Lab Report
Describe all the steps you performed during the experiment, when pipetted a specific liquid. List
the order in which the component’s reaction are added into a tube.
1:On the first day of lab our professor presented us a power point about pipeting. On those
slide we learned basic information about pipeting.
2:Firstly,in the lab we need it to put on safety equipment and cleaned our work space and pipets
with 70% ethanol.
3: After I took the micropipete,I adjusted it based on the tip I was going to use.
4:I took the device in my hand, I opened the box of the tips and put the micropipette above one
of them and pressed into it until the tip was firmly attached.
5:I put the plunger into its first stop and dipped inside the test tube which contains liquid.After
that,I released the plunger calmly with the tip still in liquid.
6: Then I inserted the tip into the second test tube, which is empty.i close the test tube without
touching the interior.
7: Lastly, I cleaned the micropipette with 70% ethanol and put the pipette back to its place
What did you learn during this session? What are some basic rules during pipetting in
laboratory, which you would advise others to consider?
In this laboratory lesson I managed to learn about different types of pipetts. I had the
opportunity to learn how to properly connect the tip the pipette.
One of the most important rule about the laboratory is that we should have a clean and non
contaminated environment.
Another rule is to keep the pipette as vertical as possible.
Most of the time it can happen because of some issues during the lab experiment like:
Forcing the solution out of the pipet causes too much to be delivered.
1. Using a dirty pipet causes too little or contaminated solution to be delivered. Leaving little
droplets behind on the walls (except for the small amount in the tip) causes too little
solution to be delivered.
2. Forcing the pipet into the bulb usually causes liquid to be sucked into the bulb. This
contaminates the bulb which must then be thoroughly rinsed and dried.
3. Allowing the tip of the pipet to rise above the liquid in the container usually causes the
liquid to be sucked into the pipet bulb.
4. A broken or chipped pipet can reduce the amount of liquid held after transfer. This
causes too much liquid to be delivered.
List the main parts of a micropipette and write the respective function.
Plunger: The topmost part of the micropipette. Pushing down the plunger helps in the aspiration
and dispensation of the desired amount of liquids. It consists of two stops, the first stop is for
aspiration and the second stop is for dispensation.
Tip ejector button: A button beside the plunger helps in the removal of pipette tips.
Volume adjustment knob: It is a knob attached to the plunger. Turning it helps in adjusting the
desired volume in the adjustable volume micropipette
Digital volume display window: It displays the volume that the micropipette can withdraw.
Plastic shaft: A cylinder structure filled with air in air micropipette.Once you press the piston
,some amount of air from the shaft releases helping in the aspiration of the liquid. In contrast
releasing the piston helps dispence the liquid and the air again fills up the shaft.
Ejector arm: It is an extension of the tip ejector button. It puts pressure on the pipette cone once
the ejector button is pressed.
