Coli surface antigen 6 (CS6) is one of the most prevalent colonization factors among enterotoxigenic Escherichia coli (ETEC) isolated in developing countries. Although it is known that CS6 is encoded by a plasmid, there are no reports on the sequence analysis of the CS6-encoding plasmid or genes exhibiting similar behavior to CS6. Here, we report the isolation of the CS6-encoding plasmid, pCss165Kan, from 4266 DcssB::kanamycin (Km) and its complete nucleotide sequence. This plasmid consisted of 165,311 bp and 222 predicted coding sequences. Remarkably, there were many insertion sequence (IS) elements, which comprised 24.4% of the entire sequence. Virulence-associated genes such as heat-stable enterotoxin, homologues of ATP-binding cassette transporter in enteroaggregative E. coli (EAEC), and ETEC autotransporter A were also present, although the ETEC autotransporter A gene was disrupted by the integration of IS629. We found that 2 transcriptional regulators belonging to the AraC family were not involved in CS6 expression. Interestingly, pCss165 had conjugative transfer genes, as well as 3 toxin-antitoxin systems that potentially exclude other plasmid-free host bacteria. These genes might be involved in the prevalence of CS6 among ETEC isolates.

Methylglyoxal (MG), which forms MG-derived AGE, is elevated in diabetic subjects with vascular disease. Detoxification of MG occurs through the glyoxalase system incorporating glyoxalase-1 (GLO1) and glyoxalase-2. Perturbations of the glyoxalase-1 gene (GLO1) may result in vulnerability to vascular complications through alterations in AGE interactions.

It is desirable to verify the structural performance based on a multi-hazard approach, taking into account the critical actions the structure in question could be subjected to during its lifetime. This study presents a proposal for a probabilistic model for multi-hazard risk associated with the limit state of collapse for a reinforced concrete (RC) structure subjected to blast threats in the presence of seismic risk. The annual risk of structural collapse is calculated taking into account both the collapse caused by an earthquake event and the blast-induced progressive collapse. The blast fragility is calculated using a simulation procedure for generating possible blast configurations, and verifying the structural stability under gravity loading of the damaged structure, using a kinematic plastic limit analysis. As a case study, the blast and seismic fragilities of a generic four-storey RC building located in seismic zone are calculated and implemented in the framework of a multi-hazard procedure, leading to the evaluation of the annual risk of collapse.

Apache Spark is an engine for large scale data processing, best described as a more flexible version of the venerable MapReduce framework with the capability to perform in-memory processing to speed up computation. BLAST (Basic Local Alignment Search Tool) is a tool used to determine the similarity of DNA and protein sequences that is used extensively in bioinformatics studies. To illustrate the use of Apache Spark in porting bioinformatics tools to the world of cluster computing, I demonstrate the creation of a distributed version of BLAST with minimal coding and simple cluster deployment. I measure performance, result accuracy, and cost efficiency to determine the viability of this approach to BLAST and other bioinformatics programs.

Rice (Oryza sativa) s one of the most important food crops of India in term of area, production and consumer preference. But production of Basmati rice is limited due to losses cause by disease at various stages from nursery to harvesting. Conducted a survey in Basmati rice growing areas in Western U.P in khrief 2013, observed major diseases which cause significance losses in Basmati. During the survey, Blast and Bakanae diseases were found at significant level. For their control farmers used application of pesticides which leaves residual effect on grain surface and it plays major role to reject a consignment in export.

After sexual reproduction in ciliated protozoa a new macronucleus differentiates from a micronuclear derivative. In the course of macronuclear development dramatic DNA-and chromatin reorganisation processes occur, which include splicing of DNA sequences such as IES (internal eliminated sequences) and transposon-like elements during formation of polytene chromosomes, degradation of the polytene chromosomes and specific elimination of micronuclear-specific DNA, de novo addition of telomeres and specific amplification of DNA sequences. In order to understand the molecular basis of this nuclear differentiation process, analysis of developmentally regulated genes seems to be a necessary prerequisite. We performed a microarray analysis to identify genes differentially expressed during macronuclear differentiation. 467 sequences from cDNA libraries were identified as possible candidates from which 384 sequences were further characterised by sequence analysis. These sequences were identified, if possible, by DNA and protein BLAST analysis. Expression of one of these sequences was silenced by RNAi and a preliminary functional analysis performed. Results presented in this study provide the basis for a functional characterisation of genes differentially expressed during this nuclear differentiation process.

AASHTO has specified probability-based design methodology and load factors for designing bridge piers against ship impact and vehicular collision. Currently, no specific AASHTO design guideline exists for bridges against blast loading. Structural engineering methods to protect infrastructure systems from terrorist attacks are required. This study investigated the most common types of concrete bridges on the interstate highways. A 2-span 2-lane bridge with Type III AASHTO girders was used for modeling. AASHTO Load and Resistance Factor Design methods were used for the model bridge design. The girders, pier caps and columns loading were analyzed for probable blast loading. The model bridge failed under the probable blast loads applied over and underneath the bridge. The research findings show that typical AASHTO girder bridges are unable to resist probable blast loads. Una catástrofe supone, además de la destrucción de bienes materiales, un daño sobre personas de tal magnitud que se puede comprometer la correcta asistencia sanitaria de los centros disponibles. En los últimos años hemos observado terribles atentados terroristas que cada vez nos afectan de una forma más cercana. A continuación se describe y analiza la actuación de un departamento de traumatología de un hospital universitario ante un ataque terrorista que ocasionó cerca de 2.000 heridos y 191 muertos. Las lesiones de las víctimas del atentado se caracterizaron, como es habitual, por la gravedad y extensión del compromiso tisular, e incluyeron heridas penetrantes, lesiones por estallido y quemaduras. El análisis crítico de catástrofes previas, descritas por colegas nuestros en la bibliografía, puede sernos útil para evitar errores futuros.

The behavior of reinforced concrete panels, or slabs, retrofitted with glass fiber reinforced polymer (GFRP) composite, and subjected to blast load is investigated. Eight 1000 · 1000 · 70 mm panels were made of 40 MPa concrete and reinforced with top and bottom steel meshes. Five of the panels were used as control while the remaining four were retrofitted with adhesively bonded 500 mm wide GFRP laminate strips on both faces, one in each direction parallel to the panel edges. The panels were subjected to blast loads generated by the detonation of either 22.4 kg or 33.4 kg ANFO explosive charge located at a 3-m standoff. Blast wave characteristics, including incident and reflected pressures and impulses, as well as panel central deflection and strain in steel and on concrete/FRP surfaces were measured. The post-blast damage and mode of failure of each panel was observed, and those panels that were not completely damaged by the blast were subsequently statically tested to find their residual strength. It was determined that overall the GFRP retrofitted panels performed better than the companion control panels while one retrofitted panel experienced severe damage and could not be tested statically after the blast. The latter finding is consistent with previous reports which have shown that at relatively close range the blast pressure due to nominally similar charges and standoff distance can vary significantly, thus producing different levels of damage.

Studies of traumatic brain injury from all causes have found evidence of chronic hypopituitarism, defined by deficient production of one or more pituitary hormones at least 1 year after injury, in 25-50% of cases. Most studies found the occurrence of posttraumatic hypopituitarism (PTHP) to be unrelated to injury severity. Growth hormone deficiency (GHD) and hypogonadism were reported most frequently. Hypopituitarism, and in particular adult GHD, is associated with symptoms that resemble those of PTSD, including fatigue, anxiety, depression, irritability, insomnia, sexual dysfunction, cognitive deficiencies, and decreased quality of life. However, the prevalence of PTHP after blast-related mild TBI (mTBI), an extremely common injury in modern military operations, has not been characterized. We measured concentrations of 12 pituitary and target-organ hormones in two groups of male US Veterans of combat in Iraq or Afghanistan. One group consisted of participants with blast-related mTBI whose last blast exposure was at least 1 year prior to the study. The other consisted of Veterans with similar military deployment histories but without blast exposure. Eleven of 26, or 42% of participants with blast concussions were found to have abnormal hormone levels in one or more pituitary axes, a prevalence similar to that found in other forms of TBI. Five members of the mTBI group were found with markedly low ageadjusted insulin-like growth factor-I (IGF-I) levels indicative of probable GHD, and three had testosterone and gonadotropin concentrations consistent with hypogonadism. If symptoms characteristic of both PTHP and PTSD can be linked to pituitary dysfunction, they may be amenable to treatment with hormone replacement. Routine screening for chronic hypopituitarism after blast concussion shows promise for appropriately directing diagnostic and therapeutic decisions that otherwise may remain unconsidered and for markedly facilitating recovery and rehabilitation.

The paper focuses on the numerical analysis of blast-induced stress wave propagation and related spalling damage in a rock plate or wall. Firstly, a large-scale contact explosion is simplified as a one-dimensional (1-D) strain problem, and a cross-format centered finitedifference scheme is presented. Secondly, the finite-difference code is used to explore the wave propagation in a rock plate, and compared with the commercial software LS-DYNA. Thirdly, a continuum damage constitutive model for rock is introduced and successfully incorporated into the LS-DYNA through its user-defined subroutine. Coupling with the erosion technique, the improved LS-DYNA is used to simulate the blast-induced spalling damage at the back-side of rock plate. Numerical results show that the 1-D finite difference code is convenient and accurate. Also, the user-defined subroutine can well describe the process of spalling damage. r

The incidence of traumatic brain injury (TBI) among military personnel is at its highest point in U.S. history. Experimental animal models of blast have provided a wealth of insight into blast injury. The mechanisms of neurotrauma caused by blast, however, are still under debate. Specifically, it is unclear whether the blast shockwave in the absence of head motion is sufficient to induce brain trauma. In this study, the consequences of blast injury were investigated in a rat model of primary blastTBI. Animals were exposed to blast shockwaves with peak reflected overpressures of either 100 or 450 kPa (39 and 110 kPa incident pressure, respectively) and subsequently underwent a battery of behavioral tests. Diffusion tensor imaging (DTI), a promising method to detect blast injury in humans, was performed on fixed brains to detect and visualize the spatial dependence of blast injury. Blast TBI caused significant deficits in memory function as evidenced by the Morris Water Maze, but limited emotional deficits as evidenced by the Open Field Test and Elevated Plus Maze. Fractional anisotropy, a metric derived from DTI, revealed significant brain abnormalities in blast-exposed animals. A significant relationship between memory deficits and brain microstructure was evident in the hippocampus, consistent with its role in memory function. The results provide fundamental insight into the neurological consequences of blast TBI, including the evolution of injury during the sub-acute phase and the spatially dependent pattern of injury. The relationship between memory dysfunction and microstructural brain abnormalities may provide insight into the persistent cognitive difficulties experienced by soldiers exposed to blast neurotrauma and may be important to guide therapeutic and rehabilitative efforts.

Time-dependent changes in blood-based protein biomarkers can help identify the -pathological processes in blast-induced traumatic brain injury (bTBI), assess injury severity, and monitor disease progression. We obtained blood from control and injured mice (exposed to a single, low-intensity blast) at 2-h, 1-day, 1-week, and 1-month post-injury. We then determined the serum levels of biomarkers related to metabolism (4-HNE, HIF-1α, ceruloplasmin), vascular function (AQP1, AQP4, VEGF, vWF, Flk-1), inflammation (OPN, CINC1, fibrinogen, MIP-1a, OX-44, p38, MMP-8, MCP-1 CCR5, CRP, galectin-1), cell adhesion and the extracellular matrix (integrin α6, TIMP1, TIMP4, Ncad, connexin-43), and axonal (NF-H, Tau), neuronal (NSE, CK-BB) and glial damage (GFAP, S100β, MBP) at various post-injury time points. Genome comparison has shed light on many fields of both basic and applied research, including the study of species phylogeny. Grass carp (Ctenopharyngodon idella) belongs to Cyprinidae, the largest freshwater fish family; but which subfamily it belongs to remains a controversial issue. In this study, the complete mitochondrial genome (mitogenome) sequence of grass carp was determined and phylogenetic analyses of all mitochondrial protein-coding genes and a nuclear gene (RAG 2) were conducted to explore the evolutionary relationship of grass carp with other cyprinid species. The mitogenome of grass carp is 16,609 bp in length. As with most other vertebrates, it contains the same gene order and an identical number of genes or regions, including 13 protein-coding genes, two rRNA genes, 22 tRNA genes and one putative control region. Phylogenetic analyses using two different datasets (mitochondrial and nuclear) and three different computational algorithms (Bayesian, MP and ML) all revealed two distinct groups with high statistical support, indicating that Cyprininae and Leuciscinae are two separate, valid subfamilies. Importantly, our phylogenetic result provides strong molecular evidence in support of the placement of Ctenopharyngodon in Leuciscinae rather than in Cyprininae.

Primary approach to prevent cervical cancer includes the development of human papillomavirus (HPV) vaccines. Currently available HPV vaccines (Gardasil and Cervarix) predominantly consider HPV16 and HPV18 strains. However, due to ignorance of the other high-risk strains aside from HPV16 and HPV18 during vaccine development, the critical need is to synthesize a vaccine with possible protection against all the high-risk HPV types. One feasible approach is to design a vaccine containing conserved immunogenic peptides that represent the genotypic diversity of all the current and future high-risk HPV types. While the epitopes derived from sequentially conserved regions may undergo mutations, it is worthwhile to explore the structurally conserved regions as a new dimension for epitope prediction. In the present study, 81 structurally conserved peptides were predicted to have immune relevance as T-cell epitopes of all the reported high-risk HPV proteins studied. A small dataset of three epitopes was also recognized as potential vaccine candidates generating both CD8+ and CD4+ responses.

The formation of the American Society for Surgery of the Hand was related to world conflicts and hostilities. Therefore, it is appropriate that upper-extremity surgeons understand injuries resulting from modern-day combat. Because of ongoing warfare, many countries have experienced a large increase in the number of wounded service members and civilians, particularly wounds of the extremities. As a result of increased rate of survival in battlefield trauma in part because of the use of modern body armor, there is increasing complexity of extremity injuries that require complex reconstructions. Decreased mortality and a consequent increase in the incidence of injured extremities underline the need for the development of new treatment options. Impact of blast shock waves (SW) with the body wall produces blast lung injuries characterized by bilateral traumatic hemorrhages. Such injuries often have no external signs, are difficult to diagnose, and therefore, are frequently underestimated. Predictive assessment of acute respiratory distress syndrome outcome in SWrelated accidents should be based on experimental data from appropriate animal models. Blood plasma transferrin is a major carrier of blood iron essential for proliferative "emergency" response of hematopoietic and immune systems as well as injured tissue in major trauma.

Wheat blast disease caused by Pyricularia grisea (telemorph Magnaporthe grisea) has become a serious restriction on increasing the area and production of the crop, especially in the tropical parts of the Southern Cone Region of South America. First identified in 1985 in the State of Paraná in Brazil, it has become an endemic disease in the low lying Santa Cruz region of Bolivia, south and south-eastern Paraguay, and central and southern Brazil in recent years. Severe infections have also been observed in the summer planted wheat crop in north-eastern Argentina. So far, only sporadic infections have been seen in Uruguay, especially during the wet and warm years. Spike infection (often confused with Fusarium head blight infection) is the most notable symptom of the disease and capable of causing over 40% production losses. However, under severe infection, the loss of production can be almost complete in susceptible varieties. Wheat blast is mainly a spike disease but can also produce lesions on all the above ground parts of the plant under certain conditions. Depending upon the point of the infection on the rachis, the disease can kill the spike partially or fully. The infected portion of the spike dries out without producing any grain which can be visibly distinguished from the healthy portion. While virulence diversity in the fungus has been reported in the literature and is under further exploration, genetic resistance in the host species has been more difficult to identify. Earlier, Brazilian cultivars such as BH 1146, CNT 8, several IAC and OCEPAR selections were credited as demonstrating different levels of field resistance, but this was not confirmed under artificial inoculation studies. However, other cultivars such as BR18, IPR 85, CD 113, have shown moderate levels of resistance over the years in many locations. Recently, several cultivars and advanced lines derived from the CIMMYT line, Milan, have been observed to carry a high level of resistance to blast disease throughout the endemic region. However, to date, the genetic basis of this resistance is not very clear due to extreme variation in the pathogen. Cultivars showing complete resistance against a few isolates under controlled conditions in the glasshouse, may or may not show field resistance in commercial cultivation. Due to an increase of the area under Milan based resistant wheat cultivars in Bolivia, Brazil and Paraguay, it needs to be combined with other sources of resistance urgently to prevent the selection of a virulent pathotype in the fungus. Besides genetic resistance, avoidance of early dates of seeding and chemical control can reduce the disease severity. Fungicides combining triazols with strobilurins can, under some situations, be effective in disease control at the heading stage. Even when all components of integrated disease management of wheat blast are not in place yet, it is seen as an essential strategy to reduce production losses in this region. Given the threat that the blast disease may pose to world wheat growing areas in the future, more research efforts are deemed urgent and necessary.

2011 3 SYNOPSIS The report presents a constitutive model for simulating the high strain-rate behavior of sands. Based on the concepts of critical-state soil mechanics, the bounding surface plasticity theory and the overstress theory of viscoplasticity, the constitutive model simulates the high strain-rate behavior of sands under uniaxial, triaxial and multiaxial loading conditions. The model parameters are determined for Ottawa and Fontainebleau sands, and the performance of the model under extreme transient loading conditions is demonstrated through simulations of split Hopkinson pressure bar tests up to a strain rate of 2000/sec. The constitutive model is implemented in a finite element analysis software to analyze underground tunnels in sand subjected to internal blast loads.

Over the past decade, much research has focused on the behaviour of structures following the failure of a key structural component. Particular attention has been given to sudden column loss, though questions remain as to whether this event-independent scenario is relevant to actual extreme events such as explosion. Few studies have been conducted to assess the performance of floor slabs above a failed column, and the computational tools used have not been validated against experimental results. The research program presented in this paper investigates the adequacy of sudden column loss as an idealisation of local damage caused by realistic explosion events, and extends prior work by combining the development of accurate computational models with large-scale testing of a typical floor system in a prototypical steel-framed structure. The floor system consists of corrugated decking topped by a lightly reinforced concrete slab that is connected to the floor beams through shear studs. Fruit aroma is a complex trait, particularly in terms of the number of different biosynthetic pathways involved, the complexity of the final metabolites, and their regulation. In order to understand the underlying biochemical processes involved in apricot aroma, four cDNAs (Pa-aat, EU784138; Pa-adh EU395433; Pa-pdc EU395434; and Pa-lox EU439430) encoding an alcohol acyl transferase (AAT), alcohol dehydrogenase (ADH), pyruvate decarboxylase (PDC), and lipoxygenase (LOX), respectively, were isolated and characterized at four stages of maturity in Prunus armeniaca L. cv. Modesto. We observed a reduction in aldehyde and alcohol production between early-harvested fruit and late-harvest fruit, concomitant with an increase in ester production. qPCR analyses showed that the expression levels of the adh gene and the lox gene stayed constant at all stages. Interestingly, aat levels showed a sharp increase in the lateharvest stages concurrent with the changes observed in ester levels. The significance of these changes in relation to aroma production in apricot is discussed.

HPLC analyses of Xanthoria elegans cultivated on different media and either aposymbiontically or with its photobiont revealed that the carbon source and the presence of the algal partner have an impact on the secondary metabolism of the mycobiont. The aposymbiotically (without photobiont) grown mycobiont contained up to 70% more of the main compounds in its thallus than in resynthesis stage. Although this is speculative, the induction of the polyketide pathway may be a feedback mechanism to the absence of the photobiont. All cultures produce a variety of substances which were not detectable in the voucher specimen. Besides physcion (the major substance), we were able to identify emodin as well as physcion-bisanthrone, teloschistin monoacetate and derivatives. A strong inducible effect on the production of physcion, physcion-bisanthrone and on their precursors and derivatives was found for mannitol. By contrast, supplementation of ribitol had negligible effects, if any, on polyketide quantities although it is the main carbon source for the mycobiont in free-living lichens with Trebouxia photobiont.

Probability analysis is commonly used to estimate the structural damage subjected to the static loads as well as dynamic loads such as earthquakes, wind and blast loads. Blast loads is difficult to predict accurately due to the parameters that influence the uncertainty in the blast shock wave propagation and shock wave-structures interaction. However, probability analysis of the structural damage can be carried out by considering all the blast load parameters and the structural properties. Instead, scale distance factors (producing various pressures and impulses) also affect the uncertainty of variations in structure damage to blast load and analysis of blast-resistant design. This study presents a reliability analysis of unstrengthened and FRP strengthened RC columns to blast loads. Three different parameter examples of unstrengthened reinforced concrete (RC) columns and Fibre Reinforced Polymer (FRP) strengthened RC column are used. The failure probabilities of RC columns under different level blast load corresponding to different range of scaled distances are estimated and presented. The results indicate reliability analysis gives range of scaled distances with different probabilities of column collapse.

This report describes an implementation of a Godunov-type solver for gas dynamics equations in MATLAB R. The main attention is paid to providing a generic code that can be easily adapted to particular problems in one, two or three dimensions. This is achieved by employing a cell connectivity matrix thus allowing one to use various structured and unstructured meshes without modification of the core solver. The code has been thoroughly tested for MATLAB Version 7.6 (Release 2008a).

Research conducted by Department of Defense laboratories and facilities, collaborating with leading academic institutions, has demonstrated that currently available landmine protective footwear does not prevent severe injury. This footwear potentially reduces injury severity against some antipersonnel mines. Volume 2 of the Lower Extremity Assessment Program (LEAP 99-2) discusses the change of injury pattern in a cadaver model wearing mine protective footwear during an antipersonnel blast mine detonation. Our analytical methodology developed to assess mine-protective footwear and injury severity associated with blast mines across the spectrum of threat is presented.

In a multi-institutional collaborative project, 1473 patients with myeloproliferative neoplasms (MPN) were screened for isocitrate dehydrogenase 1 (IDH1)/IDH2 mutations: 594 essential thrombocythemia (ET), 421 polycythemia vera (PV), 312 primary myelofibrosis (PMF), 95 post-PV/ET MF and 51 blast-phase MPN. A total of 38 IDH mutations (18 IDH1-R132, 19 IDH2-R140 and 1 IDH2-R172) were detected: 5 (0.8%) ET, 8 (1.9%) PV, 13 (4.2%) PMF, 1 (1%) post-PV/ET MF and 11 (21.6%) blast-phase MPN (P<0.01). Mutant IDH was documented in the presence or absence of JAK2, MPL and TET2 mutations, with similar mutational frequencies. However, IDH-mutated patients were more likely to be nullizygous for JAK2 46/1 haplotype, especially in PMF (P=0.04), and less likely to display complex karyotype, in blast-phase disease (P<0.01). In chronic-phase PMF, JAK2 46/1 haplotype nullizygosity (P<0.01; hazard ratio (HR) 2.9, 95% confidence interval (CI) 1.7–5.2), but not IDH mutational status (P=0.55; HR 1.3, 95% CI 0.5–3.4), had an adverse effect on survival. This was confirmed by multivariable analysis. In contrast, in both blast-phase PMF (P=0.04) and blast-phase MPN (P=0.01), the presence of an IDH mutation predicted worse survival. The current study clarifies disease- and stage-specific IDH mutation incidence and prognostic relevance in MPN and provides additional evidence for the biological effect of distinct JAK2 haplotypes.

Genes in an organism's DNA (genome) have embedded in them information about proteins, which are the molecules that do most of a cell's work. A typical bacterial genome contains on the order of 5,000 genes. Mammalian genomes can contain tens of thousands of genes. For each genome sequenced, the challenge is to identify protein components (proteome) being actively used for a given set of conditions. Fundamentally, sequence alignment is a sequence matching problem focused on unlocking protein information embedded in the genetic code, making it possible to assemble a "tree of life" by comparing new sequences against all sequences from known organisms. But, the memory footprint of sequence data is growing more rapidly than per-node core memory. Despite years of research and development, high-performance sequence alignment applications either do not scale well, cannot accommodate very large databases in core, or require special hardware. We have developed a high-performance sequence alignment application, ScalaBLAST, which accommodates very large databases and which scales linearly to as many as thousands of processors on both distributed memory and shared memory architectures, representing a substantial improvement over the current state-of-the-art in highperformance sequence alignment with scaling and portability. ScalaBLAST relies on a collection of techniques-distributing the target database over available memory, multilevel parallelism to exploit concurrency, parallel I/O, and latency hiding through data prefetching-to achieve high-performance and scalability. This demonstrated approach of database sharing combined with effective task scheduling should have broad ranging applications to other informatics-driven sciences.

IgM detection is considered as the gold standard for mumps diagnosis. Currently, most cases in developed countries occur in highly vaccinated populations due to secondary vaccine failure. In these patients, pre-existing vaccine-induced antibodies are not able to neutralise the virus, but prevent the typical primary response, so that specific IgM is not always elicited. Consequently, acute infection has to be demonstrated by direct detection of the virus by viral isolation or genomic amplification. RT-PCR allows a diagnosis with the maximum sensitivity to be made and also forms the basis for genotype characterisation by sequencing the SH gene, according to WHO recommendations. However, none of the RT-PCR techniques properly evaluated for the diagnosis of acute mumps infection yields an amplification fragment useful for genotyping, and none of the amplification techniques described for genotyping has proved to be sensitive enough for diagnosis.Development of a RT-PCR for the mumps virus diagnosis and genotyping, properly evaluated in comparison with serological gold-standard technique.195 suspected mumps cases and six wild type MuV genotypes were studied.Our method was able to detect 0.001 TCID50 of mumps virus. Fifty-eight of these showed positive results, of which 54 (93.3%) showed mumps RNA in saliva, while only 20 (34.5%) had mumps IgM in serum. Genotypes G1, G2, H1, H2, D1 and C were identified in positive samples.The technique described could be a very useful tool for mumps surveillance, management and control.

Invertebrate metallothioneins (MT) have mainly been reported in digestive tissues, but data about the existence of a ubiquitous isoform expressed also in nervous tissue, are not available. Here we report the identification of a new metallothionein gene (MTPA) from the lobster Panulirus argus, putatively encoding a 59 residue polypeptide including 19 Cys. Tissue specific analysis indicated that MTPA is ubiquitously expressed in the hepatopancreas, intestine, nervous tissue and muscle, with the highest levels in the hepatopancreas and the lowest in muscle and nervous tissue. In addition, our data showed that MTPA is differentially regulated by metals: tissue explants exposed to Cd exhibited increased MTPA mRNA levels in all cases, except in muscle, with the highest effects in the nervous tissue, while Zn was effective only in the hepatopancreas. Interestingly, Cu showed no effects in any of the analyzed tissues. Taken together, these results suggest that MTPA in the hepatopancreas likely plays an important role in Cd detoxification and Zn homeostasis. The potent Cd-inducibility of MTPA in the nervous tissue might suggest a key function of this protein in protecting this highly sensitive tissue from cadmium-induced neurotoxicity.

The work presented in this paper concerns a project on the optimization of protections subjected to explosions (IED's threat). Explosions generate two types of threats for a protective structure: blast and impact of fragments. Perforating and non-perforating impact tests were performed in our laboratory with steel spherical projectiles impacting a target based on Kevlar ® textile layers and a crushable material, Crushmat ® . These tests required to develop a specific experimental test setup in order to contain the composite protective structure and to be able to measure the relevant parameters. The experiments allow us to determine the ballistic performance and basic parameters of the protection, and to validate finite element numerical models (LS-DYNA) resulting in a performant prediction tool. The approach used for the simulation consists in the representation of the full textile architecture with solid elements. Therefore, the textile material is explicitly represented in the model in order to have a good representation of the physical phenomena occurring during impact. For the crushable material, a representation using SPH was chosen in order to take the granular behaviour of this material into account. Good results are obtained with such models. However, these models are very complicated and computing time consuming. The geometry has to be well adapted and symmetry has to be exploited. On the other hand, representation of a granular material with SPH does not take into account some characteristics of this material during impact, such as the pulverisation process of the granular material. Solutions to take these phenomena into account in the model are proposed.
