Online, or on-the-fly, parameter identification methods are therefore necessary. The challenge of online methods is that one must continuously estimate parameters as experimental data becomes available. The existing techniques in the context of time-dependent partial differential equations exclude the case where the system depends nonlinearly on the parameters. Here, we test the idea that “foraging tempo” -- the rate of successive target selections -- helps determine patterns of behaviour in these tasks. Previously, we established that the majority of target selections during unconstrained foraging happen at regular, rapid intervals, forming the “cruise phase” of a foraging trial. Furthermore, we noted that when the temporal interval between cruise phase responses was longer, the tendency to switch between target categories increased. To directly explore this relationship, we modified our standard iPad foraging task so that observers had to synchronize each response with an auditory metronome signal. Hepatitis C virus (HCV) infection may cause chronic hepatitis, liver cirrhosis, and hepatocellula... more Hepatitis C virus (HCV) infection may cause chronic hepatitis, liver cirrhosis, and hepatocellular carcinoma. HCV exploits cellular machineries to establish persistent infection. Herein we demonstrated that ubiquitin conjugating enzyme E2S (UBE2S), a member of ubiquitin conjugating enzyme family (E2s), was downregulated by ER stress caused by HCV in Huh7 cells. UBE2S interacted with domain I of HCV NS5A and degraded NS5A protein through Lys11-linked proteasome-dependent pathway. Overexpression of UBE2S suppressed viral propagation, while depletion of UBE2S expression increased viral infectivity. Enzymatically inactive UBE2S C95A mutant exerted no antiviral activity, suggesting that ubiquitin-conjugating enzymatic activity was required for the suppressive role of UBE2S. RAD51-associated protein 1 (RAD51AP1) is a member of the multiprotein complexes postulated to car... more RAD51-associated protein 1 (RAD51AP1) is a member of the multiprotein complexes postulated to carry out RAD51-mediated homologous recombination and DNA repair in mammalian cells. In the present study, we showed that hepatitis C virus (HCV) NS5A directly bound RAD51AP1 and increased protein level of RAD51AP1 through modulation of the ubiquitin-proteasome pathway. We also demonstrated that RAD51AP1 protein levels were increased in the liver tissues of HCV-infected patients and NS5A transgenic mice. Importantly, NS5A impaired DNA repair by disrupting RAD51/RAD51AP1/UAF1 complex and rendered HCV-infected cells more sensitive to DNA damage. Silencing of RAD51AP1 expression resulted in a decrease of viral propagation. We further demonstrated that RAD51AP1 was involved in the assembly step of the HCV life cycle by protecting viral RNA. This article proposes a conceptual framework to bring about a broader awareness of clients' lives and transactional processes of change over the life course. The framework draws attention to transactional processes by which individuals, situated in life contexts, change and adapt over the life course and, in turn, influence their contextual settings and broader environments. This framework is based on (a) basic tenets derived from foundational theories taking a life course perspective to change, and (b) transactional processes identified from relevant pediatric rehabilitation models that bring these foundational theories into the pediatric rehabilitation sphere. The aim of this 'perspectives' paper is to apply the six core tenets of the LCHD model and the concept of health development trajectories to individuals with lifelong neurodevelopmental conditions. We share the perspective that modifiable aspects of the environment often restrict health development; we then advocate that children, beginning at a young age, should engage in 'real-world' experiences that prepare them for current and future social roles.
