This study aimed to investigate the impact of Borreria hispida extract on gene expression in NRK-... more This study aimed to investigate the impact of Borreria hispida extract on gene expression in NRK-52E cells under conditions of glucotoxicity. Gene expression analysis was conducted using RT-PCR following the exposure of cultured NRK-52E cells to glucotoxic conditions and varying concentrations of Borreria hispida extract. The results demonstrated a dose-dependent increase in SIRT1 gene expression and a concomitant decrease in HIF-1λ, ET-1, and VEGFR-2 gene expressions upon treatment with Borreria hispida extract. Additionally, molecular docking studies suggested the potential inhibition of Rho-kinase as a mechanistic explanation for these effects. Borreria hispida extract may confer renoprotective benefits against glucotoxicity-induced cellular damage. The potential therapeutic utility of Borreria hispida extracts in managing renal complications associated with conditions such as diabetes. Furthermore, our docking studies shed light on the potential molecular mechanisms underlying the observed effects, suggesting interactions between phytochemicals present in Borreria hispida extract and the JNK-1 protein. These interactions may contribute to the augmentation of SIRT1 activity, further bolstering the extract's therapeutic utility in DKD.

The pro tein pro files were analysed by NA -TIVE and SDS -PAGE, re sults in di cated both high and low mol e c u lar weight pro teins rang ing from 135 to 25 kDa. The crude ex tract was tested for its an ti cancer ac tiv ity against hu man colon can cer cell line (HT 29) by MTT as say, the In hi bi tion con cen tra tion (IC 50 ) was ob tained on 600 μg/ ml at 24 h. Flu o res cent mi croscopy was used to study the cell mor phol ogy, nu clear con den sa tion, early and late apop totic stages by def i nite staining meth ods. DNA frag men ta tion and flow cy to met ric re sults re vealed that the skin ex tract in duces chro matin con den sa tion, apop totic cell death and also in duce cell cy cle ar rest at G 0 /G 1 phase on colon can cer cell line.

The skin mucus share protective activities against environment that are rich in infectious agents. The current study aims to analyses the anticancer, antibacterial and haemolytic properties of crude epidermal mucus from marine catfish Tachysurus dussumieri. Cell viability and Cytotoxcity was assessed by MTT method on HT 29 (human colon carcinoma) and Vero (kidney epithelial cells of African Green Monkey) cell line. In Vero up to 1000 µg/ml of crude mucus shows cell viability of 80% and hence mucus concentration for treatment of HT 29 cells was fixed between100 to 1000 µg/ml. The proliferation inhibition on HT 29 at a concentration of 400µg/ml mucus from T. dussumieri at 24 h. Cellular and nuclear morphological variations was observed in different concentration of mucus, followed by DNA fragmentation to confirm the induction of apoptosis. Moreover, mucus sample was tested against Gram positive and negative bacteria, The maximum zone of inhibition was observed in S. aureus (15.61 ± 0.7 mm), followed by B. subtilis (8.32 ± 0.32 mm) and E. coli (8.14 ± 0.08 mm). Interestingly mucus from T. dussumieri showed haemolytic activity on different blood samples of goat, rat, sheep, buffalo, rabbit , cow, hen and human. Mucus showed maximum haemolytic activity against goat, rat, sheep and buffalo when compared to other RBCs. All these results showed that the epidermal mucus has more efficacy on therapeutic application in near future..

Biologically stabilized gold nanoparticles were synthesized from the flower aqueous extract of T.... more Biologically stabilized gold nanoparticles were synthesized from the flower aqueous extract of T. divaricata. The synthesized nanoparticles were characterized by UV-Vis spectrophotometer, Zeta sizer, FTIR and TEM analysis. T. divaricata reduced gold nanoparticles having particle size and potential of 106.532 nm and -10.2 mV, respectively, with a characteristic peak of 550 nm in UV-visible spectrophotometer. FTIR graph after comparison between the crude flower extract and gold nanoparticles showed three major shifts in the functional groups. The morphology and size of the gold nanoparticles were examined by HRTEM analysis, which showed that most of the nanoparticles were nearly spherical with size of 100 nm. The gold nanoparticles synthesized demonstrated potent anticancer activity against MCF-7 cell line. The findings conclude that the antioxidant molecule present in T. divaricata may be responsible for both reduction and capping of gold nanoparticles which possess potential applications in medicine and pharmaceutical fields.
