1. GENERAL DESCRIPTION OF THE MACHINE

The MICROSED-SYSTEM machine consists of:

_ A mechanical support body for test tubes, under which the reading plate moves.

_ A reading plate consisting of an electronic circuit composed of 10 selected pairs of transmitters and infra-
red light receivers. The infra-red sensors check the transparency of the samples: an additional pair of infra-
red sensors is on the reading plate to register the upper end of stroke.

_ A motor block composed of:


A motor with mechanism to reduce rotation speed. An electronic circuit with some anti-blocking filters, the
temperature sensor and the sensor for noting the position of the plate, consisting of an encoder and an infra-
red optical sensor.
_ A CPU board to control timer functions, analysis, calculation, printing and memorisation.

_ A supply accepting input tensions between 220 and 240 Vac with frequencies between 50 and 60 Hz. The
supply gives the 12V AC tensions necessary to the functioning of the machine. The maximum output
currents are of 500 mA on 12V.

_ A serial port for connecting a printer.

1.1 DESCRIPTION OF MECHANICAL CHARACTERISTICS AND LIMITS

The reading of the level is carried out by an encoder assembled on a helical screw which directly activates
the reading plate. Resolution is 5 steps per mm.
The upper end of stroke is registered by an optical sensor on the reading plate. The lower is fixed and
measured by the electronic encoder. The number of steps, from the upper to the lower limit, is fixed at 265 ,
equivalent to a 50 mm stroke.

The minimum starting level accepted is 49 mm +/- 1 mm, from the bottom of the test tube.
The maximum starting level accepted is given from the maximum stroke permitted by the mechanism, equal
to 65 mm from the bottom of the test tube.

1.2 GENERAL DESCRIPTION OF THE SOFTWARE FUNCTIONS

Initially the machine is in “STAND-BY” state. The display is updated every 15 seconds. The reading plate is
stationary in the low position.
Reading of the plate is also carried out every 15 seconds. In this phase the presence or absence of samples
is registered.
Inserting one or more samples in the machine, the “STAND-BY” gives way to the “OPERATE” state. A timer
is associated with each sample, activated when presence of a sample is registered.
After the first 3 minutes, reading of the initial level is effected.
After a further 30 minutes, the final level is read. Then the final levels at 30 minutes are converted into
percentage of sedimentation and, by means of a correlation table extrapolated from a non-linear function,
further converted in mm/hour of the "Westergren" method of reference.

If the temperature correction is activated, the values are further converted to a base temperature of 18° in
accordance with Manley ' s table. At the end of the analysis the machine display and prints the result. If the
printer is not available or not connected, but the configuration provides for its use, the machine will try to print
for about 45 seconds and then abandon the operation.
It should be noted that if the results are many, the machine will be occupied in the printing phase for a fairly
long time. Analysis operations will proceed in any case
When the sample has been removed from its position, the sample data will be deleted on the display.

2. DESCRIPTION OF THE PARTS

2.1 General

The equipment consists of two basic solid connected parts: a front panel for holding the test
tubes (fig.1),and a back panel which contains the CPU board, the mechanical unit and
Infrared sensors for reading the erythrosedimentation rate and the power supply connector
and the general (fig.2).

2.2 CPU board


The microprocessor board is shown in fig.12 (display side view) .

2.3 I.R. sensors board (Infra-red)


The I.R. board is shown in fig. 15 (assembled) and fig.14 (PCB), consisting of 10 pairs
(emitter-receiver) of infra-red sensors.
This part is assembled on a mobile plate and enables recording of the sedimentation of the
erythrocyte.

2.4 Motor
The motor- group is shown in fig. 19.

2.5 Printer (optional)

Shown in fig. 2b
Printer is a serial input standard printer with internal universal 100- 240 VAC power supply.
The printer cable is a standard 9 pin female serial cable. It is supplied with the printer.

DISASSEMBLY OF THE MACHINE

3.1 Opening of the machine

Make sure the power supply is disconnected.


Take the warrantee-seal out as indicated in (fig.3):this
warrantee-seal must be taken out only by technical
personnel authorised for the assistance. After the
reparation, it has to be replaced by another seal.
If the seal is damaged by the operator the warranty
expires.
Unscrew the eight screws placed in the back panel
(fig.4).
Remove the front panel letting it slide lightly towards
you(it is better to keep the instrument laid on the table
together with the back panel). Be careful with the
internal cabling (fig.5).

3.2 Defective module replacement

At this point, having identified which one of the modules


is defective, replace it making sure that each part is
correctly assembled.
In order to take the CPU card out it is necessary to take
out before the mechanical part lifting the two steel
guides at the same time as shown in (fig.6).
The detector-module IR will be removed by the joint (fig.
7).
Also lift the endless screw from their position (fig. 8).
Now unscrew the fixing screws of the card to the back
panel as shown in (fig.10,11) and take the card
out(Fig.12).
Do not screw the fixing screws of the display for this
operation because it is not necessary.
The motor-group can be simply disassembled unscrewing the
two screws as shown in (fig.9)
The motor-group is supplied already assembled on its
support.

3.3 Setting

The modules are supplied set and tested, so no setting is


necessary after their replacement.
Should it be necessary to check correct functioning, a
CPU board test card appears in the appendix of this
manual.

3.4 I.R. board replacement

The IR card(fig.14) is usually supplied already assembled
and tested as shown in (fig.15 ). Please use the maximum
attention in the handling of this part because each pair
of detectors is aligned in laboratory for a good
functioning. If the detectors are lightly bent by the
card, you certainly could have reading problems.
Do not use liquids or solvents to clean this part. Use
only a clean brusher to take the dust off (if it is
necessary)

During replacement of this part pay attention because the


I.R. sensor pairs are very sensitive to knocks and dirt.

3.5 Instrument reassembling

In order to assemble the instrument again repeat the


reversal operation of the assembly, starting from the
card, then from the endless screw , then with the IR
module together with the ways (be sure that ways and
endless screws to be well introduced in their own
position). The IR module must be inserted in the apposite
slit-bush in Teflon (fig.7)
Finally assemble again the motor-group (fig.19), because
it must be well fixed.

Connect the flat cable of the IR module fastening with


the related metal fixings.
Insert the (front and back) panels slowly, taking special
care with the plastic runners with which they are
equipped. The back panel cabling must be put inside the
cavities under the base plate.
Before closing the equipment, check that the connectors
have been inserted in the right positions.
Press the central module before re-tightening the eight
screws in the sides.
To avoid breaking the plastic runners, don't over-tighten
the screws.

5. ERROR INFORMATION AND WARNINGS

5.1 Warnings with information

Messages may appear on the display :

ERL: Indicates that the sample is not of a level that would permit the instrument to
read it. If the sample has a level higher than permitted (around 60mm) it may be corrected
by aspirating off the surplus with a syringe. If the level is too low (> 10mm) the sample
should be taken again because outside these limits the results of the analysis may be
altered.

ERS: Indicates that the test tube has been removed from the position in which it had
been placed. The test tubes are checked in their positions at constant 15 second intervals.
Do not touch the test tubes during the whole period of analysis because, for the reason
described, the result will be lost.

PRINTER NOT READY...: Indicates that the printer is not connected. After 15 seconds
the MICROSED-SYSTEM automatically disable the printer output port.

5.2 SYSTEM ERROR WARNINGS

“ ERROR: Mechanical problems..”

This warning will be given if the instrument should have problems with the mechanical
movement of the reading plate.
After this indication the instrument will definitively stop its operation and technical
assistance must be requested.

6. MAINTENANCE

6.1 Maintenance

Its simplicity and component parts mean that the MICROSED-SYSTEM does not require
special maintenance. The most sensitive part is the infrared sensors inside the instrument.
Care and the attention is required above all with regard to the cleanliness of the upper part
(test tube positioning plate) which must be covered when not in use with the dust cover
supplied as an accessory..
Do not clean the upper plate with liquids or damp cloths.
The entry of liquids or solid material into the holes can cause considerable damage to the
instrument.

6.2 Cleaning Instructions

Any dust can be removed using an ordinary vacuum cleaner.


A detail with regard to test tube insertion: it must remain well closed and the cap should
absolutely not be removed.
The label must be correctly positioned and adhere well to the test tube surface. Label
fragments could fall into the test tube and obstruct a correct reading function during
analysis.

A. TESTING CPU BOARD MICROSED-SYSTEM

PHASE N° DESCRIPTION OF THE OPERATION


( ) 1) Connect CPU to the following peripherals (after ascertaining their
correct functioning):
A) IR board
B) Motor
C) PRINTER
D) Power supply

( ) 2) Switch on the power and observe:

A) Display light and visualisation of:


the name of the machine, the software version (the “T.ERR” signal points out a defect or
disconnection of the temperature sensor).
The temperature correction (“TEMP REF TO 18C”) is activated by setting the first SW1
switch at “ON” (Dip Switch 4 pole on CPU board).

B) If the printer is connected and switched on it will print:


- Name of the machine
- Software version
- Message of temp. correction (if activated)
- Temperature level
- Heading of the analysis results

( ) 3) Check that the motor starts automatically bringing IR plate to the top.
If this does not occur probably the IR cabling is not well connected or there is an anomaly
in the end of stroke sensor . That causes collision of the plate with the transmission belt
and subsequently the signal “ERROR: Mechanical problems...”.
If on switching on the motor it doesn’t start, an anomaly is signalled: “ERROR: Mechanical
problems...”: check the connections to the motor board.

( ) 4) Reading of the test tubes is carried out during the ascent phase of the IR board.

( ) 5) When the IR board reaches a distance of approximately 1.5 mm from the upper
plate, the sensor optocoupler notes the end of stroke and inverts the motor. If this is not
noted the IR board comes into collision with the upper mechanical part making the motor
stop and consequently giving the signal “ERROR: Mechanical problems...”.
This anomaly may be caused by the inadequate course to the sensor.
If the defect turns up again, the problem could be due to a defect in the sensor (in this
case replace the whole CPU board).

( ) 6) After the IR plate stops in the low position, check that, in the absence of test tubes,
the symbol “---” appears on the display in correspondence to the position of each tube.
The presence of the symbol “run” in any position precludes setting anomalies or defects in
the sensor pairs.

( ) 7) Insert the TEST tubes; within about 15 secs. the symbol “run” should appear in
correspondence to the positions in which they were introduced. After 3 minutes the IR
plate starts moving to the top, checking the level of the samples inserted. A permanent
“run” symbol indicates that the level has been accepted and the machine setting is right.

( ) 8) Wait 30 minutes for the analysis result on the display, and in printout.

( ) 9) Remove the test tubes and check if, in about 15 secs., the result disappears and is
replaced by the “---” symbol.

MICROSED SELF-TEST FUNCTION

This function permits to check all the necessary parameters for ESR calculation..

They are:

- Internal temperature
- Level check calibration
- Three levels measurement at environmental temperature
- Control of the conversion at 18° C.
- Reading reproducibility.

HOW TO ENTER INTO “SELF TEST”FUNCTION

Switch off MICROSED and insert the special tubes calibrators at three levels (60, 50,
30), respectively in the positions of the channels 5, 6 and 7.
Be sure that the other channels are free and switch on the instrument.
After the normal initialisation, MICROSED will enter automatically in SELF TEST
procedure.

On the display the following information will appear:

On the left, at the bottom of the display the visualisation of the internal temperature will
appear.

The values in the brackets of the high row and marked with a. t. represent the values in
mm of the theoretical levels of sedimentation whose values must be included between
1+/-0, 24 +/-4, 93+/-6 at environmental temperature and in the row below there are the
same results corrected at the temperature of 18°.
Please refer to the table (table 2) to check the correct compensation of the data.

Example measurement at instrument temperature of 22° C: column no.5 of the table:

If all values fall out of these ranges, the instrument needs to be checked by the service
department.

CONTROL OF THE LEVEL CALIBRATION

On the right of the display it is possible to visualise the motor steps connected to the
measurement of the theoretical level of the tube 60 mm.
The number of the STEPS must be around 216 +/- 5 which corresponds to a tolerance of
calibration of +/- 1 mm in respect to the theoretical one.

If the data belong to the pre-defined tolerances, sample levels within the limits declared
from 48 mm to 64 mm +/- 1 mm. will be accepted.

(*)NOTE:sw5,sw6,sw7,sw8 will be useful for next applications.

After selection you can see the configuration mode on the LCD display during the
initialisation of the instrument.

Display of the results:

Results appear on the display in this way:

• Configuration 1/hour Westergren: results appear on the lower line after 30’.
• Configuration 30’/1 hour Westergren: 30’results appear on the upper line and 1 hour
result on the lower line simultaneously after 30’.
• Configuration 1 hour/ 2 hours: 1 hour results appear on the upper line after 30’ and 2
hours results on the lower line after 60’.
• Configuration 30’, 1 hour and 2 hours: after 30’ on the upper line it appear the 30’ result
and 1 hour result in the lower line.
After 60’ appear in the upper line the 1 hour Westergren and in the second line the 2
hours result.
Pay attention do not make mistake for the result 30’, 1h and the 1 h e 2 h. Result of 1
hour and 2 hours are prefixed with sign:( = )

2. Pre-indication of results :
The instrument, makes a level reading of the tube after 9 minutes (3 initial + 9 of analysis). The
sedimentation ratio is calculated and a special symbol is displayed on the screen. This gives an
anticipated result indication of the samples under analysis. This is the table of symbols
displayed at different ratio. (the symbol is displayed instead of " run", after 9 minutes and remains
until the final analysis result appears).

3. 30/60' time mode:


when the instrument works on the second hour of analysis (after 30'
) and the level of the blood
falls under the maximum allowed, a special result is printed out. The result of the 2h is printed as
"?>" symbols and a value that is the maximum allowed by the instrument at the current
temperature. (eg. 2h = ?>140 ). Please note that it is impossible to detect if the user takes out
the sample from the instrument and for this reason the "?" is used to mark an abnormal situation.

B.2 Printer configuration

The printer is already equipped configured.
