Determination of the
diameter of field of view
(dFOV)

Objectives

• By the end of this Experiment, the student should be able


to:
• Determine the diameter of field of view by using ruler under 4x
& 10x objective lenses .
• Determine the diameter of FOV for medium & high-power lens
(using equation)..
• Describe the conversion of scales from cm to mm and µm.
• Calculate the size of an object ( Cell etc.)

Theory and Principle
 Field of view is the diameter of the circle of light that you see when
you look through the eye pieces

(If you know how wide your field of view is, you can estimate the size
of things you see in the field of view)

As magnification increase the FOV get smaller (decrease )

For microscopic measurement of the size and diameter of
an object, we usually use metric system Which is based on
the ‘meter

But when we use a MICROscope we tend to use


MICROmeters.

To convert from millimeters to micrometers →

Units

➢ Visual :
• 1 meter (m) =100 centimeter (cm)
or 1000 millimeter (mm)

• 1 cm =10 mm
• 1 mm =1000 micrometer (µm)
➢ Microscopic
• 1 µm =1000 nanometer (nm)
• 1 nm =10 angstrom (A0)
Most cells are between 1 and 100 µm in diameter and
therefore are visible only under the microscope.

Measure diameter of field of view (dfov)
dFOV (mp) = diameter of field of view of medium power lens (..mm)

dFOV (hp) = diameter of field of view of high-power lens (…mm)

dFOV (lp) = diameter of field of view of low power lens (…mm)

Mag. lp = magnification power of low power lens (10x)

Mag.mp = magnification power of medium power lens (40x)

Mag.hp = magnification power of high-power lens (100x)

 Two steps:
➢ Using ruler under 4x and 10x only

➢ Using equation (formula) for 40x and 100x

Finding Field of View (F.O.V)

Under Low Power(10X): Use millimeter ruler

• Diameter of field is 1.5 mm →

• 1000 × 1.5 mm = 1500 micrometers

❖Under Medium or High Power: provide


a field of view that is less than 1 mm in
diameter, so it cannot be measured with a
ruler
