Serum lacks clotting factors while plasma contains them. Serum volume is less than plasma. Serum is isolated after clotting while plasma is isolated before clotting. Serum separation does not require anticoagulants but plasma separation does.

Serum is isolated by allowing blood to clot and then centrifuging to separate the clot from the liquid portion. Plasma is isolated by collecting blood into a tube containing anticoagulant and then centrifuging.
