SECTION 4 ADJUSTMENT
4.1 OPTICAL ALIGNMENT

4.1.1 Required Tools


- Hex wrench (1.5 mm and 2.5 mm)
- Screw Driver (Philips and slot headed)
- Laser power meter
- Latex particles (LATEX particles 4207A (10ML/VIAL) and LATEX PS-FLUO RED-2.5) [AA]
- Oscilloscope

4.1.2 Optical Alignment Outline


When replacing Laser Diode
Follow from below step 1
When replacing
Flow Cell Follow from below step 2
(1) Adjust the angle of the LD unit so that incident and reflect beam are aligned using a compact mirror
after removing Flow Cell and FSC Detector unit.
(4.1.8 Adjusting Laser Diode Angle (when replacing Laser Diode)
(2) Install Flow Cell unit, and then adjust the angle of Flow Cell so that incident and reflect beam are
aligned. (4.1.9 Installing Flow Cell Unit No.11 and 4.1.10 Adjusting Flow Cell Unit No.11 Angle)
(3) Adjust the initial position of the LD unit.
(4.1.12 Confirmation/Adjustment of Laser Diode Initial Position)
1) Z-direction rough adjustment by aligning the corner of the laser diode cylinder and laser diode
base.
2) X-direction rough adjustment by aligning optical base slotted portion and LD base.

(4) Adjust LD unit X-direction roughly so that laser beam can position at the center of the Flow Cell
image behind the FSC Detector location (now unit removed).

(5) Install FSC Detector at the specified position from the Flow Cell.
(4.1.14 Installing Photo Detector No.30)
(6) Adjust X and Y direction of the LD unit so that laser beam can be positioned at the center of the
beam stopper on the FSC Detector.

(7) Adjust the initial position of the SFL Detector (X,Y and Z direction).
(4.1.16 Confirmation/Adjustment of Photo Detector No. 28 (1)) In case of Flow Cell replacement
only (no LD unit replacement), skip this step.
(8) Check FSC signal using an oscilloscope. (4.1.19 Optical Alignment (Rough)) Adjust X and Z-direc-
tion of LD unit so that FSC signal width will become narrower than specified limit. Adjust X-direction
of FSC Detector so that FSC signal is lowest.
(9) Adjust following units by observing SFL signal using an oscilloscope. Verify the results in Fine
mode menu, and then fix the units. (4.1.20 Optical Alignment (fine))
- Z-direction of SFL Detector: to make signal width narrower
- X-direction of SFL Detector: to make uniform signal
- X-direction of SFL Detector: to make higher signal
- Y and Z-direction of SFL Detector: to make uniform signal

Cautions)
In case LD unit or Flow Cell angle adjustment is not well done, uniform signal will be seen for a
short time only (unstable). If it is difficult to get SFL signal, check if Flow Cell image at the slit of the
SSC Detector is clear and sharp when shining a flash light toward flow cell. If NOT, adjust SFL
Lens X-direction to get sharp and clear image on the slit.

(10) Verify the results in Fine mode menu, and then fix the units. (4.1.20 Optical Alignment (fine))
(11) Adjust the SSC Detector left or right so that Flow Cell image can be aligned at the slit of SSC
Detector. (Step (9) in 4.1.20 Optical Alignment (fine))
(12) Adjust X-direction of the FSC Detector so that FSC signal becomes the lowest. (This means direct
laser beam is interrupted at the beam stopper.) And then, fix the FSC Detector.
(Step (14) in 4.1.20 Optical Alignment (fine))

Caution)
Adjustment for FSC(W) data is not required as long as FSC(W) data is within the specified limit.

4.1.3 Replacing Flow Cell
(1) Disconnect tubing from nipple on SV 11.

(2) Connect a syringe and push CELLPACK in the flow cell to waste chamber.

(3) Carefully remove thermistor No.48 upward so that a strong stress is not applied onto the flow cell.

4.1.4 Removing Photo Detector No.30


(1) Remove Photo Detector No.30 by loosening fixing screws (Screw Binding M3X6). Cut a tie wrap
tying wiring cord. Disconnect the connector from the PCB No.2168.

4.1.5 Removing XS_FCM Detector Block Cover B
(1) Remove XS_FCM Detector Block Cover B by loosening fixing screws (Screw Binding M3X6).

(2) Lift and move FCM Detector Block toward you by loosening fixing screws.

4.1.6 Removing Flow Cell Unit
(1) Remove fixing screw (Screw Hex-Socket Bolt M3X8) for flow cell unit and carefully pull the flow cell
unit downward so that the flow cell is not damaged.

(2) Disconnect tubing from SV 34 and Reaction Chamber.

4.1.7 Adjusting Laser Power
(1) Turn on IPU computer.
(2) Enter “sysmex” in the Logon Name.
(3) Enter “c”, “9”, “.” and “0” in the password.
(4) Turn on XS main unit.
(5) When below screen appears, select “Ignore all Errors except Sampler Errors” and click OK.

(6) Select MENU – Controller.


(7) When below screen appears, double-click Service.

(8) When below screen appears, double-click Laser Power to activate laser. (Wear protection goggle
when the LD is activated.)

(9) Set the wave length of the laser power meter to 633 nm and place the probe in front of the laser at
a little inclined angle as shown to prevent the reflect beam from going back to the laser diode.
(Laser power controlling device in the laser diode may malfunction when a beam is incoming into
the laser diode.)

(10) Adjust VR1 so that the laser power is 3.4 +/- 0.1 mW with a ceramic screw driver. (A ceramic screw
driver is recommended to prevent occurring static accident.)

4.1.8 Adjusting Laser Diode Angle (when replacing Laser Diode)
(1) Remove Flow Cell by Step 4.1.3 Replacing Flow Cell.
(2) Place XS Laser Diode Angle Adjustment Tool or equivalent (hand mirror). (Place the tool or a hand
mirror by pressing to arrow directions to reflect the laser beam to the laser diode.)

(3) Place a hex wrench between flow cell and laser diode, and adjust the laser diode angle so that the
reflected light and laser light are one line.

(4) Click OK to turn off the laser.

4.1.9 Installing Flow Cell Unit No.11
(1) Tighten temporary the Flow Cell Unit No.11 with Screw Hex-Socket Bolt M3X8.

4.1.10 Adjusting Flow Cell Unit No.11 Angle


(1) Double-click Laser Power on the service screen to activate laser.
(2) Place a hex wrench between flow cell and laser diode, and adjust the flow cell angle so that the
laser beam and reflected beam from the flow cell on the wrench are one line.

(3) Tighten fixing screws so that the flow cell angle doe not move.
(4) Confirm that the laser beam and reflected beam on the wrench are one line after tightening the fix-
ing screws.
(5) Click OK on the Laser Power screen to turn off the laser.

4.1.11 Installing XS_FCM Detector Block Cover B
(1) Tighten fixing screws so that tubing of the bottom of the flow cell is parallel to the bottom surface of
the FCM Detector Block Cover B.

4.1.12 Confirmation/Adjustment of Laser Diode Initial Position


(1) Confirm/Adjust the Laser Diode initial position (forward/backward) as follows.

(2) Confirm/Adjust the Laser Diode initial position (right/left) as follows.

(3) Do not loosen following fixing screws.

4.1.13 Adjusting Laser Diode Position
(1) Double-click Laser Power on the service screen to activate laser.
(2) Place a white paper as shown and adjust Laser Diode R/L position so that two lines of flow cell are
both sides of the beam by turning adjustment screw.

(3) When step (2) is unclear to observe the two lines of the flow cell, adjust the Laser Diode R/L posi-
tion by observing laser beam in flow cell as shown below. When laser beam hits the sample path in
the flow cell, reflected beam can be seen.

4.1.14 Installing Photo Detector No.30
(1) Click OK on the Laser Power screen to turn off the laser.
(2) Fix the Photo Detector Block No.30 so that the distance between flow cell and Photo Detector Bloc
No.30 is 7 mm by tightening screw binding M3X6.

4.1.15 Adjusting Beam Stopper Position


(1) Double-click Laser Power on the service screen to activate laser.
(2) Adjust beam stopper position so that the beam image is on the center of the beam stopper as
shown.

(3) Click OK on the Laser Power screen to turn off the laser.

4.1.16 Confirmation/Adjustment of Photo Detector No. 28 (1)
(1) Confirm/Adjust the adjustment plates position as follows. (Loose fixing screw when moving adjust-
ment plates.)

4.1.17 Confirmation/Adjustment of Photo Detector No. 28 (2)
(1) Confirm/Adjust the Photo Detector No.28 initial position as follows. (Loosen fixing screws (Screw
Hex-Socket Bolt M3X6) when moving Photo Detector No.28.)

4.1.18 Connecting Oscilloscope
(1) Remove rear panel.
(2) Connect probes of the oscilloscope as shown. (Both FSC and SFL should be connected because
SFL signal is triggered by FSC signal.)

(3) Set the oscilloscope as follows.


Gain: 10mV
Time: 10 µ sec.

4.1.19 Optical Alignment (Rough)


(1) Select Menu  Controller ( Maintenance ( Auto Rinse.
(2) Confirm that no background error is generated after performing Auto Rinse.
(3) Click Close to close the screen.
(4) Prepare latex material for optical alignment as follows.
FSC Latex: CELLPACK 30mL and 4207A six drops
SFL Latex:CELLPACK 50mL and PS-FLUO RED-2.5 two drops [AA]
For XS-1000iC instruments, CELLSHEATH may be used in place of CELLPACK.

Warning
Since undiluted latex is easy to coagulate, shake it enough and add to CELLPACK.
Use the latex materials within 3 hours after making it. Do not add latex to made latex
materials after a time. (Use the latex materials within an hour after making for confir-
mation of Optical Alignment (fine))
Since the fluorescence intensity becomes weak when PS-FLUO RED-2.5 (latex for
fluorescence) receives light, store PS-FLUO RED-2.5 in dark place. [AA]
Use diluted latex in a bottle.Store latex in room temperature and do not freeze it.

(5) Double-click Optical Axis Adjustment (rough) in service.
(6) Pour 1 mL of FSC latex to reaction chamber and press start switch.

(7) Adjust laser diode position in the following manner:


1. Turn adjustment screw A so that the FSC wave on the oscilloscope becomes narrower.
2. Turn adjustment screw B so that the FSC wave on the oscilloscope has minimum variation.
(8) Adjust the beam stopper R/L position so that FSC wave is lowest by turning adjustment screw C.

(9) Adjust FSC(W) approximately 0.3 or lower. If the value does not become 0.3, repeat steps (6) to
(8).
(10) Slightly loosen the screws circled in red (4 locations) shown in the diagram on the next page.
(11) Pour 1 mL of SFL latex to reaction chamber and press start switch.
(12) Adjust Photo Detector position so that the SFL wave on the oscilloscope is highest by turning
adjustment screw D.
(13) Adjust Photo Detector Position so that the SFL wave on the oscilloscope is highest and has mini-
mum variation by turning adjustment screw E and F.
(14) If the SFL wave has variation, repeat steps (12) and (13).
(15) Adjust the Photo Diode Position so that the SFL(W) is approximately 0.3 or lower by turning adjust-
ment screw D, E and F.
(16) Click Close in the screen.

4.1.20 Optical Alignment (fine)


(1) Double-click Optical Axis Adjustment (fine) in service.
(2) Pour 1 mL of FSC latex to reaction chamber and press start switch.
(3) Enter appropriate value into Gain and click Send so that the FSC(X) value is within the below
range.
FSC(X): 60 +/- 30ch
(4) Run FSC latex three times after adjusting Gain and confirm that the FSC(W) is within the below
range.
FSC(W)0.35
(5) Pour 1 mL of SFL latex to reaction chamber and press start switch.
(6) Enter appropriate value into Gain and click Send so that the SFL(X) value is within the below
range.
SFL(X): 130+/-50ch
(7) Run SFL latex three times after adjusting Gain and confirm that the SFL(W) is within the below
range.

(8) Pour 1 mL of SFL latex to reaction chamber and press start switch. Tighten following fixing screws
while checking SFL related values remain unchanged.
1) Fixing screws for laser diode leaf spring: four screws
2) Fixing screws for Laser diode base: two screws
3) Fixing screws for Photo Diode No.28: four screws

(9) If the value is out of the range, repeat steps (5) – (8).

(10) Shine the flow cell with a flashlight perpendicularly to flow cell and observe the image of the Photo
Detector No.29 on the mirror of the Photo Detector No.28. Adjust the Photo Detector No.29 posi-
tion so that the slit is in the center of the flow cell shadows.

(11) Change the probe of the oscilloscope from FSC to SSC.


(12) Pour 1 mL of SFL latex to reaction chamber and press start switch. Adjust Photo Detector No.29
position so that the SSC wave is highest and minimum variation by turning adjustment screw D.
(13) Pour 1 mL of SFL latex to reaction chamber and press start switch. Confirm that below values are
within the below range.
SSC(X): 50+/- 40 ch
SSC(W): 0.35

(14) After completing adjustment, fix two fixing screws.

(15) Pour 1 mL of FSC latex to reaction chamber and press start switch. Adjust beam stopper position
so that the FSC wave is lowest by turning adjustment screws C.
(16) After completing adjustment, tighten four fixing screws.

4.2 Mechanical Adjustment

4.2.1 Piercer Position Adjustment


(1) Replace adapter with XS Adapter (13 mm diameter) (P/N: 442-3086-2).

(2) Select Menu  Controller  Service Piercer/Pipette Adjustment.


(3) Select Normal for Sample Tube Type.

4.2.2 Adjusting Rinse Cup Position

(1) Select Menu  Controller  Service Piercer/Pipette Adjustment.


(2) Click Init. Position and adjust the piercer height so that the piercer tip and Rinse Cup bottom are
same height by clicking Up or Down key.

4.2.3 Adjusting Aspiration Position

(1) Select Menu  Controller  Service Piercer/Pipette Adjustment.


(2) Click Aspiration Position and adjust the piercer position so that the piercer tip is as shown position
by clicking Up, Down, Front and Back key.

4.2.4 Adjusting RBC Chamber

(1) Select Menu  Controller  Service Piercer/Pipette Adjustment.


(2) Click RBC/HGB Chamber and adjust the piercer position so that the piercer tip is in the center of
the chamber by clicking Up, Down, Front and Back key.

4.2.5 Adjusting Reaction Chamber

(1) Select Menu  Controller  Service Piercer/Pipette Adjustment.


(2) Click WBC Chamber and adjust the piercer position so that the piercer tip is in the center of the
chamber by clicking Up, Down, Front and Back key.

(2) After adjusting, click Save and Close.

4.2.6 Sampler Adjustment
(1) Select Controller  Service  Tube Holder Adjustment and click Set Position.

(2) Adjust Sample Set Unit Cover position so that the cover and chassis are flat by clicking Back or
Front.

(3) Click ID Reader Position.

(4) When movement completes, click Start in the Tube Rotation. Adjust the Sample Set Unit position
so that the test tube rotates by clicking Back and Front key. Adjust the Sample Set Unit position so
that the test tube rotates discontinuously by clicking Front key.
Move Sample Set Unit backward – 15 pulses by click in Back key.
Confirm that the test tube rotates continuously. Click Save for saving the position. Click Stop in the
Tube Rotation and Close key.

(5) Pull Sample Set Unit manually and set the test tube with the barcode facing to the barcode reader.
Select Tube Holder Adjustment and click ID Reader Position. Find the BR Unit No. 1 position so
that the barcode reader can read barcode at front most position and back most position. Move the
BR Unit No. 1 position so that the BR Unit No. 1 is placed the center of the previous two positions.

(6) Tighten left fixing screw of the BR Unit No. 1. Click Close to close the window. Tighten right fixing
screw of the BR Unit No.1
(7) Set two racks to the sampler, select Controller  Service  Hand Adjustment and click Set Posi-
tion and Down.

(8) Adjust Hand height position so that the gap between rack and hand is 1 to 2 mm by adjusting rod
height position.

(9) Click Set Position  Catch. Place a test tube in the hand and click Down. Adjust the test tube posi-
tion so that the test tube is center of the Sample Set Unit by clicking Back, Front, Left or Right.

(10) After completing adjustment, click Save  Up and remove the test tube from the Hand.
(11) Click Rack-01  Catch. Place a test tube in the hand and click Down. Adjust the test tube position
so that the test tube is center of the Rack-01 by clicking Back, Front, Left or Right.

(12) After completing adjustment, click Save  Up and remove the test tube from the Hand.
(13) Click Rack-02  Catch. Place a test tube in the hand and click Down. Adjust the test tube position
so that the test tube is center of the Rack-01 by clicking Back, Front, Left or Right.

(14) After completing adjustment, click Save  Up and remove the test tube from the Hand.

4.3 Hydraulic Adjustment

4.3.1 Adjusting RBC Clog Level

(1) Select Menu  Controller  Service  RBC Clogs Level.


(2) When “RBC Clog Error” occurs, click Close to close error screen.
(3) RBC Clog Level appears in real time.
(4) Enter “99” in Target box and click “>>>” -> “Send” ->”Continue”.
(5) Repeat step (4) until RBC Clogs is 99 +/- 1. Click Close when RBC Clogs becomes 99 +/- 1.

(5) Confirm RBC Clogs is 99 +/-1

(3) Click Send

(4) Click Continue

(1) Enter 99 (2) Click >>>

4.3.2 Adjusting HGB Blank


Note: Digital VR values inversely adjust blank values, i.e., increasing digital VR value, decreases HGB
blank value.

(1) Confirm that 30 minutes past after turning on the main unit.
(2) Select Menu  Controller  Service  Sensitivity.
(3) Enter appropriate values into HGB blank and ASP. Sensor blank, and click Send. Repeat this step
until value comes within Adjustment Value range as shown in table below.

4.3.3 Adjusting Aspiration Sensor
Note: Before performing this adjustment make sure the HGB blank and HGB Asp. sensor blank values
are in range (step 4.3.2).

(1) Select Menu  Controller  Service  Mode. Select Control 3 in Analytical Mode and Off in Con-
tinuous, and click OK.

(2) Select Aspiration Sensor.
(3) When Aspiration Sensor appears, run e-CHECK (level 2).
(4) After analysis, enter “HGB assay value for e-CHECK Level 2 X 105” into Target box (For HGB
assay value, refer to g/dl value in the assay sheet). Then, click “>>>” “Send” and press start
switch.
(5) Confirm that the value is HGB assay value for e-CHECK Level 2 X 105 +/- 50 when analysis is
over.
(6) Repeat until sample – blank value is in the range.

(4) Press Start Switch

(5) Confirm the value is HGB assay (1) Enter HGB assay value (2) Click “>>>” (3) Click Send
value for e-CHECK Level 2 X 105 +/- 50 for e-CHECK Level 2 X 105

(7) Click Close when value becomes HGB assay value for e-CHECK Level 2 X 105 +/- 50.

4.3.4 Adjusting Sensitivity


(1) Confirm that Aspiration Sensor adjustment is completed. (Select Menu  Controller  Service 
Mode. Select Control 3 in Analytical Mode.)
(2) Run e-CHECK (level 2).
Note: For American Markets, the X-CAL product is used rather than e-CHECK level 2 when per-
forming Sensitivity adjustments.
(3) Select Sensitivity in Service screen. (e-CHECK cannot be run in the Sensitivity screen.)
(4) Enter each assay value in the Target box and click >>> and Send.

(5) Click Close.


(6) Run e-CHECK (level 2) again.

(7) When the analysis is over select Sensitivity and confirm that values are within the following range.

Target range
DIFF-X: assay value +/- 3ch
DIFF-Y: assay value +/- 2ch
FSC-X: assay value +/- 2ch
MCV: assay value +/- 1 fL
MPV: assay value +/- 1fL
HGB: assay value +/- 0.2 g/dL

Make sure that the DIFF scattergram and FSC histogram are discrete and displayed normally.
(8) Click Close when the values are within the range.
(9) Repeat step (6) and (7) until the values are within the range.

For adjusting DIFF-Y using human blood, aspirate ten normal blood samples and Calculate mean
value. Enter target value in DIFF-Y and click ‘Send’ so that the result value will be in the following tar-
get value range.
Target Value: 43.5 +/- 3.0

Pick up the closest sample to the mean value.
