This service manual is intended primarily for service technicians who are trained by Leica
Microsystems and for authorized Leica Microsystems employees. Duplication and distribution
to other persons is strictly prohibited.
With this service manual and a training certified by Leica Microsystems CMS GmbH, you can
correctly set up the device described here, operate it and carry out all required service work.
Detailed information about unpacking and installing the microscope you will
find in the Operation Manual "Leica DM5500B, DM6000B, DM6000M"
(11933966).
This service manual must be read carefully before assembling, commissioning and using the
device and must be kept for later reference. Also observe other documentation such as the
user manual for the microscope or manuals for other components.
As soon as a new version of the service manual is published, you will automatically be notified
by e-mail. The current version is provided in the Leica Microsystems CMS GmbH intranet.

2 Requirements for the Service Personnel

This service manual assumes a measurable level of knowledge on the part of the Leica service
personnel in order to understand and carry out the instructions described here in a manner that
complies with Leica's quality standards.
The service personnel must be able to document the following knowledge and completion of
the following Leica training units:
• Experience with microscopes in general: replacing components, performing setup

and teach-in work. This also includes experience with Koehler illumination and
changing and adjusting mercury and xenon lamps.
• Leica DM4000/5000/6000 FS Training
The service personnel must be physically capable of moving heavy objects (such as table
boards and packaging).

3 Intended Use
3.1 IVD Devices
3 Intended Use
The owner/operator and users of the device are responsible for intended and safe use, for safe
maintenance and for compliance with the applicable safety regulations. The owner/operator
and users of the device are fully liable for all consequences resulting from the use of the device
for any purposes other than those listed in the user manual or the online help.
The manufacturer assumes no liability for damage caused by, or any risks arising from, use of
the microscopes for purposes other than those for which they are intended, or not using the
microscopes within the specifications of Leica Microsystems CMS GmbH. In such cases, the
Declaration of Conformity shall be invalid.
In the standard configuration the microscopes are not suitable to couple laser radiation (e.g.
via the camera port) without additional laser safety measures, as then the user is risked to be
harmed by laser radiation (especially eye injury). Leica Microsystems offers special
microscope variants suitable for use with lasers, with additional safety installations. Laser
couplings require safety appliances accordingly, that need to be checked and installed by
qualified personnel.
Further intended use and standards depend on the purpose of use of the microscope. Apply
the regulations valid for your device. The order number allows you to determine if athe
microscope is declared as lab device or industry device:
3.1 IVD Devices

The Leica DM5500 B and DM6000 B microscopes, to which this user manual belongs, are
designed for biological routine and research applications.
This includes the examination of samples taken from the human body with a view to providing
information on physiological or pathological states or congenital abnormalities, or to
determining the safety and compatibility with potential recipients, or to monitoring therapeutic
measures.
3.2 Industry Devices (Non-IVD Devices)
The Leica DM 6000 M, DM5500 Q, DM6000 CS and DM6000 CFS microscopes are intended
for materials science examinations and applications in laboratory settings - not for in vitro
diagnostics (IVD) applications.

You can find the serial numbers of the Leica DM5000, DM5500 and DM6000 on the base of
the device. Leica Microsystems requires the serial number of the device to process installation
logs, service reports and inquiries correctly.

8 General Safety Notes
8.1 First Contact with the Device
8 General Safety Notes

The safety notes are intended for your protection. You should observe them without fail.
You will find the safety notes:

• in this service manual
• in the corresponding operating manual
• as safety labels on the device
• in included instructions and safety data sheets, e.g. for additional auxiliary
devices
8.1 First Contact with the Device

• Make sure that the device is free of hazardous substances and infectious agents.
This applies particularly to devices in biological-medical research labs.
• Make sure that all safety features are ready to operate. Safety features that have
been disabled, damaged or modified can cause severe injury to the eye or other
parts of the body or property damage.
• Make sure that missing or damaged safety labels are immediately attached at the
described locations.
• Do not place any combustible or flammable objects near hot surfaces.
8.2 Before Switching on the Device

• Keep your hands away from the microscope’s specimen area, both before
starting the device and when it is already in use. There is a risk of injury from
rotating objectives and the motorized specimen stage.
• Make sure that the device is completely dry before connecting it to the power
supply or turning it on.
• If country-specific: Only use grounded outlets. The protective effect must not be
nullified by using an extension cable without a protective conductor.
• Do not unplug the device or individual components unless the device is de-
energized.
• With an inverted microscope: Tilt the illumination arm back so that the motorized
specimen stage cannot cause any damage to the condenser, objective or
specimen during the initialization.
8.3 If Service Work Is Performed

• Avoid creasing, bending, crushing, tightly coiling, or in any other way damaging
the cables and cooling tubes.
• Ensure that the customer has backed up his or her existing data.

If the following cases of malfunction occur, find the causes and remedy them. Start by
immediately switching off the device if:
• Coolant leaks
• Cables are damaged
8.5 Constraints on Components

• Do not connect any external equipment. The responsible Leica office or CMS
support must be consulted whenever making alterations, modifications or
combinations with non-Leica components that are outside of the scope of this
manual.
• Use only the power cables approved by Leica to connect the individual peripheral
devices to the power supply.
• Only use fuses that are listed in this manual. The type of current and amperage
must not deviate.
8.6 Safety Notes for the Owner/Operator

• The owner/operator is responsible for the safe operation and maintenance of this
device in accordance with the intended purpose.
• The owner/operator is responsible for all safety features being in ready-to-operate
condition.
• The owner/operator must comply with the specified ambient conditions (see
chapter 6).
9 Warning Messages Corresponding to Specific

Situations and Actions
The warning messages that correspond to specific situations and actions are intended to warn
you about dangers that can occur in a specific situation or during a specific action. In order to
prevent these dangers, the warning messages contain specific instructions or conditions that
must be strictly observed and complied with.
9.1 Risk Level of a Warning Message
Each warning message contains a signal word which is used to classify the level of risk:
• WARNING: Hazard with moderate risk which could result in death or serious
physical injury if not avoided.
• CAUTION: Hazard with low risk which could result in minor or moderate physical
injury if not avoided.

9.4 Messages with the i Symbol
Such a declaration either provides extra information about a specific topic or

contains special instructions for handling the product.
10.1 ESD Protection

All service work with ESD sensitive components or assemblies must be performed in a
workplace that is set up properly for that purpose. Such an ESD protected workplace requires
a portable antistatic field service kit, ESD protected tools and wearable components:

10.1 ESD Protection
• A foldable, grounded conductive antistatic mat as worktable covering to place and

work on ESD sensitive components. The mat requires a connector for a wrist
strap, a ground cord (with crocodile clip or grounding connection for country-
specific sockets) and a series resistor of 1 MΩ in order to discharge at a slow
rate. The resistor allows high-voltage charges to leak through to earth and also
prevents a shock hazard when working with low-voltage parts.
WARNING Risk of electric shock
When using a wrist strap, make sure that the connections to the ground have
a series resistor of 1 MΩ.
• A wrist strap that is connected to the grounded antistatic mat on the worktable or
to grounding connections. Your wrist strap must be grounded and used at all
times when working on ESD sensitive components.
• ESD protected tools, e.g. screwdrivers, Allen keys, pliers, tweezers.
• ESD protection clothing (e.g. ESD jacket or ESD T-shirt) which creates a
wearable version of a Faraday cage. This shields ESD sensitive components
from static charges from your normal clothing. Additionally recommended are
either ESD shoes or a pair of heel grounders worn over your shoes to provide
consistent grounding while in motion. Neither of these can replace the wrist strap.
Additionally observe the following instructions:

• Always store and handle components or assembly groups with special ESD
protective packaging when you leave an ESD protected zone: Pack all ESD
sensitive components and spare parts, even defective ones, in shielding, anti-
static packaging intended especially for this purpose and labeled noticeably with
the corresponding ESD safety sign.

• Connect the crocodile clip of the ground cord from your antistatic mat to the
grounding screw of the device, for example the CTR box or the supply unit. These
devices of course must be grounded as well.

• Before you exchange a component, switch off the device.

• When working on the main circuit, disconnect the power plug.
The grounding of the device is lost after disconnecting the power plug. In this
case, you have to ground the antistatic mat and the wrist strap with an earth
bonding point plug with a series resistor of 1 MΩ.

• Note the manufacturer’s safety data sheets for the immersion fluids.
• Only use immersion fluids that are intended for the objective.
• Only use one small drop of immersion fluid. The immersion fluid must not
contaminate or run in the microscope.
• Remove the immersion fluid from the specimen slide before changing to a dry
objective (air objective).
• For how to remove immersion fluid, see chapter 12.3.3.

11 Electrical Acceptance Test
11.1 General
11.1 General
You have to carry out the electrical acceptance test after each service call on the device where
components in the primary circuit (power circuit) have been changed. This applies mainly to
power supplies that are implemented in the device. This does not apply to external power
supplies that are closed.
Example: You replaced a power supply of a CTR box.
The mandatory electrical acceptance test that we prescribed is a minimum standard. If country-
specific regulations exist that go beyond the measurements that we prescribed, you also have
to comply with the country-specific requirements.
You must shut down the device if limits are exceeded when measuring. In this case, please
contact cms.support@leica-microsystems.com.
Fill in the attached inspection report during the measurement. You can also find it individually
in the documentation library. Sign it, save it (serial number_EIR) and send it. Please find a
detailed description below.
If you are a user of "Mobile Service", select the "CMS5 TSB 2016/08 Work on mains circuit"
activity code in your "Mobile Service Technical Report".
11.2 Components
The electrical acceptance test consists of the following parts:
• Visual inspection
• Measuring ground wire resistance
• Insulation measurement
• Measuring protective conductor current
• Functional test
11.3 Measuring Equipment
For measuring the protective conductor current, you need the following adapter cable. You can
order it from us: 15000010000084 Adapter cable for testing IEC-C14G+C13.
The remaining measuring equipment for the electrical acceptance test must be obtained
locally. We recommend the following measuring equipment:
• Fluke 1507 insulation tester

• Amprobe AC50A leakage current clamp multimeter

11.4 Measurements in Detail
11.4.1 Visual Inspection
Here, you have to visually inspect the power supply cord for damage. If you opened the
housing, you also have to visually inspect the protective ground cable (PE). Proceed as
described in the inspection report.
WARNING Risk of electric shock
Close the housing after having completed the visual inspection.
11.4.2 Measuring Ground Wire Resistance
The ground wire resistance is the resistance between the protective conductor terminal (PE)
and the metal housing. It must be less than 0.3  .
The measuring points are the protective conductor terminal (PE) and electrically conductive
points on the metal housing (also refer to figure 5)

If the metal housing is coated completely and you cannot find any regular or
alternative measuring point on the device (see figure 5), you have to pierce
the varnish with the gauge tip.
Below, we will describe how to measure the ground wire resistance for the Fluke 1507
insulation tester:
1. On the insulation tester, connect one measurement sensor to the "COM" connection
and the other measurement sensor to the "" connection.
2. Join both measurement sensors together to measure the line resistance of the
measuring line.
3. Hold the blue button for a few seconds until the insulation tester display shows a
series of dashes. The line resistance is now set.

4. Connect one of the measurement sensors to the protective conductor terminal (PE)
of the power plug. (The power plug must be from the original power cable!)
5. Connect the other measurement sensor to a measuring point on the housing.
6. Hold the "Test" button for a few seconds until a measurement result is displayed.
7. Repeat the measurement at another measuring point on the housing.
8. Enter the measured value into the inspection report.

11.4.3 Insulation Measurement
You measure the insulation resistance as part of the insulation measurement. That is the
resistance between the phase (L)/neutral conductor (N) and the ground protection conductor
(PE).
It must be more than 2 M. Measuring points are the protective conductor terminal (PE), the
phase (L) and the neutral conductor (N) of the power plug. The measurement occurs with a test
voltage of 1000 V.
Below, we will describe how to measure for the Fluke 1507 insulation tester:
1. Disconnect all peripheral devices.

2. On the insulation tester, connect one measurement sensor to the "COM" connection
and the other measurement sensor to the "Insulation V" connection.
3. Switch on all switches on the device.
4. Example FSU: All 3 switches on the main switch board and the power switch on the
back of the FSU.
5. Connect the first measurement sensor to the protective conductor terminal (PE) of
the power plug. (The power plug must be from the original power cable!)
6. Connect the second measurement sensor to the phase (L) of the power plug.
7. Hold the "Test" button for a few seconds until a measurement result is displayed.
8. Connect the second measurement sensor to the neutral conductor (L) of the power
plug. Repeat the measurement.

Even though the limit value for the insulation resistance is 2 M, the
insulation resistance is typically in the G range. If the value you measured
is not in the G range and you cannot find a reason for this behavior, please
contact cms.support@leica-microsystems.com.

11.4.4 Measuring Protective Conductor Current
The protective conductor current (or leakage current) is the current through the ground
protection conductor (PE). It must be less than 3.5 mA. Below, we will describe the procedure
with an adapter cable and the Amprobe AC50A leakage current clamp multimeter.
1. Disconnect all peripheral devices.

2. Connect the device to the line voltage using the adapter cable and the power supply
cord.
3. Switch on all switches on the device.
4. Clamp the leakage current clamp multimeter onto the ground protection conductor
(PE) of the adapter cable.
5. Set a measurement range from 40 mA.
6. Measure the protective conductor current. It must be less than 3.5 mA.
8. Only FSU: Repeat the measurement for the second power supply cord.

11.4.5 Functional Test

1. Reconnect the original power cable to the device.
2. Switch on the device.
3. Check whether the device is working properly.
11.5 Electrical Inspection Report
If you are a user of Mobile Service, select the "CMS5 TSB 2016/08 Work on mains circuit"
activity code in your "Mobile Service Technical Report".
Fill in the electrical inspection report during the measurement. You can also find it individually
in the documentation library. Use your own laptop, not the customer system. After having filled
in the electrical inspection report:
1. Sign it: Use the digital signature field, or use the field right down at the bottom with

12 Care and Maintenance
12.1 Cleaning Surfaces

• Protect the microscope from dust and grease.
• When the microscope is not in use, cover it with a plastic cover or a clean, lint-
free cotton cloth. After using the device, allow it to cool down to room temperature
before covering it with a dust cover. This way you will prevent condensation from
forming underneath.
• Always place dust caps over the nosepiece positions when no objectives are in
place.
• Disconnect the power supply before performing cleaning and maintenance work.
• Make sure that no fluid runs into the individual components during cleaning.
• Make sure that parts made of glass and rubber do not come into contact with
benzine, oil or lubricant, unless that is expressly required.
• To care of the individual components and peripheral devices, observe the
operating manuals provided for them.
12.1 Cleaning Surfaces

• Never use acetone, xylene or nitro thinners as they attack the lacquer.
• Never use abrasives. Abrasives can scratch the surface and thus have a negative
effect on the protection of the parts.
• Remove dust and loose dirt particles using a soft brush or lint-free cotton cloth.
• You may clean uncoated or plastic surfaces only using a dry cotton cloth or one
moistened with a little water. Other cleaning agents can attack and tarnish the
surface and cause it to become porous.
12.2 Cleaning Mechanical Parts

• Use benzine to clean the parts and assemblies. If possible, clean them outside of
their housing.
• When cleaning lacquered parts and parts with an engraving only use a soft
leather or brush.
• Remove sticky dirt as necessary either with a low-concentrated soap solution,
petroleum ether or ethanol (ethyl alcohol).
• Clean dirty exterior surfaces with a textured lacquer finish by using an oiled cloth
and wipe them dry.
• Remove lubricant and adhesive residue before treating parts with new adhesive.
• Only use lubricants or adhesives that Leica Microsystems CMS GmbH has
recommended and that can be purchased from Technical Service. Other
materials are not permitted, as only the lubricants and adhesives we tested
ensure proper function.

12.3 Cleaning Optical Components
The places on the device where lubricants and adhesives are to be found are
displayed in part C in the figures and indicated by numbers or letters.
Lubricants are only allowed to come into contact with designated places,
since they can dissolve or damage plastics.
• Clean and dry parts before using lubricant.

• Keep the optical system of the microscope clean at all times.
• Observe that optical components are not contaminated with dust, grease or other
dirt.
• Keep the tools, auxiliary equipment and work place clean at all times. This is a
prerequisite for successfully cleaning optical components.
• Check the condition of the optical parts before you begin cleaning them. To do so,
use a cotton cloth or a hair pencil to wipe them at a place where no light beams
pass through the optical medium (for example, at the edge). If you find traces of
wiping, the surface coating is soft and it will hardly be possible to be cleaned
without damaging the coating.
• To clean the optical part (for example illuminating lens or mirror), remove it from
the mount if possible.
• Never open the objectives for cleaning.
12.3.1 Tools and Accessories

You can also make your own polishing sticks. Chamois leather becomes hard after being used
multiple times for cleaning and has to be replaced regularly. The chamois leather surface can
be periodically re-roughened with the cutting edge of a razor blade. Clean the brush and
polishing stick by repeatedly dipping them in alcohol and then blowing them dry. Cotton swabs,
optical cloths and lens cleaning paper are consumables. Dispose of them after using them
once.
12.3.2 Cleaning Agents

• Ethanol (ethyl alcohol)
• AR19
• Distilled water
• Benzine
NOTICE Risk of damage to components

Never clean with acetone, xylene, ammonia glass cleaner or nitro-containing
thinners.
12.3.3 Cleaning Front Lenses of Immersion Objectives
Immediately remove the immersion medium from front lenses after use (e. g. type F immersion
liquid, in the past referred to as "immersion oil“):
1. First, remove the immersion medium using a clean cloth.

2. Once most of the immersion medium has been removed, lay a piece of fine lens
tissue over the immersion end of the lens.
3. Apply a drop of suitable solvent and gently wipe the tissue across the lens surface.
4. Repeat this operation until the lens is completely clean. Use a clean piece of lens
tissue each time.
12.3.4 Cleaning Glass Surfaces

• If a vacuum connection is available, vacuum the glass surfaces.
• Use a soft dust brush or rubber squeeze blower to clean large dust particles off of
glass surfaces. In the case of built-in optics units, make sure that no other parts
inside of the device or assembly get dirty.
• Remove persistent dirt from glass surfaces using pure alcohol or another suitable
cleaning agent.
• Begin in the middle of the surface and move the cleaning tool toward the edge in
concentric circles.

• When using ethanol (ethyl alcohol):

Soak a clean cotton cloth with ethanol, wipe off the glass surfaces and quickly
polish with soft leather.
• When using AR19 or distilled water:
First wipe dry the glass surfaces and then polish them with soft leather.
• Remove remaining lint and dust particles with a fine hair pencil.
Residual lint and dust can create unwanted background fluorescence in

fluorescence images. After cleaning, check whether all residual dust has
been removed.
12.3.5 Optical Parts with Graticule Marks
In the case of cemented optical parts, line markings or scales, make sure that no cleaning
agent with the ability to dissolve gets on the cemented spots or the graticule marks.

and a cotton cloth (twice, to avoid forming an edge),
then wipe dry using moderate pressure
• The lacquer layer in the graduation must not be
cleaned away

12.3.6 Optical Parts with Mild Fungal Infestation
In rare cases fungal infestation can occur in humid-warm climate. Optical

components often have to be replaced by new ones because the fungus can
destroy the glass surface over time and then cleaning is no longer possible.
With slight fungal infestation the optical components may be rescued.
1. Before cleaning, check the surface coating.

2. Prepare the following cleaning agent to treat optics units with and without surface
coatings:
– 3% hydrogen peroxide
– 10% ammonia solution
– Prior to application: Mix 1:1
– After use dispose of the leftovers
3. Clean the glass surfaces infested with fungus using an optical cloth and the cleaning
agent you have mixed.
4. Rinse the glass surfaces with water. Additional cleaning is described in
chapter 12.3.4 and chapter 12.3.5.

2 Repair and Service the Stand
2.1 Replace Backplane
When replacing the backplane compl. 11020530031000:

Only use the release version BZ04 or higher for DiscoveryII stands. BZ03 and
lower can not be used as replacement for DiscoveryII.
BZ04 or higher however can be used as replacement part for DiscoveryI
stands.
Observe modification instructions for connection field lamp
(11020530035000) and polarity of CTR box lamp outlet, chapter 2.3.
The customer and factory settings are saved on the backplane. After exchanging the
backplane you have to program the settings on the new backplane. The following 2 options are
possible:
• Case 1: backplane defective, but still responsive via workstation

In this case you can read out the settings from the backplane via the Leica test
software. After exchanging the backplane you can save these settings on the new
backplane.
• Case 2: backplane defective without contact to the workstation
In this case you have to adjust the settings manually and save them on the new
backplane.
2.1.1 Case 1: Backplane Defective, but Still Responsive via Workstation

1. Switch on the microscope and the workstation.
2. Download the Leica test software from the Leica FTP server.
3. Backup the customer and factory settings using the Leica test software:
Start the Leica test software. After the microscope is recognized, start downloading
the settings by using the option File  Save & Restore Settings in the Leica test
software main window.
4. Click "OK" to confirm the message „Load LMD Parameter OK“.
5. Switch off the microscope.

6. Remove the screw (figure 1, position 1) of the lamp housing flange, unscrew 4
screws (position 2) and pull off the casing (position 3).

7. Loosen the 4 screws (figure 1, position 4) of the back panel and tilt the back panel
(position 5) away.
8. Disconnect all plug connections to the "SYS Basic Module DM6000" board and
remove the back panel.
9. Replace the backplane and assemble the back panel in reverse order as described in
the working instructions 6 to 8.
10. Switch on the microscope.
11. Use the type plate program to program the correct type plate information to the new
backplane as shown in the service training.
12. Start LAS/Leica Hardware Configurator and load the latest firmware to the
backplane.
13. After finishing the firmware update, close the Hardware Configurator.
14. Restore customer and factory settings using the Leica test software.
Start the upload by using the option File  Save & Restore Settings in the Leica test
software main window.
15. Confirm the successful upload by "OK" and close the Leica test software.

2.1.2 Case 2: Backplane Defective Without Contact to the Workstation

1. Remove the screw (figure 2, position 1) of the lamp housing flange, unscrew 4
screws (position 2) and pull off the casing (position 3).

3. Disconnect all plug connections to the "SYS Basic Module DM6000" board and
remove the back panel.
4. Replace the backplane and assemble the back panel in reverse order as described in
the working instructions 1 to 3.
7. Start LAS/Leica Hardware Configurator and load the latest firmware to the
backplane.
8. Use the hardware configuration setup to configure the microscope hardware again.
Perform the relevant fine tuning (in particular parfocality) and save the settings as
factory settings, using the LAS/Hardware Configurator factory mode.
9. Check adjustment of the TL shutter (see chapter 8.3).
10. Check adjustment of movable lens of the TL module (see chapter 8.4 and 8.5).

2.3 Connectors for Lamp Connection Field Changed
2.2.2 Upgrade Procedure

1. Connect the DM5000 (8 Bit) to LAS and printout the configuration (identification
sheet).
2. Depending on the release version (ÄZ) of the CTR box, exchange the parts as listed
in chapter 2.2.1.
3. Replace the backplane and the connection field power.
Observe the modification instructions for the connection field lamp
(11020530035000) and the polarity of the CTR box lamp outlet (chapter 2.3).
5. Use the latest LAS to download latest firmware version to the backplane.
6. Use the LAS setup to configure the microscope as written on the identification sheet.
7. Perform the relevant fine tuning and save settings as factory settings, using the LAS
factory mode.
8. Check the adjustment of the TL shutter (see chapter 8.3).
9. Check the adjustment of movable lens of the TL module (see chapter 8.4).
10. Verify that the transmitted light lamp intensity can be properly controlled by changing
the lamp intensity between maximum and minimum value. There must be no irregular
"jumps" in lamp intensity. If there are jumps in the lamp intensity, replace the existing
power supply with a new power supply (11360006017000).

In rare occasions the exchange of a "rear cover complete" on a Discovery microscope or the
"lamp power interface" of a CTR 4000 box may result in the subsequent damage of the just
replaced board. In order to avoid such damage repeatedly it is mandatory to also modify the
below listed connection fields and cables:

The reason is that the cable lugs connecting to the TL/IL lamp sockets may show a transition
resistance high enough to cause a raise of temperature of the cable lug, which in turn increases
the temperature of the PCB (up to 80° Celsius) and its components. This may cause damage
of some electronic components of the PCB.
In order to avoid such transition resistance, it is mandatory also to replace the cable lugs of the
TL/IL lamp connection field and cables connected to the PCB after exchanging one of the
above mentioned PCBs.

2.3.1 Interchanged Polarity of the Lamp Outlet of the CTR5000/6000
The following modification only needs to be done when a "rear cover

complete" (including backplane complete) of a DM5000 or DM6000 has to be
replaced.
Do not follow the described procedure as a preventive action.
Ensure that the polarity of the lamp outlet of the CTR box is correct.
1. Disconnect all cables of the CTR box (except main power supply) and switch on the
CTR box.
2. Use a multimeter (DC voltage in the range of 2.5 - 12 V) to measure the polarity (see
figure 3).

3. If the polarity of the lamp outlet of the CTR5000/6000 is reversed, disconnect mains
of the CRT box.

4. Interchange the cables of the lamp power supply unit connected at X2A and X3
inside the CTR box:

2.3.2 Interchanged Polarity of the Cable Lug and the Connector on

the PCB
When exchanging the "backplane complete" also the lamp connection field needs a
modification.

It is important that the positive (+) voltage is connected to X8, otherwise

electronic components (V8) may be damaged.
1. Remove the lamp connection field from the stand.

2. Use an ohmmeter to check whether you have the same polarity as shown below.

3. If the polarity is interchanged (X8 = negative) you can either order a replacement of
the "connection field lamp" (11020530035000) or correct the polarity yourself:
1. Contact cms.support@leica-microsystems.com and order the required cable lugs
(3 pcs. per "connection field lamp")
2. Cut off the X8 cable lug.
3. Unsolder the cable at the negative connector of the lamp socket.
4. Interchange both cables.
5. Cut off the existing 3 cable lugs (X8, X9, X10).
6. Use a suitable crimping tool (see figure 8) to ensure the best possible contact
between cable and cable lug.

Should you not have a proper crimping tool, please order the modified
"connection field lamp" (11020530035000) via SAP.

7. After the new cable lugs are accurately crimped onto the cables, check the
polarity again.
8. If the polarity is correct, stick the delivered "MODIFIED" label onto the connection
field as shown below.

4. Remount the lamp connection field and connect it to the "backplane complete" as
usual.
5. Check the functionality of the microscope, especially switching between TL/IL.

1. Insert 2 triangle rails (figure 11, position 11) with lubricant 311 into the right side of
the guide box.
2. Insert the roller cage (figure 11, position 12) and 14 rollers (position 13) with lubricant
311 into the right side of the guide box.
3. Insert 4 triangle rails (figure 11, position 14) with lubricant 311 into the guide plate
(position 10).
4. Insert the roller cage (figure 11, position 16) and 14 rollers (position 13) with lubricant
311 into the left side of the guide plate (position 10).
5. Insert 2 triangular rails (figure 11, position 15) with lubricant 311 into the guide rail
(position 9).
6. Mount the guide rail (figure 11, position 9) onto the left side of the guide plate
(position 10) and insert the whole package into the guide box.
7. Slightly mount the guide rail (figure 11, position 9) with 4 screws (position 8).
8. With the 4 screws (figure 11, position 7) set a swift and easy movement of the guide
plate (position 10) free from any backlash.
9. Tighten the 4 screws (figure 11, position 8).
10. Check the gear and correct if necessary.
11. Assemble the rail (figure 11, position 6) with the screws (position 5).
12. Assemble the rail (figure 11, position 2) and the rack (position 3) with the screws
(position 1).
13. Fit the guide box and the z-drive.

1. Insert the guide box (figure 10, position 15) into the stand, push it to the right against
the stops (position a) and screw it in place with 4 screws (position 14).
2. Hook the 2 springs (figure 10, position 12) into the screws (position 13).
3. Insert the z-drive (figure 10, position 11) into the stand from the left and fix it with 2
screws (position 10).
Pay attention that the drive knob does not grind against the stand.
4. Assemble the washers (figure 10, position 4) and the right-hand fine drive knob
(position 3).
5. Assemble the cover (figure 10, position 9) with the 2 screws (position 8).
Make sure that the screws grip into the thread of the cover.
6. Mount the 2 metal plates (figure 10, position 2) with the 4 screws (position 1).
7. Assemble the stage bracket with the stage, condenser carrier and condenser.

1. Remove the z-drive (see previous chapter).
2. Unscrew the 2 screws (figure 13, position 1) and remove the flange (figure 13,
position 2).
3. Unscrew the locknut (figure 13, position 3) using a 22 mm jaw spanner while holding
the adjusting ring with a second 22 mm jaw spanner.
The wrench opening must have a thickness of less than of 3.5 mm.
4. Unscrew the adjusting ring (figure 13, position 4).
5. Remove the spring washer (figure 13, position 5) and plastic washer (figure 13,
position 6).
6. Remove the axis (figure 13, position 7) of the bearing (figure 13, position 12).
7. Carefully warm the bearing ring (figure 13, position 8) with a hot air gun to about
70° C to melt the Loctite in the thread.
8. Hold the gearwheel of the coarse drive axis (figure 13, position 9) with a rubber cloth
or a large pair of rubber-jawed tongs and unscrew the bearing ring (figure 13,
position 8) with a stud wrench or scissor-type wrench.
9. Remove the coarse drive axis (figure 13, position 9), balls (figure 13, position 19) and
washers (figure 13, position 11).
10. Clean all parts thoroughly in benzine and afterwards dry them.

1. Grease all parts with the lubricant specified (410) and as indicated in figure 13.
2. Insert the 2 washers (figure 13, position 11).
3. Insert 18 balls (figure 13, position 10) into the bearing (position 12) on the side of the
locking ring (position 13).
4. Carefully put the axis (figure 13, position 9) into the bearing (figure 13, position 12).
Avoid lubricant getting onto the thread.
5. Carefully put the second set of balls (figure 13, position 1) into the bearing
(position 12).
6. Screw the bearing ring (figure 13, position 8) onto the axis (position 9) about 2 turns
of the thread and apply a small amount of "Loctite 222" to the thread.
7. Screw the bearing ring fully onto the axis (figure 13, position 9), ensuring smooth and
easy movement free from any backlash.
8. Reapply the plastic washer (figure 13, position 6), spring washer (position 5) and
adjusting ring (position 4).
9. Use the adjusting ring (figure 13, position 4) to set a torque of 8 - 10 N on the coarse
drive knob. This corresponds to an axial spring travel of approximately 0.3 - 0.4 mm.
10. Lock the adjusting ring (figure 13, position 4) with the locknut (figure 13, position 3).
11. Assemble the flange (figure 13, position 2) with the 2 screws (figure 13, position 1).
4.4 Assemble the z-Drive in the Microscope

1. Insert the z-drive (figure 12, position 11) into the left side of the stand and fix the 2
screws (position 10). Take care that the drive knob does not grind against the stand.
2. Assemble the washers (figure 12, position 4) and the right-hand fine drive knob.
3. Assemble the cover (figure 12, position 9) with 2 screws (position 8).
Make sure that the screws grip into the thread of the cover.
4. Use the 4 screws (figure 12, position 1) to fix the 2 metal plates (figure 12,

5.3 Adjust the End Switches (Lower and Upper End Position)
1. Insert the guide box and mount it onto the stand pressing it against the mechanical
stops (figure 22, position 1) and fastening it with the screws (figure 20, position 2).

2. When mounting the casing (figure 17, position 1), pay attention not to screw the
screws (figure 16, position 1) too tight.
3. When assembling the handwheels, make sure that they do not rub against the panel
of the stand.
4. Check all cable connections.
5. Check all functions.
5.3 Adjust the End Switches (Lower and Upper End Position)
To adjust the 2 end switches for monitoring the mechanical end position, the guide box has to
be disassembled.
1. Loosen the 2 screws (position 1) of the end switch strip (figure 23, position 2) and
slide right up to the top.

2. Turn the clean dumper (figure 24, position 1) by hand to move the dovetail guide
down to the lower mechanical stop. Turn the dumper about 1 1/2 turns back.
3. Loosen the screws (figure 23, position 3) of the lower end switch and position it that
way it works correctly.
4. Tighten the screws.

5. Move the guide box with the dumper to the upper mechanical stop. Turn the dumper
back by about 2 turns.
6. Position the upper end switch so that it just switches.
7. Check the whole range of movement between the end switch switching points. It
should be approx. 25 mm +/- 0.5 mm.
5.4 Adjusting the Wheel Coder of the Handwheel

The wheel coder disk has to be adjusted within the light barrier fork.
The distance of the 2 receivers of the light barrier must not be more than 0.9 mm (figure 25).

1. Disconnect connector X8 from the IL axis adapter board.
2. Remove the IL turret (see chapter 7) to reach the screws of the nosepiece.
3. Unscrew the 2 DIC turret holding screws from the nosepiece and remove it.
4. Unscrew the 4 screws while holding the nosepiece.
5. Remove the nosepiece and place it down carefully.

1. Unscrew the 3 screws (1) and take off the ring.
2. Unscrew the 3 screws (2) and take off the encoder disk.
3. Manually rotate the encoder disk until an index notch is opposite to the
corresponding objective thread at a click-stop position (see figure 27).

1. Thread the X8 cable through the hole (figure 29, position 1).
2. Insert the nosepiece, press it against the stops and fix it with 4 screws. Make sure
that the gear ring of the nosepiece engages in the pinion of the motor.
3. Place the DIC turret in the slit between the IL axis housing and nosepiece.
4. Use 2 screws to fix the DIC turret. Mount the screws from the bottom of the
nosepiece.
5. Lay the cables in accordance with the cable routing.
6. Plug connector X8 into the IL axis adapter board.
7. Assemble the IL turret (see chapter 7).

4. Loosen the 2 grub screws of the pinion (the screws are secured).
5. Remove the pinion together with the encoder disk.
6. Unscrew the 2 screws that fasten the motor and remove the motor.

2. After loosening the 2 screws (figure 30, position 2) behind the metal plate (figure 30,
position 3), arrange all the cables and wires so that the 2 holes in the casing panel
remain free.
3. Make sure that the cables do not lie in the light path and that no moving parts are
blocked.
6.6 Align the Click-stop Sensor (DM6000)

1. Connect the DC-multimeter, range 5V/10V, to the green and white cable of connector
X3/X3.
2. Align the click-stop sensor such that a voltage of 4.2 or higher is measured when it is
in a click-stop position.
The voltage between the 2 click-stop positions should be 0.2 V or less.
3. Place the nosepiece in the snap-in position and move the click-stop sensor until you
reach the above mentioned specifications.
4. Check the values for all other snap-in positions as well as the low signal value for the
nosepiece positions in between.

7.1 Disassemble the 8-position IL Turret

1. Unscrew the 3 screws (figure 33, position 1) and take off the mount.
2. Take off the front cap (figure 33, position 2) and remove any filters.
3. Unscrew the 2 pins (figure 34, position 1). The pins can be loosened with a 1.5 mm
Allen key in the holes.

4. Remove the X2 cable from the adapter board IL and disconnect the plug of
cable X1-X4.
5. Unscrew 3 screws (figure 35, position 1).

Assembly is carried out in the reverse order.
1. Before assembling the IL turret, check that the cables run through the cable groove
(figure 37, position 1).

2. Insert the IL turret by pushing it against the back stop and the 2 stops on the right
(figure 37). Make sure to fix it with the 3 proper screws (figure 35, position 1).
Observe to use the correct screws.
7.3 Adjust the 4/5- and 8-position IL Turrets
Due to the inaccessibility of the sensor board screws, the 8-position IL turret
can only be adjusted when disassembled. Apart from that the adjustment
process is equivalent to the 4-position or 5-position turret.

The distance between the fork light barrier (receiver side /imprint TCUT 1200) and the encoder
disk must be large enough to prevent rubbing but should not be more than 0.9 mm.

The light barrier should be assembled in the click-stop position +/- 0.2 mm in the center of the
rectangular opening of the encoder disk.
1. Slightly loosen the screws (figure 37, position 3 and figure 39, position 1).

2. If there is no centricity mark at position 1 of the encoder disk, mark the position with a
pen to obtain an optimal adjustment.

3. Check the clearance between the gear wheel and the driving pinion or intermediate
wheel. There should not be too much play between them.

1. Turn the microscope carefully upside down to reach the TL module in the stand
bottom side.
2. Unscrew the screws of the bottom cover and remove it.
3. For better handling also unscrew the 4 screws to remove the rear base plate.
4. Remove the flat ribbon cable from the graphic display.
5. Unfix the 3 holding clamps (figure 42, position 1) of the TL module and lift the module
out of the stand.

8.2 Center the Aperture Diaphragm in the TL Diaphragm Module
8.2 Center the Aperture Diaphragm in the TL Diaphragm Module
When an objective with a magnification factor of less than 10x is turned into the light path, the
condenser top automatically moves out of the light path. That way the aperture diaphragm gets
the function of a field diaphragm.
After exchanging the TL diaphragm module, the aperture diaphragm (integrated in the TL
diaphragm module) has to be centered in the microscope in order to get a well centered image.
1. Screw the adjustment objective (figure 44) into an empty position of the nosepiece.

2. Remove the condenser from the condenser holder.

3. Turn the microscope carefully upside down to reach the TL module in the stand
bottom side.
4. Unscrew the screws of the bottom cover and remove it.
5. If the adjustment carriage (figure 45, position 1) is secured with adhesive (position A)
to prevent maladjustment, the adhesive has to be removed carefully with a knife.

8.3 Adjust the Shutter
6. Put the microscope in a position in which it is possible to look through the eyepiece
and reach the adjustment screws (figure 45, position 3) of the TL diaphragm module
at the same time.
7. Close the aperture diaphragm (field diaphragm function) with the keys (see figure 47)
until it becomes visible in the eyepiece. If the image of the diaphragm is not sharply
focused, adjust the movable lens (see figure 43).
8. Loosen the fixing screw (figure 45, position 2).
9. Using the 2 adjustment screws (figure 45, position 3), center the diaphragm to the
scale of the adjustment objective (figure 46).

10. Tighten the fixing screw (figure 45, position 2) check the centricity and correct if
necessary.
11. Secure the adjustment carriage with adhesive (figure 45, position A).
12. Plug the flat ribbon cable into the graphic display.
After exchanging the transmitted light diaphragm module, the adjustment of

the movable lens (see figure 43) must be checked and corrected if necessary.
The "Closed" position of the shutter can be adjusted with individual commands in the
hyperterminal program. Only 2 commands are required. Do the following steps to adjust the
shutter. Do not vary from the order specified.
The hyperterminal program is installed on every PC with the Microsoft

Windows operating system. If no hyperterminal program is on your PC, install
this program.

1. Start the hyperterminal program.

2. Enter the command "84020 0" to initialize the aperture diaphragm.
This command closes the aperture diaphragm to the minimum opening (=

trigger point of the fork light barrier = stop 1) and defines this position as the
starting position.
In this position the remaining, minimal aperture diaphragm opening is not

completely covered by the shutter. Light still passes around the edge of the
shutter.
3. Look into the eyepieces to check, if there still is light visible.

4. If light is visible, permanently press the pre-programmed "AP" key (figure 47) to
reduce the aperture opening.
The shutter position is changed step by step. To have the aperture diaphragm
opening completely covered by the shutter, the "AP close" key has to be pressed
repeatedly until the shutter interrupts the light beam. The end of the adjustment
range must be reached in five steps at most; usually 2 or 3 steps are adequate.

5. Enter the command "84025" into the hyperterminal program to store the shutter offset
value.
This command stores the offset value (-1 to max -5) between the initialization
position and the "shutter closed" position. After this, the shutter completely
releases the diaphragm opening again ("shutter open" position) and the
diaphragm is opened to the stop that was stored with the particular objective.
6. After executing the command the stored offset value (e.g. 84025 -2) is returned by
the hyperterminal program.
7. The current AP diaphragm opening can be read out with the command "84023"
(GET_ POS_APBL_TL).
You will receive the command with a return value for the current position:
• -1 to -5: offset values for the "shutter closed" position
• 1 - 38: current position of the aperture diaphragm (1 = minimum opening)

8.4 Adjustment of the Movable Lens (Achromat) in the TL

Diaphragm Module
When an objective with a magnification factor lower than 10x is turned into the light path, the
aperture diaphragm assumes the function of the field diaphragm. To obtain a sharp image of
the aperture diaphragm, an achromat has to be moved in the TL diaphragm module. This
happens automatically when an appropriate objective is turned into the light path.
If the distance between the aperture diaphragm and the lens has been altered mechanically or
the module has been removed, the “Field diaphragm position“ of the lens has to be readjusted
and stored by the "AdjustAdaptingLens.exe" software.
The same applies after exchanging the "MAN1 Basic Module" board (DM 4000) or "DM 5000
Basic Module" (DM5000) or "SYS Basic Module" (DM6000 B/M). The stored values are then
lost.
1. Install the "Leica Application Suite" (LAS) software package, version 2.0 R1 or higher
on the PC.
2. Check if the microscope works correctly.
3. Place a specimen on the stage.
4. Adjust the Koehler illumination.
5. Start the "AdjustAdaptingLens.exe".
6. Click "Connect" (figure 49, position 1) to get a connection to the microscope.
With DiscoveryII microscopes only USB connection is possible. Therefore the

latest version of the "TL Adapting Lens Adjusting" software has to be used
(download from the FTP server).
Add "-usb" to enable using the software via USB:

7. Select "Fix Pos 1" and check the focus of the specimen image.

"Fix Pos 1" designates the position of the movable lens for objectives of 10x
magnification or higher.
"Fix Pos 2" designates the position of the movable lens for objectives of less
than 10x magnification.
8. Set position 2 for objectives with less than 10x magnification:

Select "Fix Pos 2" (figure 49, position 3) and move the adjustment slider (figure 49,
position 3) until a sharply focused image of the aperture diaphragm (now working as
a field diaphragm) is obtained in the eyepieces (maximum value = 249)
9. After successful adjustment, click "Save As Fix Pos 2" (figure 49, position 4).
10. Check the positions once again and then burn the new value into the EEPROM of the
microscope by clicking the button "BURN" (to factory settings, figure 49, position 5).
11. Click "Disconnect" to end the program.

9 Firmware
9.1 Update Firmware via Hardware Configurator

You can do the firmware update for all modules via the Hardware Configurator. The instructions
in chapter 9.1 are an example for a firmware update. In chapter 9.2 the exception of the touch
display is explained.
9.1 Update Firmware via Hardware Configurator
The touch panel of DiscoveryII microscopes are delivered with the current Leica STP firmware
(current at the time of delivery). If you have to update the firmware, proceed as follows:
1. Start the Hardware Configurator.
2. Click "Firmware Update". The module overview window opens.
3. Choose "Touch Panel Intern" and open the menu via right-click.
4. Click "Start Update".
5. After finishing the update, close the Hardware Configurator.
6. Restart the microscope.
9.2 Download Display Firmware from FTP Server
If you have to replace the display board, the new board comes without the Leica firmware. The
firmware update can not be done via the Hardware Configurator. Download the firmware from
the Leica FTP server:
1. Download the firmware file from the FTP server to a USB stick.
The latest build of the firmware is available in the following folder:
ftp://ftp.leica.de/Firmware_Software_Patches_Tools/latest_Firmware_Files-Patches/
Touchpanel/
Pay attention to use the correct firmware version corresponding to the board of the
display (see table below).

2. Execute the correct file (see table above) by double-clicking. The following window
opens:

3. Select the drive, which represents your USB memory stick, as destination folder.
Do not create any subfolders as the destination folder.
4. Click the "Install" button to install the required files onto your USB memory stick.
5. Rename the folder on the USB stick corresponding to the board of the display:

6. Remove the USB stick from the PC.

7. Remove the USB cover on the right side of the microscope to reach the USB port.
9. Insert the USB memory stick into the USB port on the right side of the microscope.
10. After the USB memory stick is recognized, power cycle (off/on) the microscope.
The memory stick is recognized automatically, and the firmware installation will start.
11. Once the firmware installation has been completed, the calibration routine
automatically starts. You have to set points, marked as crosses, to calibrate the
display.
12. Touch the first cross with a suitable pen until the cross jumps to the next position.
Repeat touching the crosses until you have set all points.
Make sure to touch the calibration crosses very precisely. The routine will not
come to an end until all calibration crosses are touched correctly.

In the ServiceMode of the touch display the following features are available:
• activate data logging (only activate for debugging, for normal operation always
de-activate logging)
• save log files
• activate manual control
• test light barriers and push buttons
• calibrate the touch panel
2. Wait for the progress bar and press the touch display in the labeled area for 4 times:

After restarting the microscope should work in ServiceMode.

3. To terminate the ServiceMode restart the microscope.
