beta-methyl-gamma-octalactone, and vanillin) was studied in wine and model solutions containing oak chips. Given that the physicochemical properties of the wine or the model solutions might also influence extraction, the model solutions were adjusted to three different titratable acidity values and three different alcohol percentages. The extraction rate and final quantities of guaiacyl derivates and lactones in model solutions were very similar to those observed in wines. However, the behavior of other compounds, such as furfuryl and vanillin, depended on the type of matrix in which the oak chips were soaked. The physicochemical properties of the matrix should also be taken into account when studying the extraction of oak-derived volatile compounds, because these properties can affect the accumulation of some compounds, such as lactones.

Carcavelos fortified wine is an appellation of origin and demarcated as D.O.P. (Denominação de Origem Protegida). This study examines the effects of barrels made from botanical species (Quercus pyrenaica, and Quercus robur) and toasting method (medium and high) on a single vintage wine that has been aged for 8 years. Twenty barrels were used, with five replicates for each factor. The barrels were fabricated and toasted using the same cooperage, J.M. Gonçalves in Portugal. Significant differences were seen between the species Q. robur and Q. pyrenaica, with an impact on total phenolic content, including both flavonoids and non-flavonoids. The total phenols of the wine aged in Q. pyrenaica barrels was significantly higher than in the Q. robur barrels, and Q. pyrenaica contained more flavonoids than Q. robur in medium and high toast barrels. Q. pyrenaica showed more non-flavonoid compounds than Q. robur inhigh and medium toasted barrels, but this difference in non-flavonoids was only statistically significant in the high toasted barrels. The degree of toasting had significant effects on the flavonoid content of the wine, as well as the tanning power. Flavonoid content increased for both Q. pyrenaica and Q. robur in the wines that were aged in high tasted barrels compared to those that were medium toasted. The tannin power decreased for both Q. pyrenaica and Q. robur when the toasting increased.
