Determination  of  Orthophosphate  

INTRODUCTION

Phosphates that respond to colorimetric tests on sample without preliminary hydrolysis

or oxidative digestion of the sample are termed as 'reactive phosphates". Reactive

phosphate consists largely of ortho-phosphate, with a small fraction of condensed

phosphate such as di-phosphate and tri-phosphate. In this method, the water sample is

allowed to react with a composite reagent containing molybdate acid, ascorbic acid anti

trivalent antimony. The resulting complex is reduced to a blue solution, which is

measured at 880 nm.

Reagents

KH2PO4 standard solution: (0 – 1.0 mg PO4/L).

Ammonium molybdate solution: Dissolve 4.0 g ammonium paramolybdate,

(NH4)6Mo7O2.4H2O in 100 ml distilled water.

Acid Sulfuric solution (5 N): Add 140 ml concentrated sulfuric acid (sp. gr. 1.82) in 1 L

volumetric flask contained 800 ml of distilled water. Top up the solution to 1 L.

Ascorbic acid solution (0.01 M): Dissolve 1.76 g ascorbic acid, C6H8O6 in 100 ml

distilled water. Freshly prepare prior to use.

Determination  of  Orthophosphate  

Potassium antimonyl-tartrate solution: Dissolve 1.3715 g potassium antimonyl-

tartrate, K(SbO)C4H6O6.½ H2O in 500 ml distilled water.

Mix reagent solution. Mix together 75 ml ammonium molybdate solution, 250 ml sulfuric

acid solution, 150 ml ascorbic acid and 25 ml potassium antimonyl-tartrate solutions.

Prepare this reagent when needed and discard any excess. Do not store.

Procedure

1. Prepare different concentrations of standard solution, 0.05, 0.10, 0.03, 0.05, 1.00

mg/L PO43-.

2. Filter all water samples prior to analysis.

3. Pipette 5.0 ml of each standard solution, water sample and control (distilled water)

into test tubes. Add 0.5 ml mix reagent to each test tube and stir homogenously.

4. After 1 hour, record Absorbance using spectrophotometer at wavelength 880 nm.

Determination of orthophosphate concentration.

Standard curve

Prepare 100 ml of each standard solution using the standard solutions (Blank, 0.05,

0.10, 0.03, 0.05, 1.00 mg/L PO43-)1. Use deionized water or artificial seawater (depend

on sample) for dilution.

F factor

Concentration (mg/ l) = Sample absorbance x F factor

Factor, F = 1.0 mg/l / (Es-Eb)

Where,

Es = absorbance of standard solution

Eb = blank solution absorbance

 Perform  a  series  of  dilution  instead  of  one,  the  dilution  factor  should  not  exceed  100  x
