DNA BARCODING
 It is a technique in which species
identification is performed by using DNA
sequences from a small fragment of the
genome.

 DNA barcoding first came to the attention of


the scientific community in 2003 when Paul
Hebert’s research group at the university of
Guelph published a paper titled ‘Biological
identification through DNA barcodes’.
 Dr. Paul D.N. Hebert is the father of DNA
barcoding.

 DNA barcode is the widely used computational &


molecular based identification system.
WHICH GENE FRAGMENTS

 Prokaryotes : rRNA genes are used for


species identification.

 Animals,Birds& Fishes: Small regions of the


mt COI gene.

Plants : rbcL gene sequence & mat K gene


sequence of chloroplast.
 Two international initiatives working towards
the development of DNA barcoding are:

1.CBOL
 Consortium for the Barcoding Of Life.
 Established in 2004.

2.iBOL
 International Barcoding Of Life.
Largest biodiversity genomes initiative.
STEPS
i. Collect plants, animals or products in the local
environment or neighborhood.

ii. Extract & purify DNA from tissue or processed


material.

iii. Amplify a specific region of the chloroplast ,


mitochondrial or nuclear genome by PCR &
analyze PCR products by gel electrophoresis.
iv. Use the Basic Local Alignment Search Tool
(BLAST) to identify sequences in databases.

v. Use multiple sequence alignment to analyze


phylogenetic relationships.

Once the barcode sequence has been


obtained, it is placed in the Barcode Of Life
Data (BOLD) systems database .
BOLD –A reference library of DNA barcodes
that can be used to assign the identities to
unknown specimens.
 DNA barcode sequence are very short (<1000
bp) relative to the entire genome & they can be
obtained reasonably quickly & cheaply.

 All the products of one type on a super market


shelf (like a 2 l bottle Coca Cola) share exactly
the same 11 digit barcode , which is distinct
from all other barcodes.

 DNA barcodes vary among individuals of the


same species , but only to a very minor degree.
 The importance of plant DNA barcode is the
accurate species identification
