Liquid-liquid extraction using reversible chemical complexation for reactive extraction was chosen to be the separation method. Over 50% yield of lactic acid was obtained from fermented broth in a single extraction step, when using the tertiary amine as the extractant, 1-dekanol as the diluent and trimethylamine (TMA) as the stripping solution. The effect of complex media on the extraction behaviour has hardly been examined previously.
Recently we showed the applicability and sensitivity of the RNA-based sandwich hybridisation assa... more Recently we showed the applicability and sensitivity of the RNA-based sandwich hybridisation assay (SHA) for detection of Gram-negative cells in environmental samples . Sensitive genus-specific detection of Legionella by a 16S rRNA based sandwich hybridization assay. J. Microbiol. Met. 62,[167][168][169][170][171][172][173][174][175][176][177][178][179]. In this study the aim was to test and optimise this method for the detection of Gram-positive cells from brewery yeast slurries that contain up to 10 9 yeast cells/ml. Eleven new oligonucleotide probes were designed for group-specific detection of different beer-spoiling lactic acid bacteria of the genera Lactobacillus and Pediococcus. Functionality of the designed probes was shown by testing individual and paired probes using in vitro transcribed 16S rRNA and crude cell extracts as samples. Various simple and fast cell disruption methods were evaluated for the efficient disruption of lactobacilli and pediococci. The applicability of the designed oligonucleotide probes and the SHA for detection of brewery contaminants was demonstrated using both artificial and actual yeast slurry samples from brewery fermentation tanks with either fluorimetric readout or an electric biochip analyser.

Effective disruption of Escherichia coli cells is achieved by the intracellularly accumulated rec... more Effective disruption of Escherichia coli cells is achieved by the intracellularly accumulated recombinant murein hydrolase (Lactobacillus bacteriophage LL-H muramidase) after the addition of 5 mM thymol. Thymol destroys the integrity and electric potential of the cytoplasmic membrane, and as a consequence the muramidase can access and hydrolyze the cell wall murein leading to cell lysis. Lysis occurred within 5 min after the addition of thymol and seemed to be efficient at high culture densities. This lysis method does not require cell harvesting or addition of other cell wall weakening substances or exogenous enzymes. As a cell disruption method, thymol-triggered lysis is as efficient as sonication in the presence of 1% Triton. Furthermore, thymol did not interfere with the purification steps of Mur by expanded bed adsorption chromatography (EBA), suggesting that the lysis method presented here is well suited for large-scale production and purification of intracellular proteins of E. coli.

The fresh juice had a total soluble solids content of about 16.58Brix. The effect of centrifugation followed by depectinization with two commercially available pectinase enzyme preparations (Panzym Super E and Trenolin Rot DF) on the permeate flux has been evaluated. For the description of the concentration process, the resistance-in-series model has been used. It was concluded that the applied RO membrane is suitable for concentration of blackcurrant juice up to 28.68Brix, which was achieved with the juice that had been treated with Panzym Super E. The total resistances were calculated and compared, taking into account membrane fouling and polarization layer resistance.

Maximum yields of anthocyanins were achieved at pH2 with an extraction time of 10 min with a microwave power of 700 W. The anthocyanin yields in MAE were compared with those obtained by conventional solvent extraction (CE) using citric or hydrochloric acids, and citric acid and water with a sulfur concentration of 50 ppm. A significant reduction of extraction time was achieved using MAE; maximum anthocyanin yield was obtained after 10 min using MAE, while using CE only after 300 min. Furthermore, the amount of solvent used halved; the mass ratio of marc to solvent was 1:40 in CE and 1:20 in MAE.
