This study aimed to evaluation the effect of different equilibration time and post thawing cryopr... more This study aimed to evaluation the effect of different equilibration time and post thawing cryopreservation in gaga chicken semen. The research material was 10 male Gaga chickens who were 10 months old, healthy, and normal reproductive organs. Semen was collected using massage methods. This study uses a complete random design of factorial patterns with factor I being the equilibration time and factor II being the different thawing conditions. Fresh semen was evaluated macroscopically and microscopically. The sperm used had a motility of over 80%. The semen was diluted Lactate Ringer with 10% egg yolk and the addition of 7% DMSO cryoprotectant. Then the semen was equilibrated at different times and then frozen. The frozen semen was then stored in a liquid nitrogen container at a temperature of-196°C.After 24 hours, the semen was thawing at a different temperature. The evaluation of semen quality included motility, viability, plasma membrane integrity, and acrosome integrity. All data obtained were analyzed with two wayanalysis of variance. The result showed that equilibration time treatment and differential temperature of thawing yield good results on the motility and viability of Gaga chicken sperm on all treatments (P>0,01). Equilibration time of treatment for 2 hours was capable of preserving Plasma Membrane Integrity (PMI) and acrosomal integrity with a thawing temperature of 38 °C for 30 seconds or a 60 °C Thawing temperature for 5 seconds (P<0.01).

Some factors affect the success of artificial insemination, including the quality of the frozen semen. This research examined the effects of carrot extract addition in egg yolk diluent tris aminomethane on the quality of frozen semen of Boer goats. Fresh semen was collected from 5 male Boer goats aged 5-7 years, with minimum mass sperm motility of ++ and individual motility of at least 70%. The semen was diluted using tris aminomethane egg yolk added with different concentrations of carrot extract. In this experimental research, 4 treatments with 5 replications and a laboratory experiment using a completely randomized design (CRD) were carried out. The treatments in this study were P0:0% carrot extract + 80% tris aminomethane + 20% egg yolk, P1:1% carrot extract + 80% tris aminomethane + 20% egg yolk, P2: 2% carrot extract + 80% tris aminomethane + 20% egg yolk, P3: 3% carrot extract + 80% tris aminomethane + 20% egg yolk. The addition of carrot extract in egg yolk tris aminomethane had a significant effect (P<0.05) on sperm individual motility and viability. To sum up, the addition of 1% carrot extract had better results than other treatments.

This study aimed to examine the effect of adding BHT (butylated hydroxytoluene) into the diluent and storage time on the quality of Gaga chicken spermatozoa during liquid storage. The semen was collected using abdominal massage methods and divided into 4 tubes filled with diluent. For the analysis, the diluent selected was egg yolk Ringer's lactate added with BHT at concentrations of 1, 2, and 3 mM, respectively as well as a control treatment without BHT. Liquid semen was stored for 72 hours at 5°C, with spermatozoa quality monitored every 24 hours in 10 replicated experiments. The results showed that sperm motility, viability, and plasma membrane integrity were significantly higher (P<0.01) when 2-3 mM BHT was added, while acrosome integrity was elevated after the addition of 1 and 3 mM BHT. Furthermore, the addition of BHT did not significantly change semen pH. DNA damage was lower (P<0.05) when 3 mM BHT was added and mitochondrial activity was enhanced under the same condition. The concentration of malondialdehyde (MDA) was lower (P=0.05) with the addition of 3 mM BHT at 24 hours of storage. In conclusion, the addition of 3 mM BHT showed promising potential to maintain chicken sperm quality after liquid storage.

Water clover extract (WCE) was supposed to retain the semen content during freeze storage with the antioxidant compounds. The objective of this analysis was to determine the impact on the consistency of post-thawed Boer Goat Semen of the water clover extract supplement. The Boer semen was collected from 3 Boer bucks aged at 3 to 3.5 years Boer bucks which weighed at 40 to 42 kg. The semen was obtained by artificial vagina two days a week and the semen used for this research should have at least 80 percent individual motility and feasibility and maximum anomalies of 10 %. Data were collected through a survey method. The research materials were 228 first-parity (P1), 226 second-parity (P2), 147 third-parity (P3), and 73 fourth-parity (P4) heads acquired in 2011–2017. We then observed the service per conception (S/C), days open (DO), conception rate (CR), kidding interval (KI), litter size, birth weight, and fertility index (FI). The obtained data were analyzed with compare means procedure (one-way ANOVA) by using SPSS version 20 and then Least Significant Difference to determine any significant differences. The CR was computed by chi-square test. Results showed that the PE parities significantly affect (P<0.05) the S/C, DO, CR, litter size, and birth weight, but not the KI (P>0.05). The objective of this study was to examine the effects of adding Moringa leaves extracts on rabbi... more The objective of this study was to examine the effects of adding Moringa leaves extracts on rabbit does' milk production and histological changes of mammary glands during their lactation phase. Three different solvents, which are n-hexane, ethanol and mixed of nhexane and ethanol (50: 50), were used to extract Moringa leaves powder (MLP). Moringa extract was given to the does' feed by seven different treatments: 0% dosage (without extract), 0.13% dosage of HeEF, 0.26% dosage of HeEF, 0.37% dosage of EtEF, 0.74 % dosage of EtEF, 0.54% dosage of EtCE and 1.08 % dosage of EtCE. Each treatment was replicated at four does. The variables were dry matter consumption, average does' daily milk production, bunnies' daily weight, and the does' mammary gland histology. The milk production was measured twice per day in the morning (7 a.m.) and the evening (4 p.m.), obtained from the bunnies' weight gap before and after suckling. The results showed that there was a significant difference (P < 5%) in treatments' effect on dry matter consumption, daily milk production and bunnies' average daily gain (ADG). From the results, treatments with the highest daily milk production were 0.54% EtCE (134.70 ± 5.77 g / day) and 0.26% EtEF (128.83 ± 10.11 g/day). The potential effect of Moringa leaves extract on the milk production was shown by an overview of mammary gland alveoli, which indicates the increasing number of the epithelial cells as well as the alveoli number and size.

Goat ovaries were collected at a local slaughterhouse and washed twice in sterilized NaCl solution. The follicular content including cumulus-oocyte complexes (COCs), were collected by aspirating the visible small antral follicles (around 2-6 mm diameter) with a 10 ml syringe equipped with an 8-gauge needle. The oocytes with more two layers were cultured for 28 hours at 38.5 o C and 5%CO 2 . After maturation the cumulus cells were removed and the denuded oocytes were stained with 2% aceto-orcein to determine the nuclear oocyte status. The result showed that the rate of nuclear maturation in ITS-treated groups was significantly higher (P<0.05) than in the ITS-untreated group, were 38.14% a ; 61,40% b and 71,55% b , respectively for medium without ITS, 5 ng/ml ITS and 10 ng/ml ITS. This research aimed to evaluate the deterioration degree of frozen semen after 5 °C cooling and freezing of Bali cattle. The samples included 10 male Bali cattle with a body weight of 542-668 kg, from which semen was collected once a week for five weeks. The deterioration of each individual’s sperm was determined by observing two distinct straws. The parameters observed included viability, abnormalities, intact plasma membrane, and intact acrosome cap. Initial observations of the parameters were conducted following the addition of semen to diluent A1 (AD) as much as the volume of fresh semen. The semen in the AD group was not cooled and frozen. The objective of this study was to determine the effect of the different ratio of young coconut w... more The objective of this study was to determine the effect of the different ratio of young coconut water and egg yolk extender and length of storage at low temperature on sperm motility and viability of Bali bull. The method used was an experiment in the completely randomized factorial design. The first factor was young coconut water and egg yolk ratio which consisted of 5 treatments, including P0 (control) = Tris aminomethane, P1 = 85% of young coconut water + 15% of egg yolk, P2 = 80% of young coconut water + 20% of egg yolk, P3 = 75% of young coconut water + 25% of egg yolk, P4 = 70% of young coconut water + 30% of egg yolk. The second factor was the length of storage at a low temperature which consisted of 3 treatments, including 1 day, 2 days and 3 days. All treatments were replicated 10 times. Variable measured was sperm motility and viability. Results showed that the sperm motility of Bali bull in P0 (control) after 3 days of storage was 38.00%, which was significantly different (P<0.05) compared to P1 (21.50%), P2 (24.50%), P3 (28.00%), and compared to P4 (33.50%). Results also showed that sperm viability of Bali bull in P0 after 3 days of storage, which was in the amount of 65.06%, was significantly greater (P<0.05) than P1 (55.64%), P2 (58.42%), and P3 (60.91%), but did not significantly different (P>0.05) compared to P4 (63.54%). The conclusion of this study is that the use of extender which composed of 70% of young coconut water + 30% of egg yolk could maintain the sperm motility and viability of Bali bull. Therefore, this study aims to determine the effect of adding L-Carnitine, hyaluronic acid, and sucrose as well as their combination in a diluent on the quality of Gaga' chicken spermatozoa after freeze-thawing. A completely randomized design was used along with 6 treatments, namely P0, P1, P2, P3, P4, and P5, containing Ringer acetate-egg yolk (RAEY) diluent (P0), RAEY + 0.063 mM hyaluronic acid (P1), RAEY + 1 mM L-carnitine (P2), RAEY + 1 mM sucrose (P3), RAEY + 0.063 mM hyaluronic acid + 1 mM L-carnitine (P4), and RAEY + 0.063 mM hyaluronic acid + 1 mM L-carnitine + 1mM sucrose (P5). Liquid semen was packed in a 0.25 mL straw, equilibrated at 5 o C for 2 hours, and placed 3 cm above the surface of liquid nitrogen for 10 minutes. It was then immersed in nitrogen for 24 hours and thawed at 60°C for 5 seconds. The results showed that there was a significant difference (P<0.05) in the motility, viability, and kinematic variables of spermatozoa after freeze-thawing. P1, P2, P3, and P4 increased progressive motility, P1 and P4 increased curvilinear velocity, while viability increased with P4 treatment. Furthermore, the addition of Hyaluronic acid, L-carnitine, or their combination in the RAEY diluent can improve the sperm quality of Gaga' chickens.

Metode yang digunakan dalam penelitian ini adalah percobaan atau eksperimental dengan jenis rancangan percobaan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan masing-masing 10 kali ulangan. Perlakuan dalam penelitian ini adalah spermatozoa kambing Boer diencerkan menggunakan Andromed dengan perbandingan 1:4 (P1), 1:8 (P2), 1:12 (P3), dan 1:16 (P4). Hasil penelitian menunjukkan bahwa penambahan pengencer Andromed memberikan pengaruh yang sangat nyata (P<0,01) terhadap viabilitas, motilitas dan abnormalitas spermatozoa kambing Boer setelah pendinginan dan pembekuan. Media maturation were used: (1) TCM 199 + FCS 10%; (2) TCM 199 + FCS 10% + ECS 5%; (3) TCM 199 + FCS 10% + ECS 7, 5%; (4) TCM 199 + FCS 10% + ECS 10%; (5) TCM 199 + ECS 10%. Supplementation of ECS had significantly difference (P < 0.05) on expansion of cumulus cells and rate of nucleus maturation. Supplementation of ECS 5% was the best result in expanded cumulus cells and metaphase II rate: 82% and 72% respectively. This study identifies the phenotypic and reproduction performance of Indonesian swamp buffalo bas... more This study identifies the phenotypic and reproduction performance of Indonesian swamp buffalo based on specific characteristics of skin color (gray, dark grey, albino or white, and red-brownish) found in East Java province, Indonesia. This phenotypic variation is thought to be the negative effect of inbreeding. The study included phenotypic identification and DNA characterization by RAPD (Random Amplified Polymorphic DNA) method using OPA 1-4 primers. A sampling from 3 heads of buffalo blood was isolated from Madura Island purposively and a comparison was done with 2 heads of gray buffalo from Malang Regency, East Java, which represented buffalo with normal and abnormal skin coloring. This study also sought to identify the reproductive performance of swamp buffalo in both locations. Data from RAPD-DNA with OPA 1-4 primers were used for genetic similarity analysis. The results showed that there were two heads of white skinned buffalo (7.14 %) in Madura and one head (1.54%) in Malang. Meanwhile, there were five heads of red-brownish buffalo and the rest were normal (gray and grey-dark black, n = 28 and 36 heads respectively). The abnormal phenotype may be a result of simple inheritance of a codominant gene associated with the appearance of the red-brownish phenotype and considered a recessive homozygote for white skin color or these characteristics may have appeared as a result of the qualitative character of the polygenes. The similarity of the red and white buffaloes was 88.9%, while both had a similarity of 85.4% with normal gray/dark grey/black buffalo groups. The availability of males, isolated location, and uncontrolled natural service allows for inbreeding pressure, with possible adverse recessive homozygote characteristics. Advanced DNA analysis is required to more accurately confirm the presence of a skin color inheritance control gene, and it is necessary for a continuous recording to be performed to obtain more evidence of the red or white buffalo skin character traits

Early pregnancy detection becomes important as it would determine the success of breeding management as well as to achieve the best productivity. This study aimed to evaluate and observe the pregnancy rate and reproductive disorders of Brahman Cross (BX) cows and compare the results to the Ultrasonography (USG) observation. The materials of the study were 80 BX cows selected through a purposive sampling method with the following criteria: aged 2-3 years, weighed 300-500 kg, has at least calved once and had the body condition score (BCS) at 4-7 (in 1-9 scale). This study was conducted as experimental research, with 40 BX cows were artificially inseminated with Ongole crossbred (PO) semen at 0-2 h interval and the other 40 BX cows were at 8-10 h interval from the estrous detection. All of the BX cows were firstly synchronized with PGF2α (LUTALYSE) and then inseminated with PO semen (>40% motility) as according to the Indonesian standard. The pregnancy rate of the BX cows was observed through rectal palpation and 17β-estradiol hormone observation and further examined for the reproductive disorders by using USG. The obtained data were then analyzed descriptively and the results showed that 0-2 h insemination had a higher success of pregnancy (32.5%) compared to the 8-10 h interval (27.5%). Moreover, the highest 17βestradiol in the inseminated BX cows was at 5,765 ng/ml, while the lowest was at is 0.41 ng/ml. The USG observation showed that 6 BX cows (7%) in this study had a follicular cyst, 2 BX cows (2.25%) had persistent corpus luteum and 1 BX cows (1.25%) had ovarian hypofunction.

One strategy to increase population is to produce frozen semen with good quality. Semen production is influenced by libido which is known through male sexual behavior. The purpose of this study was to determine the effect of sexual behavior on semen production. The research material included 3 OC cattle studs aged 4-5 years with a body weight of 500-600 kg. Semen is collected twice a week using the artificial vaginal method. The method used in the study was observational. Sexual behavior parameters include libido and ejaculation duration. Semen production variables are known by calculating total spermatozoa and total motile spermatozoa. Data were analyzed using Randomized Block Design (RBD) consisting of 3 treatments and 10 replications. If there is a difference, continue with Duncan's Multiple Distance Test. The relationship between sexual behavior and semen production was calculated by Pearson correlation and its effect was calculated by linear regression. The results showed that the length of libido was not significant (P> 0.05), the duration of ejaculation was significantly (P <0.01), the total spermatozoa was significantly (P <0.01), and the total motile spermatozoa was significantly (P <0.01) in all three male males. The relationship between the duration of libido and semen production was insignificant (P> 0.05), while the duration of ejaculation had a significant relationship (P <0.05) to total spermatozoa with R2 = 22.15% and total motile spermatozoa with R2 = 25.84 %. The conclusion of the study was that there was an influence on sexual behavior on semen production in OC cattle.
