The effect of parity (multiparous vs primiparous) and body condition score (BCS; <3.0 or ≥3.0,... more The effect of parity (multiparous vs primiparous) and body condition score (BCS; <3.0 or ≥3.0, lean vs fat) at parturition on metabolic and endocrine profiles from 1 month before to 2 months after parturition were studied in 42 Holstein cows grazing on improved pastures. BCS and milk production were determined every 2 weeks. Non-esterified fatty acids (NEFA), β-hydroxy-butyrate (BHB), insulin, IGF-I, leptin, thyroxine (T4) and 3,3′,5-tri-iodothyroinine (T3) were determined in plasma every 10 days. Progesterone was determined three times per week after parturition. Primiparous cows had a lower BCS during the early postpartum period and produced less milk than multiparous animals. Primiparous cows had higher NEFA concentrations and they presented more samples with BHB concentrations of >1 mmol/l than multiparous cows. Multiparous cows had higher T3, T4 and IGF-I concentrations, while fat cows had higher leptin and IGF-I concentrations. All hormone concentrations were diminished ...

The effect of cracked corn grain supplementation (3.5 kg⁄day) during 3 weeks before the expected ... more The effect of cracked corn grain supplementation (3.5 kg⁄day) during 3 weeks before the expected calving date on milk production and composition, body condition score (BCS), metabolic and hormonal profiles and length of postpartum anoestrus was evaluated in multiparous Holstein dairy cows under grazing conditions (Energy supplemented group, n = 10; Control group, n = 10). Body condition score was weekly recorded during the peripartum period, from days )21 to +35 (parturition = day 0). Non-esterified fatty acids, b-hydroxybutyrate, cholesterol, urea, insulin, insulin-like growth factor I (IGF-I), leptin, thyroxine (T 4 ) and 3,3¢' 5-triiodothyroinine (T 3 ) were weekly determined in plasma from days )21 to +35. The reinitiation of ovarian cyclicity was twice weekly determined by ovarian ultrasonography and confirmed by plasma progesterone concentrations. Cows fed energy concentrate prepartum had higher BCS during the prepartum and postpartum and produced more milk. Nonesterified fatty acids plasma concentrations were significantly higher in the energy group, while cholesterol was higher in the control group. Treated cows had higher levels of plasma insulin, IGF-I and leptin pre-calving. IGF-I, leptin and T 4 were diminished during the early postpartum period in both groups. Insulin levels were also diminished in the control group, but levels remained high in the energy-supplemented group. Treated cows ovulated sooner after parturition than controls. We conclude that Energetic supplementation prepartum in cows under grazing conditions increased milk production and reduced the reinitiation of ovarian activity, consistent with a better EB (BCS), higher prepartum levels of IGF-I, leptin and insulin, and higher insulin levels during early postpartum.

Involvement of thyroid hormonal regulation of seasonal patterns of reproduction as well as ovarian endocrine function are discussed.

Healthy multiparous Holstein-Friesian cows (n = 22, parity: 2–4) from a large-scale dairy herd in... more Healthy multiparous Holstein-Friesian cows (n = 22, parity: 2–4) from a large-scale dairy herd in Hungary were subjected to an intravenous glucose tolerance test 10–15 days after calving. AluI genotype of growth hormone, several plasma metabolites and metabolic hormones were determined, and current and previous lactation yields were recorded. We also used the Revised Quantitative Insulin Sensitivity Check Index (RQUICKI) and its modified version (RQUICKIBHB) for the estimation of peripheral insulin sensitivity. The majority of cows (n = 18) was leucine homozygous (LL), four were heterozygous (LV) and there were no valine homozygous (VV) animals in the population. Current average milk production was not different between AluI genotypes, but LV cows tended to have higher 305-day previous lactation yields (P = 0.13). There is growing evidence, however, for rapid, "non-nuclear" regulatory effects of E2 on lymphocytes. Such effects are likely mediated by putative membrane-associated receptor(s) (mER), but the mechanistic details and the involved signaling pathways still remained largely unknown because of their complexity. Here, we show that in lymphocytes, mERs can signalize themselves, and upon ligation, they are able to coordinate translocation of other E2Rs to the PM. Our data firmly imply existence of a complex, dynamic network of at least seven ER forms in murine lymphocytes: cytoplasmic and membrane-linked forms of ERα, ERβ, or GPR30 and a mER that can receive extracellular E2 signals. Tests for monitoring the carbohydrate/energy metabolism of (farm) animals are less commonly performed in veterinary laboratories, since most of the reliable determinations, both automated and manual, are relatively expensive. The aim of this study was to develop a precise, money- (and time-) saving automated micro method for measuring SeFa. ELISA microplates (20 µL samples and 200 µL reagents) and an automatic microplate autoreader were used. The classical nitroblue tetrazolium (NBT) stain reagent solution of Johnson et al. (1982) was modified using a SIGMA reagent to render it stable for up to one year. An experiment was carried out with young male New Zealand White (NZW) rabbits to establish live b... more An experiment was carried out with young male New Zealand White (NZW) rabbits to establish live body weight changes, body measurements, body composition and sexual maturity as a function of feeding intensity. Animals in Group 1 ('AL', n = 10) were fed ad libitum, while those in Group 2 ('RS', n = 10) received restricted feeding corresponding to 70% of the ad libitum level. The starting liveweights were practically the same (0.907 ± 0.146 and 0.911 ± 0.147 kg in Group AL and Group RS, respectively). The feeding trial lasted from 6 to 22 weeks of age. The average body weight was significantly higher in Group AL from 7 to 22 weeks of age. At 22 weeks of age the body weight of RS rabbits was 85.64% of the weight of AL animals (3.22 ± 0.52 kg and 3.76 ± 0.33 kg, respectively). Leptin and leptin receptor were studied in the mammary gland of non-pregnant dry and lactating co... more Leptin and leptin receptor were studied in the mammary gland of non-pregnant dry and lactating cows. Using RT-PCR it was demonstrated that leptin and its short (Ob-Ra) and long (Ob-Rb) receptor isoforms are expressed both in the dry and the lactating mammary gland tissue. Tissue distribution of leptin and its receptor mRNA transcripts were examined by insitu hybridisation, while the leptin protein was localised by immunohistochemistry. Although in situ hybridisation is semiquantitative, our morphological data suggest that the epithelial leptin mRNA expression of the lactating gland is higher than that of the dry gland. To compare the leptin mRNA levels between dry and lactating udders competitive PCR was used, which showed no difference in leptin expression for the whole mammary tissues. The lack of difference in total leptin mRNA levels is explained by the high adipose tissue content of the dry mammary gland. In periparturient cows the energy homeostasis turns into a negative energy balance that may shift the physiological regulation of energy balance towards pathological processes. Propylene glycol (PG), as a complementary source of energy used in the nutrition of dairy cows, alters systemic thyroid hormone economy; however, the exact mechanism through which highly glycogenic feed supplements impact liver metabolism is little known. Previous studies showed that only leptin receptors are expressed in the liver of cows, and now we report that leptin mRNA is expressed in the liver of cows as well. The basic patterns of thyroid hormones [thyroxine (T4) and 3,3',5-triiodothyronine (T3)] and ... more The basic patterns of thyroid hormones [thyroxine (T4) and 3,3',5-triiodothyronine (T3)] and the T4 and T3 responses induced by thyrotropin releasing hormone (TRH) are reported in captive female barn owls (Tyto alba) during the non-breeding period. The main findings of the study, conducted on a total of 10 owls, are as follow: (1) The thyroid gland of barn owl can be stimulated by the classical TRH stimulation test. (2) T3 response was much more pronounced both under cold (around 10°C) and warm (around 20°C) conditions, whereas T4 response ranged so widely that we could not point out any significant change in it. (3) Basal T3 plasma level was significantly (p = 0.036) higher in birds exposed to cold temperature, and they responded to TRH treatment with a lower plasma T3 elevation than the birds kept in a warm chamber. Changes of progesterone (P4) profiles and oestrous cycle were investigated up to 70 days in 20 su... more Changes of progesterone (P4) profiles and oestrous cycle were investigated up to 70 days in 20 superovulated Holstein-Friesian cows in a dry tropical environment (Brazil). Superovulated cows showed no significant differences in relation to P4 level at the time of embryo recovery (39.0 ± 27.1 nmol/L, P = 0.536), first and second (12.0 ± 6.0 and 10.7 ± 2.2 nmol/L, P = 0.543) cycle. There was a close correlation between serum P4 concentration and the number of corpora lutea (CL; 13.3 ± 9.5) at the recovery (P < 0.0001). After the embryo collection, cows returned to cycle in different ways: (i) group of donors returning to cycle after 2.2 ± 0.8 days, (ii) group with a delay of 11.0 ± 1.9 days; and (iii) animals having a long (28.8 ± 2.2 days) acyclic period, which is significant (P < 0.001). Holstein-Friesian cows were put on the Provsynch regimen in winter (n = 10) and in summer (n = 10). Blood sampling (starting 35 days post partum) and rectal ultrasound examinations (starting post insemination) were carried out once a week in each examination period. Metabolic [plasma nonesterified fatty acid (NEFA) and beta-hydroxybutyrate (BHB) levels, ferric reducing ability of plasma (FRAP) and serum beta-carotene] and endocrine parameters [plasma thyroxine (T4), triiodothyronine (T3), insulin-like growth factor (IGF-I) and insulin levels] were measured.
