EXERCISE 3
MICROBIOLOGICAL CULTURE MEDIA

PREPARATION OF SOLID PLATED AND TUBED MEDIA

Nutrient Agar container label

Directions: Suspend 28.0 grams in the 1000 mL distilled water. Heat to boiling to dissolve the
medium completely. Sterilize by autoclaving at 15 lbs pressure (1210C) for 15 minutes. Cool to
45-500C. If desired the medium can be enriched with 5-10% blood or other biological fluids.
Mix well and pour into sterile Petri dishes.

1. Determine the amount of dehydrated medium needed to prepare 50 nutrient agar plates.
Include amount for 2 additional plates as excess to compensate for compounding losses.

2. Determine the amount of dehydrated medium needed to prepare 15 butts, 20 slants, and
25 butt-slants. Include amount for 2 additional units of each as excess to compensate for
compounding losses.

3. Illustrate the different solid tubed media formed.

4. Give examples of specific applications for different types of solid tubed media
___________________________________________________________________________________________________________
Deep/Butt is used for motility, the tube of agar is stabbed into the agar without touching the
bottom of the test tube. This is to observe the movement of microorganisms. Slant allows greater
___________________________________________________________________________________________________________
surface area of microorganisms to move around, which is helpful in maintains the bacterial culture.
___________________________________________________________________________________________________________
Butt-Slant on the other hand allows the culture media to grow in both aerobic and and anaerobic.

PREPARATION OF LIQUID MEDIA

Nutrient Broth container label

Directions: Suspend 13.0 grams in 1000 mL distilled water. Heat if necessary to dissolve the
medium completely. Dispense in tubes or flasks as desired. Sterilize by autoclaving at 15 lbs
pressure (1210C) for 15 minutes.

1. Complete the table

Parameter Nutrient Broth Nutrient Agar
Composition - Simple peptone - Peptone
- Beef extract - Beef extract/ yeast extract
- Solidifying agent

Applications Cultivation of different Used to cultivate and support

variety of fastidious and non- growth of broad ranged of non-
fastidious organisms fastidious organisms

Sign of microbial growth - Turbidity Visible growths of colonies

- Precipitation and pellicle
formation

2. Give 5 examples of microorganisms that may be grown in Nutrient Broth.

___________________________________________________________________________________________________________
The five examples of microorganisms that may grow in the Nutrient Broth are:
Streptococcus pyogenes, Psdeudomonas aeruginosa, Escherichia coli, Staphylococcus aureus,
___________________________________________________________________________________________________________
and Salmonella Typhi.

3. Discuss the implication/s if spore strip was missed out during the preparation of sample for
autoclave and possible remedy for this scenario.
___________________________________________________________________________________________________________
If the spore strip was missed out during the preparation of sample for autoclave, it would
be difficult to identify whether the sample has been completely sterilized. A way to fix this
___________________________________________________________________________________________________________
situation is to incubate the sample for 24 hours and if no apparent organism grow then it has
___________________________________________________________________________________________________________
been fully sterilized.
