S. mutanscan form colonies and biofilms through adhesion mechanisms on the tooth surface. The use of chitosan as an antimicrobial material has been widely developed in dentistry, especially on the nanoscale, because nanomaterials can spread quickly throughout the biofilm structure. The use of hydroxyapatite (HAP) in conservative dentistry has been developed to protect teeth from S. mutans. This research aims to see the ability of nano chitosan hydroxyapatite (KNHP), if combined in gel form, does it prevent S. mutansfrom forming biofilms on the enamel surface. The research material was S. mutansserotype c and saliva collected from a volunteer. The biofilm mass profile was determined on the surfaces of teeth implanted on acrylic resin and incubated for 24, 48, and 72 hours, then observed under a counter colony lens. Determination of S. mutans CFU after interacting with nano chitosan hydroxyapatite for 24, 48, and 72 was then examined with spectrophotometry at a wavelength of 610 nm OD value of 0.08-0.1, which is equivalent to 0.5 McFarlan (1.5x 108). Then the determination of the interaction power of KNHP and HAP with S. mutanswas done by UV / VIS with a wavelength of 250-600 nm. The results showed that the mass profile of biofilms at 24 hours still showed colony growth after being given KNHP and HAP, whereas, at incubation times at 48 and 72 hours, there was no S. mutans growth. The interaction between KNHP and S. mutans with UV-VIS at 24, 48, and 72 hours incubation time has higher interaction power than HAP.

Composite resin is a filling or restoration material that has a similar color to the teeth, is in... more Composite resin is a filling or restoration material that has a similar color to the teeth, is insensitive to dehydration, and is relatively easy to manipulate. One of the nanotechnology developments in dentistry is nano fill and nanohybrid composite resin with minute particle sizes, which has a different value in its physical, mechanical, and optical properties. In the oral cavity, composite resin restoration material comes in contact with various salivary pH, acidic or alkali that can affect physical property changes, including surface roughness. Chitosan is a biomaterial continuously being developed because it has many medical benefits and has proven safe to use on people. This study analyzes composite resin restoration strength when nanoparticle chitosan high molecular is added 1% to its surface degradation. Samples are 36 human premolar teeth randomly classified into 12 groups (r=3). Each group is classified based on different filling materials and salivary pH. Scanning Electron Microscopy (SEM) samples are tested to see the surface degradation. The result of this study shows that using composite resin, either nanofiller or nanohybrid, will go through salivary degradation on pH 3, 5, 7, dan 10. The lower the pH, the higher the degradation, Adding 0,1% chitosan on nano fill and nano hybrid shows that there is still degradation in the restoration surface. 0,1% Chitosan added in nano hybrid composite resin on pH 7 offers surface stability, although degradation is still found. The 0,1% chitosan added into nano fill composite resin shows a different value although still lower than nano hybrid composite added with 0,1% chitosan on surface degradation. Adding 0,1% high molecular Chitosan to both nanocomposite resins shows a better value when compared to the composite resin that is not infused with Chitosan.

Together with vehicle aqueous, glycerine, polyethylene glycol, and chitosan can increase calcium ion diffusion, pH balance, and anti-bacterial power. To analyze the effect of different vehicles on alkaline pH changes, diffusion of calcium ions, roughness, and frequency of chemical elements in the root canal after applying calcium hydroxide. A total of 100 dental samples in 5 groups of vehicles were examined for pH using a pH meter, calcium ion diffusion using Atomic Absorption Spectrophotometric, and surface roughness of the root canals chemical compounds were analyzed by SEM-EDX. All the vehicles can maintain the balance of pH changes to alkaline in the root canal wall (p<0.05: 0.00). Maximum calcium ion diffusion occurs on day 30, especially in the vehicle Chitosan and Polyethylene glycol (p <0.05: 0.00). Vehicle chitosan and glycerine more strongly reduce root surface wall roughness after calcium hydroxide medicament (20% roughness score). Specifically, vehicle chitosan can increase calcium (21.71%) compared to control and other vehicles in the root canal wall (p<0.05: 0.02). The chitosan vehicles and other vehicles can maintain an alkaline pH balance, diffusion of calcium ions, and decrease in roughness and increase in the quantity of calcium in the root canal wall.

Chitosan is an irrigant in dentistry has the properties of biocompatibility, biodegradability, bioadhesion, and not toxic to human cells. Several studies have suggested the use of ultrasonics to enhancing the action of irrigants.Aims:The aim of the study was to investigate the bacterial growth and surface roughness of the root canal surface after irrigation and agitation with passive ultrasonic irrigant.Subjects and Methods:Experimental research with randomized block design obtained a sample size for each group of 9 samples with a total number of 27 teeth divided into three treatment groups. Statistical analysis used was one-way analysis of variance.Results:The irrigation material for nano-chitosan high molecular 0.2% with passive ultrasonic irrigation (PUI) activation was shown to cause lysis on surface of bacterial cell walls. There was no significant difference between the roughness values in all treatment groups.Conclusions:The irrigation of root canal treatment with 0.2% high molecular nano-chitosan with the addition of PUI activation had significant antibacterial activities against E. faecalis.

Wound healing is a complex and integrated process that requires the collective roles of various c... more Wound healing is a complex and integrated process that requires the collective roles of various cells and tissues of the palatal mucosa. During the proliferation phase, the sum of fibroblasts increases in response to damaged oral tissues. Thus, the number of fibroblast cells in the palatal mucosa is a substantial indicator of wound healing. Binahong leaves possess the potential to accelerate wound healing by stimulating fibroblast proliferation. This study aimed to formulate and evaluate the effect of Binahong leaves extract (BLE) gel on wound healing of the palatal mucosa by investigating the fibroblast cell count in Wistar rats. This experimental study was carried out in several stages which included the collection and processing of fresh Binahong leaves followed by extraction with 70% ethanol using maceration method. The extract was formulated to be a gel product with a concentration of 3%, 5%, and 7%. BLE gel was administered to Wistar rats which were deliberately injured at the palatal mucosa by excision. Wound healing was assessed using hematoxylin-eosin staining and the number of fibroblast cells was counted. The result of fibroblast proliferation was analyzed by One Way ANOVA followed by Bonferroni’s test with a significance level of 95%. There were significant differences in fibroblast cell proliferation between all test groups except between base gel and Aloclair® gel, base gel and BLE gel 7%, Aloclair® gel and BLE gel 7%, BLE gel 3% and BLE gel 5%. Application of BLE gel 3% promotes better wound healing of the palatal mucosa of Wistar rats.

This study aims to examine the potential of gel chitosan oligosaccharide (COS) as a cavity cleanser on the adhesion of the restoration to the cavity wall. Thirty-two mandibular premolars with class V restorations are divided into two groups with 2% COS gel and 2% chlorhexidine digluconate (CHX). Samples were immersed in 2% methylene blue, cut longitudinally, observed under a stereomicroscope with 1x magnification, and scored 0-3. The Mann-Whitney test was used to see the potential of gel COS as a cavity cleanser. The results showed a significant difference between the two groups at the edge of the enamel junction restoration (p=0.038) and the dentin junction restoration (p=0.027). The mean microleakage score in group 1 showed better results at the enamel junction (1.19± 1.328) and the dentin junction (2.00±1.211). It shows that 2% COS gel has the potential as a cavity cleanser. There was a significant difference between the two groups as a cavity cleanser on the adhesion of adhesive restorations to the cavity wall.

To investigate the effect of different ferrule height designs on the resistance of dental fractures following endodontic treatment, using AUTOCAD as the basis for non-linear finite element analysis. Five models of 3D maxillary first incisors were constructed using AutoCAD 2016 with five different ferrule heights (mm) 0, 0.5, 1, 1.5, and 2, meshing and input mechanical properties, followed by loading 25 N and 170 N at an angle of 135° in the tooth palatal area which was analyzed by Finite element analysis (FEA) using ANSYS v 17.0 software. Von Mises analysis shows the maximum and minimum deformation values, equivalent elastic strain, and equivalent stress. The ferrule's total deformation value in the 25 N simulation was the highest at 0 mm (0.0093 mm) and the lowest at 2 mm (0.0091 mm). The simulation load was 170 N, the highest at 0 mm (0.0632 mm) and the lowest at 2 mm (0.0618 mm). The maximum equivalent elastic strain value of ferulle is highest at 1 mm (0.034814 mm) and lowest at 0 mm (0.03245 mm). The maximum equal stress value decreases in models with ferrules. In the 25 N and 170 N simulations, the highest values on the 3D model are at 0 mm (13,008 MPa and 88,452 MPa) and the lowest at 1 mm (11,567 MPa and 78,655 MPa). Conclusion.The addition of ferrule height can increase dental fractures' resistance after endodontic treatment by reducing deformation. Ferrule decreases stress on the tooth's labial cervical area and enlarges the strain on the cervical palatal dentine by holding non-axial loads from the palatal side.

Using a suitable sealer material can increase penetration and adhesion to the root canal wall. Epoxy and chitosan resins have been reported to act as endodontic sealers with good penetration and adhesive properties. The evaluation of the penetration and adhesion properties of epoxy resin sealer and nano chitosan high molecular on the tubules dentin in root canal. The absorption and penetration assay of sealer used to the methylene blue dye assessment, while the confirmation the adhesive strength using SEM (Scanning electron microscope). The addition of chitosan nanoparticles 1% allowed the epoxy sealer group to have better penetration and absorption capabilities in tooth root canals. In addition, this group increased the adhesion of the root canal wall compared to other groups because it increased its density and adapted well to the entire dentine tubules on the root canal wall. The sealer penetration value was significantly different between the treatment groups (p<0.005);0.001) with a strong relationship (r=0.89). Epoxy resin sealer with the addition of 1% chitosan high molecule nanoparticle has excellent absorption and penetration properties in root canal walls, increasing adhesion to root canal walls.

The Glucosyltransferase (GTF) enzyme produced by S. mutans contributes to the virulence factor by converting sucrose into fructose and glucans used in biofilm formation. Probiotics such as yogurt and kefir of bacteria could also affect the oral ecology and potentially inhibit the GTF enzyme produced by S.mutans. The assessment of probiotic yogurt and kefir to inhibition of glucosyltransferase enzyme of Streptococcus mutansATCC25175.This research was done in an experimental post-test-only group design using Microtiter Plate Assay by measuring the Optical Density (OD) of the ELISA reader. Two aspects were analyzed; first, the evaluation of probiotic products concerning their biomass index, biostability, and pH alteration; second, the assessment of GTF enzyme activity and fructose release by S.mutans. Both tests aimed to measure probiotics' effectivity in inhibiting the GTF enzyme's movement to glycolate glucose into fructose. Both test materials were analyzed in 25%, 50%, and 100% concentrations within 24 hours, 48 hours, and 72 hours incubation. In evaluating fructose release by the GTF enzyme of S.mutans, probiotic yogurt produced a more significant effect than probiotic kefir (p<0,05). This activity was not influenced by incubation periods or probiotic concentrations (p>0,05). The probiotic yogurt and kefir inhibited the GTF enzyme activity and reduced the fructose release by S.mutans.

The possible connection between chronic oral inflammatory processes, such as apical periodontitis... more The possible connection between chronic oral inflammatory processes, such as apical periodontitis and periodontal disease (PD), and systemic health is one of the most interesting aspects faced by the medical and dental scientific community. Chronic apical periodontitis shares important characteristics with PD: 1) both are chronic infections of the oral cavity, 2) the Gram-negative anaerobic microbiota found in both diseases is comparable, and 3) in both infectious processes increased local levels of inflammatory mediators may have an impact on systemic levels. One of the systemic disorders linked to PD is diabetes mellitus (DM); is therefore plausible to assume that chronic apical periodontitis and endodontic treatment are also associated with DM. The status of knowledge regarding the relationship between DM and endodontics is reviewed. Upon review, we conclude that there are data in the literature that associate DM with a higher prevalence of periapical lesions, greater size of the osteolityc lesions, greater likelihood of asymptomatic infections and worse prognosis for root filled teeth. The results of some studies suggest that periapical disease may contribute to diabetic metabolic dyscontrol.

Still, it has the disadvantage of leavinga residue on the surface of the root canal wall. Hence, it can interfere with the setting time and adhesion of the sealermaterial to the root canal wall. This study aims to determine the ability of calcium ion diffusion in one-third of theroot canal after administration of nanohydroxyapatite medicament with a different vehicle than calcium hydroxide.This study used 24 mandibular premolars extracted, decorated, and prepared, then grouped into four treatmentgroups. Based on the paired T-test, the study results showed no significant effect of calcium ion diffusion in group B(p = 0.739) on the 7th and 14th days. The ANOVA test results showed no significant difference in calcium ion diffusion between groups A, B, and C. on the 7th day of testing (p=0.773) and the 14th day (p=0.661).
