Blood Group Serology Practical

Title: ABO and Rhesus Cell grouping


Aim: To determine the specific antigen present in a given blood sample
Method: Tile
Materials/reagents: blood samples, white tiles, applicator sticks, Anti sera A, B,
AB and D
Procedure:
 Wash all tests and control samples at least 3 times and prepare 20% cell
suspension
 place one drop each of anti A, Anti B, Anti AB and Anti D on to a cleaned
white tile
 add one drop of already prepared 20% test and control samples on to a
respective column
 mix thoroughly with a separate applicator stick and rock gently and allow to
stand at room temp for 5 minute and record the result
Protocol:
Sample Anti A Anti B Anti A+B Anti D Result

K + - + + ‘A’ Rhesus D positive

L - + + - ‘B’ Rhesus D negative

M + + + + ‘AB’ Rhesus D positive

N - - - - ‘O’ Rhesus D negative


 Sample k is from blood group A individual
 Sample L is from blood group B individual
 Sample M is from blood group AB individual
 Sample N is from blood group O individual

Title: ABO serum grouping


Aim: To determine the antibody presence in a given serum
Method: Tile method
Materials/Reagent: blood samples, white tiles, applicator sticks, STD ABO cells,
physiological saline.
Procedure:
 Prepare 5% standard ABO cell for test and control samples
 Place one drop of each 5% STD ABO cells on to a clean white tile
 Add equal volume of each test and control serum on to respective column
 Mix thoroughly with applicator stick, rock gently and read the result within
5 minutes
Protocol:
Sample STD A Cell STD B Cell STD O cell Result

K - + - A blood group

L + - - B blood group

M - - - AB blood group
N + + - O blood group
Comment:
 Serum sample K was from blood group A individual
 Serum sample L was from blood group B individual
 Serum sample M was from blood group AB individual
 Serum sample N was from blood group O individual

Aim: To determine the specific antigen present in a given blood sample
Method: Tube method
The tube technique is employed for all blood group tests. It is recommended for
confirmation of slide/tile test results when it is doubtful and whenever there is
enough time.
Materials/reagents: blood samples, test tube, racks, applicator sticks, water bath
Anti sera A, B, AB and D
Procedure:
 Prepare 5% cell suspension for each test and control sample
 Arrange the required number of precipitin tubes in to a rack
 Place one volume of unknown cells/serum of each test and control sample
 Add equal volume of known serum (Antisera)/cells (5% suspension) to the
appropriate tube
 Mix the tubes gently and allow to stands at room temperature for 1½ hours
 Incubate tubes containing ant-D at 37OC for 1½ hours
 Add one volume of bovine albumin to tube containing ant-D after 1½ hours
and re-incubate for further 30 minutes
 Read result macroscopically by gently tapping the tube and microscopically
if no observable reaction was observe macroscopically
