With this service manual and a training certified by Leica Microsystems CMS GmbH, you can
correctly set up the device described here, operate it and carry out all required service work.
Detailed information about unpacking and installing the microscope you will
find in the Operation Manual "Leica DM5500B, DM6000B, DM6000M"
(11933966).
This service manual must be read carefully before assembling, commissioning and using the
device and must be kept for later reference. Also observe other documentation such as the
user manual for the microscope or manuals for other components.
As soon as a new version of the service manual is published, you will automatically be notified
by e-mail. The current version is provided in the Leica Microsystems CMS GmbH intranet.

3 Intended Use
3.1 IVD Devices
3 Intended Use
The owner/operator and users of the device are responsible for intended and safe use, for safe
maintenance and for compliance with the applicable safety regulations. The owner/operator
and users of the device are fully liable for all consequences resulting from the use of the device
for any purposes other than those listed in the user manual or the online help.
The manufacturer assumes no liability for damage caused by, or any risks arising from, use of
the microscopes for purposes other than those for which they are intended, or not using the
microscopes within the specifications of Leica Microsystems CMS GmbH. In such cases, the
Declaration of Conformity shall be invalid.
In the standard configuration the microscopes are not suitable to couple laser radiation (e.g.
via the camera port) without additional laser safety measures, as then the user is risked to be
harmed by laser radiation (especially eye injury). Leica Microsystems offers special
microscope variants suitable for use with lasers, with additional safety installations. Laser
couplings require safety appliances accordingly, that need to be checked and installed by
qualified personnel.
Further intended use and standards depend on the purpose of use of the microscope. Apply
the regulations valid for your device. The order number allows you to determine if athe
microscope is declared as lab device or industry device:
This includes the examination of samples taken from the human body with a view to providing
information on physiological or pathological states or congenital abnormalities, or to
determining the safety and compatibility with potential recipients, or to monitoring therapeutic
measures.
The Leica DM 6000 M, DM5500 Q, DM6000 CS and DM6000 CFS microscopes are intended
for materials science examinations and applications in laboratory settings - not for in vitro
diagnostics (IVD) applications.

5 Serial Numbers
You can find the serial numbers of the Leica DM5000, DM5500 and DM6000 on the base of
the device. Leica Microsystems requires the serial number of the device to process installation
logs, service reports and inquiries correctly.
6 Ambient Conditions
Use of the Leica DM5000, DM5500 and DM6000 is permitted only in indoor areas at a
maximum height above sea level of 2000 m.
Only service personnel authorized by Leica Microsystems CMS GmbH may assemble and
disassemble the device as well as carry out repairs and service work. Unauthorized alterations
to the device shall void all rights to any warranty claims. Leica Microsystems CMS GmbH shall
not be liable for any injury or property damage caused by untrained or unauthorized persons.
The owner/operator and users are fully liable for all consequences resulting from the use of the
device if it is opened, improperly serviced or repaired by persons other than authorized Leica
service representatives.

9 Warning Messages Corresponding to Specific Situations and Actions
8.4 If the Device Malfunctions
If the following cases of malfunction occur, find the causes and remedy them. Start by
immediately switching off the device if:
• Coolant leaks
• Cables are damaged
Each warning message contains a signal word which is used to classify the level of risk:
• WARNING: Hazard with moderate risk which could result in death or serious
physical injury if not avoided.
• CAUTION: Hazard with low risk which could result in minor or moderate physical
injury if not avoided.

10.1 ESD Protection
When using a wrist strap, make sure that the connections to the ground have
a series resistor of 1 MΩ.
• A wrist strap that is connected to the grounded antistatic mat on the worktable or
to grounding connections. Your wrist strap must be grounded and used at all
times when working on ESD sensitive components.
• ESD protected tools, e.g. screwdrivers, Allen keys, pliers, tweezers.
• ESD protection clothing (e.g. ESD jacket or ESD T-shirt) which creates a
wearable version of a Faraday cage. This shields ESD sensitive components
from static charges from your normal clothing. Additionally recommended are
either ESD shoes or a pair of heel grounders worn over your shoes to provide
consistent grounding while in motion. Neither of these can replace the wrist strap.

• Connect the crocodile clip of the ground cord from your antistatic mat to the
grounding screw of the device, for example the CTR box or the supply unit. These
devices of course must be grounded as well.
The grounding of the device is lost after disconnecting the power plug. In this
case, you have to ground the antistatic mat and the wrist strap with an earth
bonding point plug with a series resistor of 1 MΩ.

11 Electrical Acceptance Test
11.1 General
11.1 General
You have to carry out the electrical acceptance test after each service call on the device where
components in the primary circuit (power circuit) have been changed. This applies mainly to
power supplies that are implemented in the device. This does not apply to external power
supplies that are closed.
The mandatory electrical acceptance test that we prescribed is a minimum standard. If country-
specific regulations exist that go beyond the measurements that we prescribed, you also have
to comply with the country-specific requirements.
You must shut down the device if limits are exceeded when measuring. In this case, please
contact cms.support@leica-microsystems.com.
Fill in the attached inspection report during the measurement. You can also find it individually
in the documentation library. Sign it, save it (serial number_EIR) and send it. Please find a
detailed description below.
If you are a user of "Mobile Service", select the "CMS5 TSB 2016/08 Work on mains circuit"
activity code in your "Mobile Service Technical Report".

For measuring the protective conductor current, you need the following adapter cable. You can

locally. We recommend the following measuring equipment:

Here, you have to visually inspect the power supply cord for damage. If you opened the
housing, you also have to visually inspect the protective ground cable (PE). Proceed as
described in the inspection report.
The ground wire resistance is the resistance between the protective conductor terminal (PE)
and the metal housing. It must be less than 0.3 .
The measuring points are the protective conductor terminal (PE) and electrically conductive
points on the metal housing (also refer to figure 5)

If the metal housing is coated completely and you cannot find any regular or
alternative measuring point on the device (see figure 5), you have to pierce
the varnish with the gauge tip.
Below, we will describe how to measure the ground wire resistance for the Fluke 1507
insulation tester:
1. On the insulation tester, connect one measurement sensor to the "COM" connection
and the other measurement sensor to the "" connection.
2. Join both measurement sensors together to measure the line resistance of the
measuring line.
3. Hold the blue button for a few seconds until the insulation tester display shows a
series of dashes. The line resistance is now set.

4. Connect one of the measurement sensors to the protective conductor terminal (PE)
of the power plug. (The power plug must be from the original power cable!)
5. Connect the other measurement sensor to a measuring point on the housing.
6. Hold the "Test" button for a few seconds until a measurement result is displayed.
7. Repeat the measurement at another measuring point on the housing.
8. Enter the measured value into the inspection report.
You measure the insulation resistance as part of the insulation measurement. That is the
resistance between the phase (L)/neutral conductor (N) and the ground protection conductor
(PE).
It must be more than 2 M. Measuring points are the protective conductor terminal (PE), the
phase (L) and the neutral conductor (N) of the power plug. The measurement occurs with a test
voltage of 1000 V.
Below, we will describe how to measure for the Fluke 1507 insulation tester:

Even though the limit value for the insulation resistance is 2 M, the
insulation resistance is typically in the G range. If the value you measured
is not in the G range and you cannot find a reason for this behavior, please
contact cms.support@leica-microsystems.com.
The protective conductor current (or leakage current) is the current through the ground
protection conductor (PE). It must be less than 3.5 mA. Below, we will describe the procedure
with an adapter cable and the Amprobe AC50A leakage current clamp multimeter.

If you are a user of Mobile Service, select the "CMS5 TSB 2016/08 Work on mains circuit"
activity code in your "Mobile Service Technical Report".
Fill in the electrical inspection report during the measurement. You can also find it individually
in the documentation library. Use your own laptop, not the customer system. After having filled
in the electrical inspection report:
1. Sign it: Use the digital signature field, or use the field right down at the bottom with
your signature pad (see also tooltip, mouseover).
2. Click "save". Save it as "XXX_EIR" (XXX= serial number).
3. Click "send". The form is delivered to cms.support@leica-microsystems.com.

Lubricants are only allowed to come into contact with designated places,
since they can dissolve or damage plastics.

You can also make your own polishing sticks. Chamois leather becomes hard after being used
multiple times for cleaning and has to be replaced regularly. The chamois leather surface can
be periodically re-roughened with the cutting edge of a razor blade. Clean the brush and
polishing stick by repeatedly dipping them in alcohol and then blowing them dry. Cotton swabs,
optical cloths and lens cleaning paper are consumables. Dispose of them after using them
once.
Immediately remove the immersion medium from front lenses after use (e. g. type F immersion
liquid, in the past referred to as "immersion oil“):

repair and maintenance:

2 Repair and Service the Stand
2.1 Replace Backplane
The customer and factory settings are saved on the backplane. After exchanging the
backplane you have to program the settings on the new backplane. The following 2 options are
possible:

2.1 Replace Backplane
6. Remove the screw (figure 1, position 1) of the lamp housing flange, unscrew 4
screws (position 2) and pull off the casing (position 3).
7. Loosen the 4 screws (figure 1, position 4) of the back panel and tilt the back panel
(position 5) away.
8. Disconnect all plug connections to the "SYS Basic Module DM6000" board and
remove the back panel.
9. Replace the backplane and assemble the back panel in reverse order as described in
the working instructions 6 to 8.
10. Switch on the microscope.
11. Use the type plate program to program the correct type plate information to the new
backplane as shown in the service training.
12. Start LAS/Leica Hardware Configurator and load the latest firmware to the
backplane.
13. After finishing the firmware update, close the Hardware Configurator.
14. Restore customer and factory settings using the Leica test software.
Start the upload by using the option File Save & Restore Settings in the Leica test
software main window.
15. Confirm the successful upload by "OK" and close the Leica test software.

2.1 Replace Backplane
2. Loosen the 4 screws (figure 2, position 4) of the back panel and tilt the back panel
(position 5) away.
3. Disconnect all plug connections to the "SYS Basic Module DM6000" board and
remove the back panel.
4. Replace the backplane and assemble the back panel in reverse order as described in
the working instructions 1 to 3.

The reason is that the cable lugs connecting to the TL/IL lamp sockets may show a transition
resistance high enough to cause a raise of temperature of the cable lug, which in turn increases
the temperature of the PCB (up to 80° Celsius) and its components. This may cause damage
of some electronic components of the PCB.
In order to avoid such transition resistance, it is mandatory also to replace the cable lugs of the
TL/IL lamp connection field and cables connected to the PCB after exchanging one of the
above mentioned PCBs.

Newly shipped instruments containing this modification can be identified by the release version

Ensure that the polarity of the lamp outlet of the CTR box is correct.

2. Use a multimeter (DC voltage in the range of 2.5 - 12 V) to measure the polarity (see
figure 3).
3. If the polarity of the lamp outlet of the CTR5000/6000 is reversed, disconnect mains

When an objective with a magnification factor of less than 10x is turned into the light path, the
condenser top automatically moves out of the light path. That way the aperture diaphragm gets
the function of a field diaphragm.
After exchanging the TL diaphragm module, the aperture diaphragm (integrated in the TL
diaphragm module) has to be centered in the microscope in order to get a well centered image.
1. Screw the adjustment objective (figure 44) into an empty position of the nosepiece.

and reach the adjustment screws (figure 45, position 3) of the TL diaphragm module
at the same time.
7. Close the aperture diaphragm (field diaphragm function) with the keys (see figure 47)
until it becomes visible in the eyepiece. If the image of the diaphragm is not sharply
focused, adjust the movable lens (see figure 43).
8. Loosen the fixing screw (figure 45, position 2).
9. Using the 2 adjustment screws (figure 45, position 3), center the diaphragm to the
scale of the adjustment objective (figure 46).

5. Enter the command "84025" into the hyperterminal program to store the shutter offset
value.
This command stores the offset value (-1 to max -5) between the initialization
position and the "shutter closed" position. After this, the shutter completely
releases the diaphragm opening again ("shutter open" position) and the
diaphragm is opened to the stop that was stored with the particular objective.
6. After executing the command the stored offset value (e.g. 84025 -2) is returned by
the hyperterminal program.
7. The current AP diaphragm opening can be read out with the command "84023"
(GET_ POS_APBL_TL).
You will receive the command with a return value for the current position:
• -1 to -5: offset values for the "shutter closed" position
• 1 - 38: current position of the aperture diaphragm (1 = minimum opening)

The same diaphragm value is also shown on the display of the DM4000 B/M
or the touchscreen of the DM5000 B or DM6000 B/M.
When an objective with a magnification factor lower than 10x is turned into the light path, the
aperture diaphragm assumes the function of the field diaphragm. To obtain a sharp image of
the aperture diaphragm, an achromat has to be moved in the TL diaphragm module. This
happens automatically when an appropriate objective is turned into the light path.
If the distance between the aperture diaphragm and the lens has been altered mechanically or
the module has been removed, the “Field diaphragm position“ of the lens has to be readjusted
and stored by the "AdjustAdaptingLens.exe" software.
The same applies after exchanging the "MAN1 Basic Module" board (DM 4000) or "DM 5000
Basic Module" (DM5000) or "SYS Basic Module" (DM6000 B/M). The stored values are then
lost.
1. Install the "Leica Application Suite" (LAS) software package, version 2.0 R1 or higher
on the PC.
2. Check if the microscope works correctly.
3. Place a specimen on the stage.
4. Adjust the Koehler illumination.
5. Start the "AdjustAdaptingLens.exe".
6. Click "Connect" (figure 49, position 1) to get a connection to the microscope.

9.1 Update Firmware via Hardware Configurator

You can do the firmware update for all modules via the Hardware Configurator. The instructions
in chapter 9.1 are an example for a firmware update. In chapter 9.2 the exception of the touch
display is explained.
The touch panel of DiscoveryII microscopes are delivered with the current Leica STP firmware
(current at the time of delivery). If you have to update the firmware, proceed as follows:
1. Start the Hardware Configurator.
2. Click "Firmware Update". The module overview window opens.
3. Choose "Touch Panel Intern" and open the menu via right-click.
4. Click "Start Update".
5. After finishing the update, close the Hardware Configurator.
6. Restart the microscope.
If you have to replace the display board, the new board comes without the Leica firmware. The
firmware update can not be done via the Hardware Configurator. Download the firmware from
the Leica FTP server:
1. Download the firmware file from the FTP server to a USB stick.
The latest build of the firmware is available in the following folder:

Make sure to touch the calibration crosses very precisely. The routine will not
come to an end until all calibration crosses are touched correctly.
