1.5 Intended Use

1.5.1 Intended Use


DH-520 Series Auto Hematology Analyzer is an in vitro diagnostic device that is
to be used together with supporting reagents, calibrators and QC materials. It is used
in medical laboratories to analyze the relevant items of CBC and DIFF of the human
blood. Anticoagulant adopts EDTA-2K, EDTA-3K and EDTA-2Na. Blood anticoagulant
ratio is guided by CLSI.

1.6 Analysis Principles

1.6.1 Sample Quantitative Method


Syringe is used to conduct quantification.

1.6.2 DIFF Measurement

When certain amount of blood cells are drawn and reacted with specific
reagent, certain amount of blood samples will be injected through the nozzle into
the conical flow chamber which is filled with diluent. With the surrounding of sheath
fluid formed by diluent, the cells will cross the central flow chamber in a single row.
When the blood cells which are floated in the sheath fluid pass through the laser
detection region, they will be exposed to a laser beam and produce scattered lights,
whose property is associated with the cell size, cell membrane and refractive index
of the internal cell structure. The forward scattering light (FSC) reflects the size of
the cells, the side scattering light (SSC) reflects the cells’ fine internal structure and
particulate matters, and the side fluorescent light (SFL) reflects nucleic acid contents
in the cell. The photodiode receives these scattered light signals and converts them
to electric pulses. According to the collected electrical pulse data, the
two-dimensional distribution diagram of blood cell size and internal information can
be obtained, which is called a scattergram. In Figure 1-3, the abscissa reflects the
information of the complexity of cells’ inside and the ordinate reflects the nucleic
acid content of the cell.

From DIFF channel, percentage of LYMPH, MONO, EOS, NEUT and IG of the WBC
total count can be obtained.

1.6.3 WBC Counting


When certain amount of blood cells react with a specific reagent, RBC and WBC
excluding BASO are dissolved. With the reaction liquid driven by constant vacuum,
the cells will one by one pass through a small hole applied with constant current
source. The number and height of pulses reflect the number and volume size of the
cells. Principles are shown in Figure 1-4.

From the WBC histogram, WBC count, BASO count and the percentage of BASO
of total amount of WBC can be obtained. Histogram is shown as Figure 1-5.

1.6.4 WBC Parameters


By analyzing the WBC histogram, the analyzer can compute White Blood Cell
(WBC) count and basophile count (BASO#), and further get the basophile percent
(BASO%). It can compute the neutrophile percent (NEUT%), the lymphocyte percent
(LYMPH%), the monocyte percent (MONO%), the eosinophile percent (EO%) and the
immature granulocytes percent (IG%) by analyzing the scattergrams and its area of
neutrophile, lymphocyte, monocyte, eosinophile and immature granulocytes.
Computing with the WBC count obtained from the WBC histogram, you can get the
neutrophile count (NEUT #), the lymphocyte count (LYMPH#), the monocyte count
(MONO#), the eosinophile count (EO#), the basophile count (BASO#) and the
immature granulocytes count (IG #). The unit of the cell count is 109/L.

1.6.5 RBC/PLT Measurement


The analyzer uses Coulter Principle to count the RBC/PLT. A certain amount of
blood cells are injected into RBC test unit after secondary dilution. There is a small
hole called detect hole in the RBC test unit. The small hole is connected with positive
and negative electrodes on the side and is charged with constant current source.
Since the cell is a poor conductor, the resistance between the electrodes will change
when the diluted cells pass through the hole, thereby forming a pulse signal with the
similar size of the cell volume at both sides of the electrodes. When cells pass
through the hole continuously, a series of pulses will be formed. Pulse number is
consistent with the cell number, and the pulse height is related to the size of the cell.
Pulse signal amplified by signal acquisition circuit is compared with the
threshold of normal RBC/PLT volume range to calculate the count of cells which fall
in the RBC/PLT range, so that we can classify the RBC and PLT and get their count.
Abscissa represents the volume and the ordinate represents the relative count of
cells so that RBC/PLT volume distribution histogram shown in Figure 1-6 can be
obtained.

variation expressed in percentage and obtained from RBC histogram.

peak height on the histogram. Unit is fL. It is shown in Figure 1-7.

1.6.8 HGB Measurement
In the HGB colorimetric pool, after the diluted sample is added to the lysing
reagent, red blood cells will dissolve and release hemoglobin, which unites with
lysing reagent and becomes hemoglobin compounds. At one end of the colorimetric
pool, illuminate the hemoglobin compounds with LED light emitted from a
monochromatic light tube with 520nm wavelength; the other end receives
transmission lights through phototube, and converts them to voltage signal after the
optical intensity signal is amplified. By comparing it with the voltage obtained from
the transmission light before the samples were added to the colorimetric pool (only
diluents before), you can get the sample’s hemoglobin concentration.

Take a vial of fresh blood or QC material. Test it 10 times or above continuously. Its
coefficient of variation meets the following requirements within 95% of the
confidence interval.

1.7.5 Counting Correlation


After calibration, the analyzer tests no less than 100 samples with the standard
device to conduct correlation comparison. Regression equation: Y=aX+b; calculate

1.7.6 Differential Correlation


After calibration, the analyzer tests no less than 100 samples with the standard

4.1 Motor Position Debugging


Regular installation does not require position adjustment. Motor position is
saved in analyzer’s driver board and computer’s database.
When motor is running, driver board value is executed and database value is
used as backup. Click “Save” to write the database value to the driver board.
Position needs to be confirmed if sample collection unit, DIFF reaction chamber
or counting chamber unit is replaced.
The motor of DH-520 series analyzer is divided to three modules:
 Sample collection unit motor module
 Mixture unit motor module
 Automatic sampling unit motor module (no adjustment needed on site)

4.1.1 Adjustment Method


Log in with account name “super”.

Modify “Backup Value”. If the value increases, motor is farther away from OC; if
decreases, motor is closer to OC.
Use 5 steps as the rough adjustment unit. After modifying database value, click

that the modification is effective. User can conduct a blank check to see whether the
position is correct.
When approaching the correct position, adjust step by step until the requirement is
met.

Database backup only applies to the software with the same version. If AUMS
version upgrades, the original backup database cannot be imported. Forced import
will crash the software, making it unusable.
This is used after installation and debugging to conduct a complete backup of
the database. If the client computer crashes or AUMS software reinstalls, backup
database can be imported directly. All the parameters do not need adjustment.

5.3 Main Control Board Replacement
Main control board replacement needs to make sure that ACMS program and
time sequence version matches.
After replacing main control board, user needs to readjust relevant gain in
“Device Parameter” since the previously backed up parameters do not match.

5.4 Driver Board Replacement


After replacing driver board, user needs to make sure whether it matches the
drivers, or the pressure might be inaccurate.
After replacing driver board, user needs to confirm motor position. Basing on
the database value, user can click “Save”-“Refresh” to write the database backup
value to the driver board.
If the driver board value and database value are inconsistent, report “Motor
moving position points inconsistent” error during start up. See “4.1 Motor Position
Debugging”.

5.5 HGB Voltage Setting


See field description in “4.3.1 Main Control Board Setting”. HGB blank voltage
requires 4.0±0.1V.
When HGB blank background voltage is lower than 3.6V or exceeds 4.4V, device
reports “HGB background voltage abnormal”.

5.7 Package Shutdown


If the device is not used for over a week, client should be informed to start up
every two days. If client cannot do that, package shutdown should be conducted.
If the device is not used for a long time, there might be risks of pressure valve
tubes adhesion therefore cannot flick, tube crystallization, optical flow chamber
blockage and so on.
Package shutdown requires purified water. User can choose purified water
(mineral water and natural water not allowed) sold on the market. Consumption is
less than 500ml.
Fluorescent dye tube also needs emptying.
After package shutdown, take out pressure valve V104, which should be put
back in again when the device resumes using.

Chapter 7 Installation

7.1 Software Installation


32 bit system (64 bit not available yet)
Note:
(1) Please turn off firewall and antivirus software.
(2) It is normal that third-part security software prompts whether to continue with
the installation. Please continue with the installation.
(3) WIN 7 system user please log in to the computer with the user name of
Administration to install AUMS.

7.1.1 AUMS Installation


Install AUMS in WIN 7, XP system:
(1) Double click AUMS installation packet. If “Next” interface appears, execute from
step (3); if not, execute from step (2).
(2) Run "SETUP.exe" as manager.
(3) Click “Next” to install until installation completes.
(4) Run “InstallDB.bat” as manager under “AUMSSetup” file.
(5) Select “A” (regardless of case) and press “Enter” until “Installation completes”
appears.
7.1.1.1 Run as Manager
(1) Right-select the file and select “Get Manager’s All Authorities” in the pop-out
menu.
(2) Right-select the file and select “Attribute-Compatibility”. Check “Run this
program as manager”.
7.1.1.2 Abnormal Condition
During database installation, if [“OSQL” is not an internal or external command, nor
an executable program or batch file] occurs.
(1) Please check whether database service pack is installed (click “Start” menu-“All
Programs”-“Microsoft SQL Server 2005”). If Microsoft SQL Server 2005
installation file is not found, please install “SQLEXPR32_CHS.EXE” manually.
(2) If (1) is successfully installed, check whether right click executing “InstallDB.bat”
as manager to process batch files.
(3) If (1) and (2) are successful but the above problem still occurs, please retry after
changing a computer or reinstalling the system.

7.1.2 Network Setting


(1) Setting- Network Connection-Local Computer Network
(2) Select the network card that needs connecting to the device. (Please select the
network card that connects to the cable. When there are dual NIC, the device
connects to the computer onboard NIC.)
(3) Enter the connecting device’s IP (192.168.7.113), subnet mask (255.255.255.0),

default gateway (192.168.7.1), DNS (192.168.7.1).
(4) Click “Save” and it prompts “IP setting successful!”
(5) Check whether AUMS’s current status is “Ready”. If so, the device is successfully
connected.
(6) Then select the NIC that needs connecting to extranet network and enter the IP
address of the Internet.
(7) If the IP of (6) is LIS system IP, please note: this IP address is not the same as the
IP address of LIS server.
(8) Click “Save” and it prompts “IP setting successful!”
(9) If device status is “Ready”, it means the device is successfully connected.
