LABORATORY EXPERIMENT NO.

COMPOUND MICROSCOPE

Introduction

The microscope is an important laboratory equipment in the study of living things. Much of the
present knowledge of cellular organization has been made possible by the development of better and more
powerful microscopes.

With such importance, this activity highlights the value of knowing the principle of light and optics
in microscopy. It further emphasizes the four important attributes of the microscope: magnification,
resolution, contrasts and microscopic measurement. It is in these attributes that the effective use of the
microscope lies.

Objectives

At the end of this activity, the students are expected to:

1.Label the parts of the compound microscope and their functions


2.Examine the prepared slides of paramecia, amoeba and other forms of protozoans under LPO and
HPO
3.Prepare a wet mount slide
4.Manipulate the microscope correctly
5.Determine the magnification of the specimens

Materials

1 Compound microscope Prepared slides:paramecia, amoeba and euglena


2 Glass slide and coverslip
Dropper
Specimen (old newspaper)

Procedure

A. The Compound Microscope


1.Label the numbered parts of the compound microscope in Figure 1.1.
2.Classify each parts of the microscope as to illuminating, magnifying and supporting. Give the functions
of each numbered parts. Record your observation in Table 1.1.
3.List down the techniques in handling the microscope.

B.Microscope Manipulation

1.Using letter “e” as the specimen

a.Cut a smallest “e” from an old newspaper.


b.Place the “e” on the glass slide in an upright position. Cover with a slip.
Drop a small amounts of water at the side of the slip.
c.Examine the specimen under LPO. Draw the image as it appears under the
microscope.
d.Move the specimen to your right and then to your left. Notice the direction
of the movement.
e.After LPO, shift your focus to the HPO. Draw the image. Observe the
differences.

2.Using hair strand as the specimen

a.Make a wet mount of two crossed hair strands and focus under the LPO,
then HPO.
b.Draw the image.

3.Examine the provided prepared slides

a.Secure the prepared slides ( paramecia, amoeba and euglena) .


b.Focus the slide under the LPO and HPO. Draw the image.
c.Determine the magnification of your drawing.
