This study examined hepatic cytochrome P450 (CYP450) response to dietary sesamin in combination with different n-6/n-3 fatty acid ratios in fish diet. Over a period of 4 months, fish were fed seven different experimental diets an n-6/n-3 FA ratio of either 0.5 or 1.0 in combination with two sesamin levels: low sesamin = 1.16 g/kg feed and high sesamin = 5.8 g/kg feed. Control diets did not contain sesamin. 2. The CYP450-associated activities of ethoxyresorufin O-deethylase (EROD), 7-benzyloxy-4-trifluoromethylcoumarin O-debenzylation (BFCOD), pentoxyresorufin O-depentylase (PROD), coumarin hydroxylase (COH), methoxyresorufin O-deethylase (MROD) and p-nitrophenol hydroxylase (PNPH) were significantly induced by dietary sesamin in a dose-related manner. 3. Expressions of the genes CYP1A1, CYP1A3, CYP3A, AhR1α, AhR2β, AhR2δ and PXR involved in the regulation of CYP450 activities, was not the primary source of this induction.

ABSTRACT microRNAs (miRNAs) are small non-coding RNAs that regulate numerous fundamental biologic... more ABSTRACT microRNAs (miRNAs) are small non-coding RNAs that regulate numerous fundamental biological functions. So far miRNAs have only been identified in few commercially important fish species. Similarly to messenger RNA (mRNA) expression analysis, miRNA qPCR analysis requires thoroughly investigated endogenous controls. In this paper, we have in a pilot study investigated miRNAs in Atlantic salmon (liver, heart, brain, kidney, spleen, intestine, gill, white, and red muscle) by qPCR analysis. The expression of seven putative endogenous control genes (ssa-let-7a, ssa-miR-16a, ssa-miR-16b, ssa-miR-194a, ssa-miR-22a, ssa-miR22b, and ssa-miR-27c) was examined with regard to their tissue distribution and use as endogenous controls microRNA expression studies. The candidates for endogenous controls were selected as they had been reported by others to be evenly expressed among tissues. The most suitable endogenous control was miR-27c in this study. Further the expression of ssa-miR 26b, ssa-miR-92a-1, ssa-miR-122, ssa-miR-722, ssa-miR-21-1, and ssa-miR-143-1 were investigated and tissue specific expression of all miRNAs except ssa-miR194a were found. The expression of sss-miR-122 and ssa-miR-722 were specific for liver, ssa-miR-26b mostly expressed in brain and ssa-miR143-1 mostly expressed in gill and kidney. This study has provided information of endogenous control genes and has also identified tissue specific expression of miRNAs in Atlantic salmon. Further investigation of endogenous control genes target genes in response to treatment would increase the knowledge of the role of miRNAs in Atlantic salmon.

ABSTRACT Sesamin (S) is a known lipid modulator and has been shown to increase the conversion of ... more ABSTRACT Sesamin (S) is a known lipid modulator and has been shown to increase the conversion of α-linolenic acid (ALA) to docosahexaenoic acid in rainbow trout (Oncorhynchus mykiss) fed vegetable oil mixtures including linseed oil. In this study, we evaluated the effects of S supplementation in linseed oil-based diets, content of α- and γ-tocopherols, fatty acid (FA) composition, as well as the gene expression of lipid-related genes. Fish with an average weight of 36.5 g were fed different combinations of commercial linseed oil (LO), purified linseed oil triacylglycerols (TAG) with polar fraction removed and a mixed linseed-sunflower oil (6:4 v/v) (MO). S was added at 0.58 g 100−1g feed and fed to the fish for a period of 58 days. Expression of PPARα was downregulated in white muscle of fish fed S containing diets (P < 0.05). The expression of PPARβ1A was not affected by S supplementation except where TAG oil was used. The expression of PPARβ1A declined significantly in TAG + S fed group (P < 0.05), which indicates that some minor compounds in linseed oil might suppress the effect of S on the expression of PPARβ1A. The expression of PPARγ(long) declined in LO + S and MO + S fed group (P < 0.05). The β-oxidation-related genes CPT1 and ACO were upregulated by vegetable oils compared to fish oil. S decreased percentage of ALA in white muscle of fish fed LO + S (P < 0.05). The increased desaturation index and the decreased ALA levels suggest that S may increase the biosynthesis of highly unsaturated FA in rainbow trout.

With declining commercial fisheries, aquaculture fish constitute a growing proportion of human consumption. Sustainable development of aquaculture requires that the fish feed used is not solely based on fish meal and oil (FO), but also contains increasing levels of vegetable oil (VO). The replacement of FO with VO influences FA composition in fish tissues by decreasing n-3 long-chain polyunsaturated fatty acids (LCPUFAs) and the nutritional value for humans. Accordingly, the last decade of salmonid research has focused on increasing the amount of n-3 LCPUFAs in fish fed VO diets e.g. addition of bioactive compounds. This thesis examined the potential effects of bioactive compounds on lipid metabolism in salmonids.
